Malate enzyme (l-malate : NADP+ oxidoreductase (oxalacetate-decarboxylating, EC 1.1.1.40)) has been purified from Pseudomonas putida to 99 per cent homogeneity by heat, ammonium suphate fractionation, gel filtration and anion exchange chromatography. Sodium dodecylsulphate-(SDS)-polyacrylamide disc gel electrophoresis analysis showed an approximate tetrameric subunit with a molecular weight of 52,000. The purified enzyme showed a pH optimum between 8.0 and 8.5 (for Tris-HCl buffer) and required bivalent cations for catalysis ; monovalent ions like K+ and NH4+ acted as very effective activators. The temperature-activity relationship for the malate enzyme from 35–80 °C showed broken Arrhenius plots with an inflexion at 65 °C. The enzyme halflife was 30s at 85 °C.The enzyme showed hyperbolic kinetics for both substrates with apparent Km values of 4.0 × 10−4 M and 2.3 × 10−5 M for l-malate and NADP+ respectively. From the study of the effects of some compounds on the enzyme, the physiological significance of those produced by fumarate, succinate and oxalacetate can be emphasized. 相似文献

The tumor promoter 12-O-tetradecanoyl-phorbol-13-acetate is mitogenic for normal chicken embryo fibroblasts and also causes these cells to express transiently many properties of cells transformed by Rous sarcoma virus. Since some mitogenic hormones stimulate a tyrosine-specific protein kinase activity, and since the transforming protein of RSV is a tyrosine-specific protein kinase, we have examined whether TPA also stimulates protein phosphorylation on tyrosine. We report here that TPA treatment of normal cells resulted in a very rapid phosphorylation on tyrosine of a protein peak of Mr 40 to 43 kilodaltons. Thus, a similar biochemical activity (tyrosine phosphorylation) is associated with the action of polypeptide mitogenic hormones, Rous sarcoma virus and a tumor promoter. In addition, TPA treatment resulted in rapid changes in phosphorylation of proteins on serine and threonine. 相似文献

Summary Three different diallel crosses were studied in Cicer arietinum; two of size 6×6, one within each of the two botanical groups macrosperma and microsperma of the cultivated subspecies, and one of 9×9 involving lines covering most of the morphological variation of chickpea. Barriers to crossability present neither a botanical nor a geographical pattern, being probably a direct consequence of interactions between genotypes. The genetic systems of twelve quantitative characters were analysed. Full dominance in a negative sense (small values dominant) is shown by leaflet length, width and shape index, rachis length, leaflet density on the rachis and pod length. Full dominance in a positive sense is shown by seeds per pod. Overdominance (in a positive sense) is evident for pods, seeds and yield per plant. Weak reciprocal differences were manifested by pod length, and pods, seeds and yield per plant. The system controlling number of leaflets per leaf is not clear. Dominance of primitive over selected characters seems to be the rule. As far as the environmental effects have permitted the analysis, no differences in genetic systems were observed between botanical groups. 相似文献

The effect of elongation factor (EF) Tu, bound to the ribosome with the help of poly(uridylic) acid, Phe-tRNA and guanyl-5-yl methylene diphosphonate, on the conformation and/or chemical environment of ribosomal proteins has been examined using, as a probe, protein iodination. Ribosomes complexed only with poly(uridylic acid) and Phe-tRNA have been used as a control. The enzymatic activity of human erythrocyte pyruvate kinase was found to decrease on incubation of the purified enzyme with red blood cell ghosts, ATP and cAMP. If was used radioactivity was found associated with the protein after gel electrophoresis. Various effectors protected the enzyme against phosphorylation. Treatment of the modified enzyme with a protein phosphatase restored enzymatic activity and also caused the loss of the radioactive label. Modification of the pyruvate kinase in this way altered the affinity of the enzyme for one of its substrates (phosphoenolpyruvate), but the binding of the other substrate (ADP) was unaffected. 相似文献

A 5 10/12 year-old boy with a 47,XYY karyotype, micropenis, scrotal hypospadias and right testicular regression is described. Fifteen lactating Holstein cows were randomly allotted to receive either 0 mg (group 0), 32 mg (group 1) or 50 mg (group 2) porcine follicle stimulating hormone (FSH-P) injected in 10 fractions at 12 hr intervals beginning on day 9 of the estrous cycle. All cows received 25 mg prostaglandin (PG) on day 11. Jugular blood samples were collected from cows in all groups at 6 hr intervals beginning on day 7 and continuing through expression of estrus. Mean duration to occurrence of estrus and preovulatory LH surge after PG injection was reduced (P<.05) by injection of FSH-P. Mean number of ovulations increased (P<.05) progressively with increased dose of FSH-P. Mean peripheral progesterone declined more uniformly in FSH-P treated cows after PG and increased earlier (P<.05) after estrus in group 2 cows compared to group 0 and 1 cows. Mean plasma estradiol-17beta elevated (P<.05) after PG injection in both FSH-P-treated groups compared to group 0 cows. Both LH and FSH increased (P<.05) for 36 hr after initiation of FSH-P injection in groups 1 and 2, then declined until after PG injection. Peak LH and FSH occurred more uniformly following PG in treated cows. Results indicate that FSH-P increased endogenous gonadotropin release, estradiol-17beta, ovulation rate and reduced duration to estrus and preovulatory gonadotropin release after PG. Injection of 50 mg FSH-P increased plasma estradiol-17beta and ovulation rate compared to injection of 32 mg FSH-P. 相似文献

A simple procedure for the simultaneous determination of morphine and monoamine transmitters was developed. The procedure consisted of (1) n-butanol extraction and (2) separation and quantitative determination by means of high-performance liquid chromatography combined with electrochemical detection. The maximum intracerebral concentration (210 ± 35 ng/g wet tissue) of morphine was detected 30 min after intramuscular injection (10 mg/kg), which agreed with previous research. Noradrenaline was significantly decreased by morphine injection, while dopamine and 5-hydroxytryptamine were unchanged. However, 3-methoxytyramine, a metabolite of dopamine, was increased, suggesting that the drug increased the turnover rate of dopamine. The procedure used revealed a direct correlation between pharmacokinetics (e.g., distribution of morphine) and pharmacodynamics (e.g. changes of monoamine concentrations) of the drug in vivo. 相似文献

Using a combination of gel filtration and hydroxyapatite chromatography, a major secretory glycoprotein (EP140) was purified from rabbit epididymal fluid. The protein had an apparent Mr of approximately 140 kDa under native conditions but dissociated into 2 equimolar amounts of glycosilated subunits, α and β of Mr 35 and 33 kDa, upon sodium dodecylsulfate polyacrylamide gel electrophoresis in absence of reducing agents. Thus EP140 appears to be a tetramer composed of 2 α and 2 β subunits, held together by noncovalent forces. Proteolytic peptide mapping, amino acid analysis, and determination of partial aminoacid sequences suggested that the 2 subunits were very similar, differing only at some punctual residues in their primary structures. The amino acid sequences obtained did not show significant similarity to any known protein. Western blot determinations with a specific antibody indicated that no EP140-immunorelated protein was detected either in testis or blood from the rabbit nor in epididymides from rats or hamsters. EP140 was shown to associated to the spermatozoon surface, mainly at the acrosomal zone and in the middle piece, and this association progressively increased during the transit of the spermatozoon through the epididymis. Glutathione (GSH) has been shown to play an important role in embryo development. In a previous study, we demonstrated that cysteamine supplementation of in vitro maturation (IVM) medium increased the intracellular GSH content in bovine oocytes and improved subsequent embryo development to the blastocyst stage. The present study was carried out to evaluate the effect of inhibition by buthionine sulfoximide (BSO) of GSH synthesis during IVM in the presence of cysteamine, on subsequent embryo development, and the effect of cysteamine during IVM on the survival of blastocysts following freezing. The effect of β-mercaptoethanol and cysteine added to the maturation medium on GSH levels in bovine oocytes, as well as the effect of these compounds on de novo GSH synthesis by oocytes during in vitro maturation, was also studied. The inhibitory effect of BSO during in vitro maturation on GSH synthesis was also evaluated. Evidence was found confirming that GSH synthesis occurs intracellularly during IVM of oocytes and is stimulated by cysteamine, β-mercaptoethanol and cysteine. Moreover, the present results suggest that the increase in the rate of embryo development exerted by cysteamine, when present during IVM, was due to its stimulatory effect on GSH synthesis. This increase in GSH levels during IVM improves embryo development and quality, producing more embryos reaching the blastocyst stage on day 6, those most suitable for freezing. © 1996 Wiley-Liss, Inc. 相似文献
