Summary The N-metabolism ofArthrocnemum fruticosum (L.) Moq., growing in a saline area north-east of the Dead Sea in Jordan, was studied over its vegetative growth period from
March to September 1981. Plant and soil samples were taken at monthly intervals. Water content, Na+, K+, Cl−, NH
4
+
, NO
2
−
and NO
3
−
concentrations were determined in the soil extracts, and the same determinations plus ash weight, soluble carbohydrates,
proline, proteins andin vivo nitrate reductase in the plant roots and shoots.
Soil humidity decreased and salinity increased from March to August, with re-wetting occurring in late July. K+ and Cl− were much lower in the soils than Na+. Plant relative dry weight increased during summer due to the absorption of Na+ in addition to increased organic dry weight. The uptake of Na+ was not balanced by a similar uptake of Cl−.
Ammonium and nitrate decreased in soil and plants in parallel with increasing salinity. Nitrite was only found in the roots
and always in very low quantities. Proline was found only in March. The total soluble carbohydrates in the roots showed a
short increase in June when the sodium in the plants also increased. It was concluded that carbohydrates may be used to balance
osmotic shocks, but that another compatible compounds is necessary to maintatin long-term osmotic equilibrium.
The nitrate reductase activity, measuredin vivo, and the soluble protein changed roughly in parallel with the internal nitrate from May to August, suggesting that nitrogen
uptake and reduction in the plant is inhibited during summer when the soil is dry and very saline. This could be a direct
effect of drought and/or salinity on the plants, or an indirect onevia an inhibition of nitrifying bacteria. 相似文献

Differentiation process of mesenchymal stem cells (MSCs) into adipocyte is involved in obesity. Multiple factors such as Ca2+ play important roles in different stages of this process. Because of the complicated roles of Ca2+ in adipogenesis, the aim of present investigation was to study the influx and efflux of Ca2+ into and out of the cells during adipogenesis. Adipose-derived MSCs were used to differentiate into adipocytes. MSCs were exposed to 2.5 mM Ca2+ or 1.8 mM Ca2+ plus calcium ionophore, A23187, for 3 days. Lipid staining, triglycerides (TG) content, and glyceraldehyde phosphate dehydrogenase (GAPDH) activity were evaluated to confirm the efficiency of the differentiation. Gene expression of GLUT4, PPARγ2, RAR-α, and calreticulin, as well as the protein levels of GLUT4 and PPARγ2 were determined. Ca2+ and in particular Ca2+ plus A23187 significantly lowered the efficiency of differentiation accompanied by decrease in intracellular TG deposits, GAPDH activity and alleviation of gene, and protein levels of GLUT4 and PPARγ2. While calreticulin and RAR-α were remarkably upregulated in A23187 group. This study showed the inhibitory effects of calcium in adipogenesis. Additionally, it indicated the greater inhibitory effect of calreticulin and RAR-α in controlling adipogenesis by higher levels of calcium. 相似文献

Mutational inactivation of p53 is a hallmark of most human tumors. Loss of p53 function also occurs by overexpression of negative regulators such as MDM2 and MDM4. Deletion of Mdm2 or Mdm4 in mice results in p53-dependent embryo lethality due to constitutive p53 activity. However, Mdm2(-/-) and Mdm4(-/-) embryos display divergent phenotypes, suggesting that Mdm2 and Mdm4 exert distinct control over p53. To explore the interaction between Mdm2 and Mdm4 in p53 regulation, we first generated mice and cells that are triple null for p53, Mdm2, and Mdm4. These mice had identical survival curves and tumor spectrum as p53(-/-) mice, substantiating the principal role of Mdm2 and Mdm4 as negative p53 regulators. We next generated mouse embryo fibroblasts null for p53 with deletions of Mdm2, Mdm4, or both; introduced a retrovirus expressing a temperature-sensitive p53 mutant, p53A135V; and examined p53 stability and activity. In this system, p53 activated distinct target genes, leading to apoptosis in cells lacking Mdm2 and a cell cycle arrest in cells lacking Mdm4. Cells lacking both Mdm2 and Mdm4 had a stable p53 that initiated apoptosis similar to Mdm2-null cells. Additionally, stabilization of p53 in cells lacking Mdm4 with the Mdm2 antagonist nutlin-3 was sufficient to induce a cell death response. These data further differentiate the roles of Mdm2 and Mdm4 in the regulation of p53 activities. 相似文献

A new series of NSAID thioesters were synthesized and evaluated for their in vitro antitumor effects against a panel of four human tumor cell lines, namely: HepG2, MCF-7, HCT-116 and Caco-2, using the MTT assay. Compared to the reference drugs 5-FU, afatinib and celecoxib, compounds 2b, 3b, 6a, 7a, 7b and 8a showed potent broad-spectrum antitumor activity against the selected tumour cell lines. Accordingly, these compounds were selected for mechanistic studies about COX inhibition and kinase assays. In vitro COX-1/COX-2 enzyme inhibition assay results indicated that compounds 2b, 3b, 6a, 7a, 7b, 8a and 8?b selectively inhibited the COX-2 enzyme (IC50?=?~0.20–0.69?μM), with SI values of (>72.5–250) compared with celecoxib (IC50?=?0.16?μM, COX-2 SI:?>?312.5); however, all the tested compounds did not inhibit the COX-1 enzyme (IC50?>?50?μM). On the other hand, EGFR, HER2, HER4 and cSrc kinase inhibition assays were evaluated at a 10?μM concentration. The selected candidates displayed limited activities against the various tested kinases; the compounds 2a, 3b, 6a, 7a, 7b and 8a showed no activity to weak activity (% inhibition?=?~0–10%). The molecular docking study revealed the importance of the thioester moiety for the interaction of the drugs with the amino acids in the active sites of COX-2. Leaf curl and yellow vein mosaic viral disease is the major constraint on okra (Abelmoschus esculentus L.) production in India. Amplified fragment sequence of DNA-β showed highest similarity of 91.7% with Bhendi yellow vein mosaic virus-Tamil Nadu (AJ308425, NC_003405) and lowest similarity of 48.5% with OKLCV (NC_004093), whereas coat protein specific amplified sequence showed highest homology with isolate of Madurai, Haryana, Ludhiana and lowest homology of 92% with Mesta yellow vein mosaic Bahraich virus (MYVMBV) (EU360303). The results obtained in the present study confirm that both the viral diseases of okra reported in southern India are caused by a begomovirus associated with DNA-β in which the plants show leaf curl symptoms and never develops yellow vein mosaic and those plants which show yellow vein mosaic, never develops leaf curl symptoms even in the same rows and field. The efficacy of different concentrations of the commercial neem-based insecticide, Nimbicidine® and the ecdysone agonist compound Methoxyfenozide (RH-2485), was evaluated against larvae of Spodoptera littoralis (Boisduval) (Lepidoptera: Noctuidae). RH-2485 and Nimbicidine® significantly (p?<?0.001) reduced adult longevity by 2.7 and 1.9 d at the higher concentrations tested, respectively, but no significant differences were observed at low concentration. The tested compounds strongly affected the reproduction of S. littoralis by producing a high percentage of sterility. Fecundity and fertility were significantly affected by both insecticides. No egg laying has been recorded with the higher concentration 0.5% of Nimbicidine® and 0.1?ppm of RH-2485, while no significant (p?>?0.05) difference was noticed on total number of eggs laid by the female when lower concentration 0.0001?ppm of RH-2485 was applied in the same stage as compared to the control. In the second part of this study, the invasion rate of Steinernema feltiae was affected by the addition of both Nimbicidine and RH-2485 to the diet of experimental host. At higher concentration of both compounds, the invasion rate was decreased despite the infection rate, while the percentage of invading nematodes increase to 56.7% with the combination treatment indicating a considerable improvement in the efficacy of S. feltiae nematode applied in combination with the lower concentration of Nimbicidine® over that of nematode alone. On the other hand, the invasion of S. feltiae to the insects that were fed on the diet with the addition of ecdyson compound was strongly decreased with increasing concentration of RH-2485 in host diet. 相似文献
