Community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) is a threat to human health worldwide. Although progress has been made, mechanisms of CA-MRSA pathogenesis are poorly understood and a comprehensive analysis of CA-MRSA exoproteins has not been conducted. To address that deficiency, we used proteomics to identify exoproteins made by MW2 (USA400) and LAC (USA300) during growth in vitro. Two hundred and fifty unique exoproteins were identified by 2-dimensional gel electrophoresis coupled with automated direct infusion-tandem mass spectrometry (ADI-MS/MS) analysis. Eleven known virulence-related exoproteins differed in abundance between the strains, including alpha-haemolysin (Hla), collagen adhesin (Cna), staphylokinase (Sak), coagulase (Coa), lipase (Lip), enterotoxin C3 (Sec3), enterotoxin Q (Seq), V8 protease (SspA) and cysteine protease (SspB). Mice infected with MW2 or LAC produced antibodies specific for known or putative virulence factors, such as autolysin (Atl), Cna, Ear, ferritin (Ftn), Lip, 1-phosphatidylinositol phosphodiesterase (Plc), Sak, Sec3 and SspB, indicating the exoproteins are made during infection in vivo. We used confocal microscopy to demonstrate aureolysin (Aur), Hla, SspA and SspB are produced following phagocytosis by human neutrophils, thereby linking exoprotein production in vitro with that during host-pathogen interaction. We conclude that the exoproteins identified herein likely account in part for the success of CA-MRSA as a human pathogen. 相似文献

Plant traits are particularly important in determining plant community structure. However, how can one identify which traits are the most important in driving community assembly? Here we propose a method 1) to quantify the direction and strength of trait selection during community assembly and 2) to obtain parsimonious lists of traits that can predict species relative abundances in plant communities. We tested our method using floristic data from 32 plots experiencing different treatments (fertilisation and grazing) in southern France. Twelve functional traits were measured on 68 species. We determined the direction and strength of selection on these 12 traits using a metric derived from a maximum entropy model (i.e. lambda). We then determined our parsimonious list of traits using a backward selection of traits based on these lambda values (for all treatments and in each treatment separately). We finally compared our method to two other methods: one based on iterative RLQ and the other based on an entropy‐based forward selection of traits. We found major differences in the direction and strength of selection across the 12 traits and treatments. From the 12 traits, plant vegetative and reproductive heights, leaf dry matter content leaf nitrogen content, specific leaf area, and leaf phosphorus content were particularly important for predicting species relative abundances when considering all treatments together. Our method yielded results similar to those produced by the entropy‐based approach but differed from those produced by the iterative RLQ, whose selected traits could not significantly predict species relative abundances. Together these results suggest that the assembly of these communities is primarily driven by a small number of key functional traits. We argue that our method provides an objective way of determining a parsimonious list of traits that together accurately predict community structure and which, despite its complementarities with entropy‐based method, offers significant advantages. 相似文献

Eggs from Amphibia are surrounded by several layers of jelly that are needed for proper fertilization. Jelly coat is composed of highly glycosylated mucin-type glycoproteins containing up to 60% of carbohydrates, which display a remarkable species-specificity. This material obtained from Triturus alpestris was submitted to reductive beta-elimination and the released oligosaccharide-alditols were further fractionated by HPLC. Structural characterization was performed through a combination of two dimensional (1)H-(1)H and (1)H-(13)C NMR and ESI-MS/MS analysis. Numerous carbohydrate chains are characterized by the presence of the Cad (Sd(a)) determinant, including respectively NeuAc, NeuGc or Kdn as a sialic acid. But the most significant O-glycan sequences which mark the difference between the jelly of T. alpestris and other studies amphibian jellies are polymers of GalNAc(beta 1-4)GlcNAc (LacdiNAc) which form part of the following sequence: HSO(3)(4)(GalNAcbeta 1-4GlcNAcbeta 1-3)(1-3)GalNAcbeta 1-4(GlcNAcbeta 1-3)(0-1)GlcNAcbeta 1-6GalNAc-ol. 相似文献

This article focuses on parameter estimation of multilevel nonlinearmixed-effects models (MNLMEMs). These models are used to analyzedata presenting multiple hierarchical levels of grouping (clusterdata, clinical trials with several observation periods, ...).The variability of the individual parameters of the regressionfunction is thus decomposed as a between-subject variabilityand higher levels of variability (e.g. within-subject variability).We propose maximum likelihood estimates of parameters of thoseMNLMEMs with 2 levels of random effects, using an extensionof the stochastic approximation version of expectation–maximization(SAEM)–Monte Carlo Markov chain algorithm. The extendedSAEM algorithm is split into an explicit direct expectation–maximization(EM) algorithm and a stochastic EM part. Compared to the originalalgorithm, additional sufficient statistics have to be approximatedby relying on the conditional distribution of the second levelof random effects. This estimation method is evaluated on pharmacokineticcrossover simulated trials, mimicking theophylline concentrationdata. Results obtained on those data sets with either the SAEMalgorithm or the first-order conditional estimates (FOCE) algorithm(implemented in the nlme function of R software) are compared:biases and root mean square errors of almost all the SAEM estimatesare smaller than the FOCE ones. Finally, we apply the extendedSAEM algorithm to analyze the pharmacokinetic interaction oftenofovir on atazanavir, a novel protease inhibitor, from theAgence Nationale de Recherche sur le Sida 107-Puzzle 2 study.A significant decrease of the area under the curve of atazanaviris found in patients receiving both treatments. 相似文献

Uncovering relationships between landscape diversity and species interactions is crucial for predicting how ongoing land-use change and homogenization will impact the stability and persistence of communities. However, such connections have rarely been quantified in nature. We coupled high-resolution river sonar imaging with annualized energetic food webs to quantify relationships among habitat diversity, energy flux, and trophic interaction strengths in large-river food-web modules that support the endangered Pallid Sturgeon. Our results demonstrate a clear relationship between habitat diversity and species interaction strengths, with more diverse foraging landscapes containing higher production of prey and a greater proportion of weak and potentially stabilizing interactions. Additionally, rare patches of large and relatively stable river sediments intensified these effects and further reduced interaction strengths by increasing prey diversity. Our findings highlight the importance of landscape characteristics in promoting stabilizing food-web architectures and provide direct relevance for future management of imperilled species in a simplified and rapidly changing world. 相似文献

A Th1 response is required for the development of Plasmodium berghei ANKA (PbA)-induced experimental cerebral malaria (ECM). The role of pro-Th1 IL-12 in malaria is complex and controversial. In this study, we addressed the role of IL-12Rβ2 in ECM development. C57BL/6 mice deficient for IL-12Rβ2, IL-12p40, or IL-12p35 were analyzed for ECM development after blood-stage PbA infection in terms of ischemia and blood flow by noninvasive magnetic resonance imaging and angiography, T cell recruitment, and gene expression. Without IL-12Rβ2, no neurologic sign of ECM developed upon PbA infection. Although wild-type mice developed distinct brain microvascular pathology, ECM-resistant, IL-12Rβ2-deficient mice showed unaltered cerebral microcirculation and the absence of ischemia after PbA infection. In contrast, mice deficient for IL-12p40 or IL-12p35 were sensitive to ECM development. The resistance of IL-12Rβ2-deficient mice to ECM correlated with reduced recruitment of activated T cells and impaired overexpression of lymphotoxin-α, TNF-α, and IFN-γ in the brain after PbA infection. Therefore, IL-12Rβ2 signaling is essential for ECM development but independent from IL-12p40 and IL-12p35. We document a novel link between IL-12Rβ2 and lymphotoxin-α, TNF-α, and IFN-γ expression, key cytokines for ECM pathogenesis. 相似文献

Background
The epidermal cells of the seed coat of certain species accumulate polysaccharides during seed development for cell wall reinforcement or release on imbibition to form mucilage. Seed-coat epidermal cells show natural variation in their structure and mucilage production, which could explain the diverse ecophysiological roles proposed for the latter. Arabidopsis mucilage mutants have proved to be an important tool for the identification of genes involved in the production of seed-coat polysaccharides.Scope
This review documents genes that have been characterized as playing a role in the differentiation of the epidermal cells of the arabidopsis seed coat, the natural variability in polysaccharide features of these cells and the physiological roles attributed to seed mucilage.Conclusions
Seed-coat epidermal cells are an excellent model for the study of polysaccharide metabolism and properties. Intra- and interspecies natural variation in the differentiation of these epidermal cells is an under-exploited resource for such studies and promises to play an important part in improving our knowledge of polysaccharide production and ecophysiological function. 相似文献110.

In plants, sugars are the main respiratory substrates and important signaling molecules in the regulation of carbon metabolism. Sugar signaling studies suggested that sugar sensing involves several key components, among them hexokinase (HXK). Although the sensing mechanism of HXK is unknown, several experiments support the hypothesis that hexose phosphorylation is a determining factor. Glucose (Glc) analogs transported into cells but not phosphorylated are frequently used to test this hypothesis, among them 3-O-methyl-Glc (3-OMG). The aim of the present work was to investigate the effects and fate of 3-OMG in heterotrophic plant cells. Measurements of respiration rates, protein and metabolite contents, and protease activities and amounts showed that 3-OMG is not a respiratory substrate and does not contribute to biosynthesis. Proteolysis and lipolysis are induced in 3-OMG-fed maize (Zea mays L. cv DEA) roots in the same way as in sugar-starved organs. However, contrary to the generally accepted idea, phosphorous and carbon nuclear magnetic resonance experiments and enzymatic assays prove that 3-OMG is phosphorylated to 3-OMG-6-phosphate, which accumulates in the cells. Insofar as plant HXK is involved in sugar sensing, these findings are discussed on the basis of the kinetic properties because the catalytic efficiency of HXK isolated from maize root tips is five orders of magnitude lower for 3-OMG than for Glc and Man. 相似文献
