Abstract Some strains of motile Aeromonas produce lectin-like adhesins, whose activity can be inhibited by d -mannose. Such strains can co-agglutinate with some strains of Salmonella . Whether or not co-agglutination occurs is dependent upon both the properties of the Aeromonas adhesin and the structure of the Salmonella lipopolysaccharide (LPS). These studies have enabled new structural information for Salmonella LPS to be deduced and have confirmed previous studies regarding the nature of the Aeromonas adhesin binding site. It is possible that the observed in vitro co-agglutination between Aeromonas and Salmonella is a reflection of an in vivo situation which could modify the virulence of either or both bacteria.  相似文献

Techniques were developed for genetic characterization of Legionella pneumophila serogroup 1 by using restriction fragment length polymorphism analysis. Allozyme analysis provided an index of the discrimination achieved by restriction fragment length polymorphism. Isolates from human cases of legionellosis were examined by both methods, and their profiles were compared with reference strains of L. pneumophila serogroup 1 obtained from the American Type Culture Collection. Eighteen distinct clones were evident among the isolates examined. Both methods could be used to trace the source of an outbreak of legionellosis caused by L. pneumophila serogroup 1.  相似文献

The detection of DNA polymorphisms by RFLP analysis is having a major impact on identity testing in forensic science. At present, this approach is the best effort a forensic scientist can make to exclude an individual who has been falsely associated with an evidentiary sample found at a crime scene. When an analysis fails to exclude a suspect as a potential contributor of an evidentiary sample, a means should be provided to assess suitable weight to the putative match. Most important, the statistical analysis should not place undue weight on a genetic profile derived from an unknown sample that is attributed to an accused individual. The method must allow for limitations in conventional agarose-submarine-gel electrophoresis and Southern blotting procedure, limited sample population data, possible subpopulation differences, and potential sampling error. A conservative statistical method was developed based on arbitrarily defined fixed bins. This approach permits classification of continuous allelic data, provides for a simple and portable data-base system, and is unlikely to underestimate the frequency of occurrence of a set of alleles. This will help ensure that undue weight is not placed on a sample attributed to an accused individual.  相似文献

Summary Complete hydatidiform moles contain only paternal chromosomes. To learn more of their origin, we used restriction endonuclease site polymorphisms found in the parental mitochondrial DNAs to demonstrate that moles contain exclusively maternal mitochondrial DNA. Thus, moles must arise from the fusion of one or two sperm with a mature but anucleate ovum.  相似文献

Eimeria nuttalli oocysts were found in 58% (21/36) and E. procyonis oocysts in 25% (9/36) of raccoons Procyon lotor in Illinois, and sporocysts of Sarcocystis sp. in 17% (2/12) of other raccoons in Illinois. The oocysts of E. nuttalli were ellipsoidal to ovoid, 15-21 X 12-17 micrometer, with a one-layered, smooth, colorless wall. The oocysts of E. procyonis were 22-28 X 18-22 micrometer, with a rough, striated, brownish, two-layered wall. The sporulated sporocysts of Sarcocystis sp. were 11-13 X 8-10 micrometer. Attempts to infect baby pigs by feeding them sporocysts of Sarcocystis sp. from the reaction failed.  相似文献

An improved method of purification of DNA methylase from Krebs II ascites cells is reported. The enzyme sediments at 8.3 S on glycerol-gradients and a major band on SDS polyacrylamide gel electrophoresis has a molecular weight of 184 000. Aggregation occurs at low salt and this may interfere with enzymic activity. The preferred double stranded DNA substrate is that rendered partially unmethylated by an in vitro repair mechanism or by isolation from methionine starved cells. Methylation of native partially methylated DNA is favoured under conditions of low salt and high temperature; conditions which encourage 'breathing' of the DNA. Methylation of native, unmethylated DNA also involves breathing but results in formation of a salt resistant tight binding complex between the enzyme and the DNA.  相似文献
