Dental diseases are among the most prevalent afflictions of humankind. These diseases are associated with the formation of biofilms harboring pathogenic bacteria. Fructosyltransferases (FTF) are extra cellular enzymes of several oral bacteria. FTF are associated with the formation of extracellular polysaccharide matrix (fructans) which play a role in biofilm formation and oral bacteria physiology. Oxazaborolidines have been shown to inhibit biofilm formation. The purpose of this study was to examine if the anti-biofilm effect is, in part, an effect on the immobilized enzymes synthesizing the extra cellular polysaccharide participating in biofilm formation. Eight different oxazaborolidines (BNO1-BNO8) were synthesized and evaluated for their affect on the synthesis of fructans by FTF using the biomolecular interaction analysis (BIAcore) system which involves the use of real-time surface plasmon resonance (SPR) technique. The tested oxazaborolidines demonstrated a significant and immediate inhibitory effect on immobilized FTF activity. This effect was reversible. Our results show that oxazaborolidines can act as enzymatic inhibitors of FTF immobilized on the surface, also at levels lower than their MIC. Part of the anti-biofilm effect of BNOs may be accounted for this enzymatic inhibition.  相似文献

Human papillomavirus (HPV) are well known to be associated with the development of cervical cancer. HPV16 and HPV 18 are known as high-risk types and reported to be predominantly associated with cervical cancer. The prevalence and genetic diversity of HPV have been well documented globally but, in the Kingdom of Saudi Arabia, data on HPV genetic diversity are lacking. In this study, we have analyzed the genetic diversity of both HPV16 and HPV18 based on their L1 gene sequence because L1 gene is a major capsid protein gene and has been utilized to develop a prophylactic vaccine. In January 2011–2012, a total of forty samples from cervical specimens of women in Saudi Arabia were collected. The association of HPV16, HPV18 was detected by polymerase chain reaction, sequenced and submitted to GenBank. The sequences identity matrix and the phylogenetic relationship were analyzed with selected HPVs. The highest sequence identity (99.5%) for HPV16 and (99.3%) for HPV was observed with selected HPVs. The phylogenetic analysis results showed that HPVs from Saudi Arabia formed a closed cluster with African, Asian, East Asian as well as American HPVs distributed into multiple linages from various geographical locations. The results provided the valuable information about genetic diversity, but there is an urgent need to generate full genome sequence information which will provide a clearer picture of the genetic diversity and evolution of HPVs in Saudi Arabia. In conclusion, the generated data will be highly beneficial for developing molecular diagnostic tools, analyzing and correlating the epidemiological data to determine the risk of cervical cancer and finally to develop a vaccine for Saudi Arabian population.  相似文献

A newly soil-isolated Staphylococcus aureus strain secretes a non-induced lipase in the culture medium. The extracellular lipase from S. aureus (SAL3) is purified to homogeneity. The purified enzyme is a tetrameric protein (180 kDa) corresponding to the association of four lipase molecules. The 15 N-terminal amino acid residues showed a high degree of homology with other staphylococcal lipase sequences. The part of the gene encoding the mature SAL3 is cloned and sequenced. The deduced polypeptide sequence, corresponding to the mature SAL3, was very similar to the mature Staphylococcus simulans lipase sequence with two additional amino acid residues (LK) at the N-terminus of SAL3. The lipase activity is maximal at pH 9.5 and 55 °C. The specific activity of about 4200 U/mg or 3500 U/mg was measured using tributyrin or olive oil emulsion as substrate, respectively, at pH 9.5 and 55 °C.In contrast to other staphylococcal lipases previously characterised, SAL3 is found to be stable between pH 5 and 12 after 24 h incubation. The enzyme retained 50% of its activity after 60 min incubation at 60 °C. This novel lipase is able to hydrolyse its substrate in presence of various oxidizing agents as well as some surfactants and some commercial detergents, then SAL3 can be considered as a good candidate for industrial and biotechnological applications.  相似文献

运用16种酶蛋白编码的23个遗传座位对突尼斯非洲跳鼠(Jaculus jaculus)和埃及跳鼠(J. orientalis)自然群体的遗传变异和分化进行了电泳分析。结果表明,与其他啮齿动物等哺乳动物的相关数据比较,发现这两个种群体的遗传变异水平较低。非洲跳鼠群体的观测杂合度 (Hobs) 为0.08—0.19,多态座位百分比(P)为26.2%—45.2%,每个座位的平均等位基因数(A)为1.1—1.4;埃及跳鼠的Hobs为0.10—0.15,P为29.3%—44.1%,A为1.1—1.7。两个种群体各自的遗传分化程度较低(非洲跳鼠和埃及跳鼠的Fst分别为0.0017和0.0019)。而两个种群体间的Fst为0.607(P<0.05),表明两个种之间高度的遗传分化。本研究支持这两个种分类地位的合法性,并强调了地理因素(环境类型和生物气候阶段)对两个种遗传结构的影响。  相似文献

This study aimed to identify the economic dimension of directing treated wastewater to the production of green fodder. To achieve its objectives, the study relied on economic equations, the method of using multiple criteria, and a scoring technique for prioritization. The study showed that green fodder is a crop that depletes water, as it consumes about 67.27% of the water used for agricultural purposes. In determining the priority of regions intending to use treated wastewater, the ranking put Riyadh region first, followed by the eastern region, the Qassim region, Hail, Makkah Al-Mukarramah, Tabuk, Asir, Al-Jawf, Jizan, Madinah, Najran, the northern borders, and Al-Baha, in this order. The area that can be cultivated with forage crops is estimated to be about 53.05 thousand hectares, with a production of 953.75 thousand tons. The cost of treating wastewater to make it usable amounts to 2126.22 million riyals, while the value of the benefits gained from its use is 2508.95 million riyals; thus, the Saudi agricultural economy achieves a net gain estimated at about 382.73 million riyals annually. Therefore, this study recommends the use of treated wastewater in the production of green fodder.  相似文献

The dystroglycans (alpha-DG and beta-DG), which play important roles in the formation of basement membranes, have been well studied in skeletal muscle and nerve, but their expression and localization in intestinal epithelial cells has not been previously investigated. Here, we demonstrated that the DG complex, composed of alpha-DG, beta-DG, and utrophin, is specifically expressed in the basolateral membrane of the Caco-2-BBE monolayer. The DG complex coprecipitated with beta(1)-integrin, suggesting a possible interaction among these proteins. In addition, we observed that activation of DG receptors by laminin-1 enhanced the interaction between beta(1)-integrin and laminin-1, whereas activation of DG receptors by laminin-2 reduced the interaction between beta(1)-integrin and laminin-2. Finally, we demonstrated that the intracellular COOH-terminal tail of beta-DG and its binding to the DG binding domain of utrophin are crucial for the interactions between laminin-1/-2 and beta(1)-integrin. Collectively, these novel results indicate that dystroglycans play important roles in the regulation of interactions between intestinal epithelial cells and the extracellular matrix.  相似文献

Erythroleukemia mitochondria (E. Mito) and chloroma mitochondria (C. Mito) were isolated from tumors grown in their hosts, DBA/2J mice and Long-Evans rats, respectively. Oxypolarographic tests showed respiratory control and ADP/O ratios typical for well-coupled mitochondria. Therapeutic concentration of chloramphenicol (CAP) had no effect on the energy transfer of those mitochondria. l-[14C]leucine incorporation into protein was comparable in both types of mitochondria. Although the incorporation at 15 min appeared higher in C. Mito, at 60 min it became similar to that in E. Mito. When CAP was used at the therapeutic concentration of 20 μg/ml about 80% inhibition was observed in both mitochondria. The exogenous amino acid mixture added to the medium was an important determinant in both the rate of leucine incorporation as well as the sensitivity to CAP. Thus, if no amino acids were added the incorporation was reduced to 18–25%. Under these conditions, however E. Mito were significantly more sensitive to the same concentration of CAP than C. Mito. The results suggest that mitochondrial amino acid pool may be involved in the greater sensitivity of erythroid precursors to CAP.  相似文献

Trivalent chromium (Cr3+) is essential for animal and human health, whereas hexavalent Cr (CrO4 2−) is a potent carcinogen and extremely toxic to animals and humans. Thus, the accumulated Cr in food plants may represent potential health hazards to animals and humans if the element is accumulated in the hexavalent form or in high concentrations. This study was conducted to determine the extent to which various vegetable crops absorb and accumulate Cr3+ and CrO4 2− into roots and shoots and to ascertain the different chemical forms of Cr in these tissues. Two greenhouse hydroponic experiments were performed using a recirculating-nutrient culture technique that allowed all plants to be equally supplied with Cr at all times. In the first experiment, 1 mg L−1 Cr was supplied to 11 vegetable plant species as Cr3+ or CrO4 2−, and the accumulation of Cr in roots and shoots was compared. The crops tested included cabbage (Brassica oleracea L. var. capitata L.), cauliflower (Brassica oleracea L. var. botrytis L.), celery (Apium graveolens L. var. dulce (Mill.) Pers.), chive (Allium schoenoprasum L.), collard (Brassica oleracea L. var. acephala DC.), garden pea (Pisum sativum L.), kale (Brassica oleracea L. var. acephala DC.), lettuce (Lactuca sativa L.), onion (Allium cepa L.), spinach (Spinacia oleracea L.), and strawberry (Fragaria ×  ananassaDuch.). In the second experiment, X-ray absorption spectroscopy (XAS) analysis on Cr in plant tissues was performed in roots and shoots of various vegetable plants treated with CrO4 2− at either 2 mg Cr L−1 for 7 d or 10 mg Cr L−1 for 2, 4 or 7 d. The crops used in this experiment included beet (Beta vulgaris L. var. crassa (Alef.) J. Helm), broccoli (Brassica oleracea L. var. Italica Plenck), cantaloupe (Cucumis melo L. gp. Cantalupensis), cucumber (Cucumis sativus L.), lettuce, radish (Raphanus sativus L.), spinach, tomato (Lycopersicon lycopersicum (L.) Karsten), and turnip (Brassica rapa L. var. rapifera Bailey). The XAS speciation analysis indicates that CrO4 2− is converted in the root to Cr3+ by all plants tested. Translocation of both Cr forms from roots to shoots was extremely limited and accumulation of Cr by roots was 100-fold higher than that by shoots, regardless of the Cr species supplied. Highest Cr concentrations were detected in members of the Brassicaceae family such as cauliflower, kale, and cabbage. Based on our observations and previous findings by other researchers, a hypothesis for the differential accumulation and identical translocation patterns of the two Cr ions is proposed.
