Neuroglobin (Ngb) is a recently discovered vertebrate globin expressed primarily in neurons. Ngb expression is induced by hypoxia and ischemia, and Ngb protects neurons from these insults. However, its normal physiological role and the mechanism underlying its neuroprotective action are uncertain. We report production of a transgenic mouse in which Ngb is overexpressed under the control of the chicken beta-actin promoter. This mouse should prove helpful for studying Ngb-mediated effects in vitro and in vivo.  相似文献

Small model peptides containing N-terminal methionine are reported to form sulfur-centered-free radicals that are stabilized by the terminal N atom. To test whether a similar chemistry would apply to a disease-relevant longer peptide, Alzheimer's disease (AD)-associated amyloid beta-peptide 1-42 was employed. Methionine at residue 35 of this 42-mer has been shown to be a key amino acid residue involved in amyloid beta-peptide 1-42 [A beta1-42]-mediated toxicity and therefore, the pathogenesis of AD. Previous studies have shown that mutation of the methionine residue to norleucine abrogates the oxidative stress and neurotoxic properties of A beta(1-42). In the current study, we examined if the position of methionine at residue 35 is a criterion for toxicity. In doing so, we tested the effects of moving methionine to the N-terminus of the peptide in a synthetic peptide, A beta(1-42)D1M, in which methionine was substituted for aspartic acid at the N-terminus of the peptide and all subsequent residues from D1 to L34 were shifted one position towards the carboxy-terminus. A beta(1-42)D1M exhibited oxidative stress and neurotoxicity properties similar to those of the native peptide, A beta(1-42), all of which are inhibited by the free radical scavenger Vitamin E, suggesting that reactive oxygen species may play a role in the A beta-mediated toxicity. Additionally, substitution of methionine at the N-terminus by norleucine, A beta(1-42)D1Nle, completely abrogated the oxidative stress and neurotoxicity associated with the A beta(1-42)D1M peptide. The results of this study validate the chemistry reported for short peptides with N-terminal methionines in a disease-relevant peptide.  相似文献

Background

It is well established that low levels of 25(OH) Vitamin D (<30 ng/dL) are a common finding world over, affecting over a billion of the global population. Our primary objective was to determine the prevalence of vitamin D deficiency and insufficiency in the asymptomatic adult population of Karachi, Pakistan and the demographic, nutritional and co-morbidity characteristics associated with serum vitamin D levels.

Methods

A cross-sectional population survey was conducted at two spaced out densely populated areas of the city. Serum levels of 25OHVitamin D were measured and GFR as renal function was assessed by using 4 variable MDRD formula.

Results

Our sample of 300 had a median age of 48(interquartile range 38–55) years. The median level of serum vitamin D was 18.8 (IQ range 12.65–24.62) ng/dL. A total of 253 (84.3%) respondents had low levels (<30 ng/dL) of 25OH vitamin D. Serum PTH and vitamin D were negatively correlated (r = −0.176, p = 0.001). The median PTH in the vitamin D sufficiency group was 38.4(IQ range28.0–48.8)pg/mL compared with 44.4(IQ range34.3–56.8) pg/mL in the deficiency group (p = 0.011).The median serum calcium level in the sample was 9.46(IQ range 9.18–9.68) ng/dL. Low serum levels of vitamin D were not associated with hypertension (p = 0.771) or with an elevated spot blood pressure (p = 0.164).In our sample 75(26%) respondents had an eGFR corresponding to stage 2 and stage 3 CKD. There was no significant correlation between levels of vitamin D and eGFR (r = −0.127, p-value = 0.277).Respondents using daily vitamin D supplements had higher 25 OH vitamin D levels (p-value = 0.021).

Conclusion

We observed a high proportion of the asymptomatic adult population having low levels of vitamin D and subclinical deterioration of eGFR. The specific cause(s) for this observed high prevalence of low 25OH vitamin D levels are not clear and need to be investigated further upon.  相似文献

Comparative studies suggest that a positive correlation between xylem water transport and photosynthesis is adaptive. A requirement for the adaptive evolution of coordination between xylem and photosynthetic functions is the presence of genetic variation and covariation for these traits within populations. Here it was determined whether there was genetic variation and covariation for leaf blade hydraulic conductivity (K(W)), photosynthetic rate (A), stomatal conductance (g(s)), and time to flowering in a population of recombinant inbred lines of Avena barbata, a Mediterranean annual grass. Significant (P < 0.05) broad-sense heritabilities (H(2)) were detected for K(W) (H(2) = 0.33), A (H(2) = 0.23) and flowering time (H(2) = 0.62), but not for g(s). Significant positive genetic covariation between A and K(W) was also observed. There was no other genetic covariation among traits. The first evidence of genetic variation for K(W) within a species was obtained. These results also indicate that there is a genetic basis for the positive association between xylem water transport and photosynthesis. The presence of significant genetic variation and covariation for these traits in natural populations would facilitate correlated evolution between xylem and leaf functions.  相似文献

Summary and conclusions From a study of the composition of the soil and the subsoil under three grasses,Imperata cylindrica, Pennisetum orientale, Pennisetum polystachyum and three legumesTephrosia candida, Medicago sativa andPueraria hirsuta and of those of natural bare soil in the neighbourhood of each, it could be observed that the soils under vegetation contained more moisture, organic matter, organic nitrogen clay and soluble salts but had lower pH values than the bare soils. The soils under grasses had less moisture, lower pH and lower salinity but higher clay content and exhibited greater aggregation than the soils under legumes. Though the soils under grasses had significantly higher quantities of organic matter than the soils under legumes there was no significant difference in the organic nitrogen contents between them.  相似文献

Poly(2-hydroxyethylmethacrylate) (pHEMA) based flat sheet membrane was prepared by UV-initiated photopolymerization technique. The membrane was then grafted with -histidine. Catalase immobilization onto the membrane from aqueous solutions containing different amounts of catalase at different pH was investigated in a batch system. The maximum catalase immobilization capacity of the pHEMA–histidine membrane was 86 μg cm−2. The activity yield was decreased with the increase of the enzyme loading. It was observed that there was a significant change between Vmax value of the free catalase and Vmax value of the adsorbed catalase on the pHEMA–histidine membrane. The Km value of the immobilized enzyme was higher 1.5 times than that of the free enzyme. Optimum operational temperature was 5°C higher than that of the free enzyme and was significantly broader. It was observed that enzyme could be repeatedly adsorbed and desorbed without loss of adsorption capacity or enzyme activity.  相似文献

Genome‐scale metabolic models (GEMs) have proven useful as scaffolds for the integration of omics data for understanding the genotype–phenotype relationship in a mechanistic manner. Here, we evaluated the presence/absence of proteins encoded by 15,841 genes in 27 hepatocellular carcinoma (HCC) patients using immunohistochemistry. We used this information to reconstruct personalized GEMs for six HCC patients based on the proteomics data, HMR 2.0, and a task‐driven model reconstruction algorithm (tINIT). The personalized GEMs were employed to identify anticancer drugs using the concept of antimetabolites; i.e., drugs that are structural analogs to metabolites. The toxicity of each antimetabolite was predicted by assessing the in silico functionality of 83 healthy cell type‐specific GEMs, which were also reconstructed with the tINIT algorithm. We predicted 101 antimetabolites that could be effective in preventing tumor growth in all HCC patients, and 46 antimetabolites which were specific to individual patients. Twenty‐two of the 101 predicted antimetabolites have already been used in different cancer treatment strategies, while the remaining antimetabolites represent new potential drugs. Finally, one of the identified targets was validated experimentally, and it was confirmed to attenuate growth of the HepG2 cell line.  相似文献

Sequential deposition has great potential to achieve high performance in organic solar cells due to the resulting well‐controlled vertical phase separation. In this work, double bulk heterojunction organic solar cells are fabricated by sequential‐blade cast in ambient conditions. Probed by the in situ grazing incidence X‐ray diffraction and in situ UV–vis absorption measurements, the seq‐blade system exhibits a different tendency from each of the binary films during the film formation process. Due to the extensive aggregation of FOIC, the binary PBDB‐T:FOIC film displays a strong and large phase separation, resulting in low current density (Jsc) and unsatisfactory power conversion efficiency. In the seq‐blade cast system, the bottom layer PBDB‐T:IT‐M produces many crystal nuclei for the top layer PBDB‐T:FOIC, so the PBDB‐T molecules are able to crystallize easily and quickly. Balanced crystallization kinetics between polymer and small molecule and an ideal percolation network in the film are observed. In addition, the balanced crystallization kinetics are favorable toward realizing lower recombination loss through charge transport processes.  相似文献
