Summary Freshly isolated hepatocytes from neonatal rats were cultured for approximately 24 h; incubated for 5, 30, or 60 min in solutions containing 0, 50, 100, or 200 μM cadmium; embedded in plastic; and sectioned for optical microscopy. The exeent of cadmium-induced hepatotoxicity was evaluated by double-blind morphometric analysis (a geometricostatistical processing of two-dimensional data for the collection of threedimensional information) whereby hepatocytes were classified on the basis of the severity of morphologic damage at the optical level. Both time and concentration effects were studied. Cultures exposed to 200 μM cadmium, for various intervals of time from 5 to 60 min, showed statistically significant reductions in the relative volume percent of normal hepatocytes, elevations (then reductions) in the relative volume percent of slightly damaged hepatocytes, increases in the relative volume percent of moderately damaged cells, and increases in the relative volume percent of severely damaged liver cells. As the concentration of cadmium was increased from 50 to 200 μM cadmium (during both 30 and 60-min exposures), significant trends were observed in cellular distribution patterns based on relative volume percent. Morphologically normal cells decreased, both slightly damaged and moderately damaged cells increased, and severely damaged cells remained unchanged. These results indicated that morphometric analysis at the optical level provided quantitative estimates for the evaluation of time- and concentration-effects of cadmium on cultured hepatocytes. Methylmethacrylate (MMA) is the most commonly used embedding medium for sectioning undecalcified bone; however, a number of problems exist with its use in a research laboratory. MMA requires a long infiltration time and temperature control, and it reacts with many polymers. We used Kleer Set resin™ as an alternative embedding medium for sectioning undecalcified bone specimens. Fluorochrome labeled bone specimens were sectioned transversely using a ground section technique and longitudinally on a sledge macrotome. The slides were viewed using both transmitted light and epifluorescence microscopy. High quality sections were obtained using Kleer Set resin™ for both sectioning techniques. We have shown that this new embedding medium is simpler, safer, quicker to use and does not interfere with visualization of fluorochromes.  相似文献

Histophilus somni (Haemophilus somnus) has been reported as the cause of epididymitis in rams. This bacterium has also been found in the preputial mucosa of rams without epididymitis lesions. H. somni is a bacterium that is difficult to characterize, since it is a pleomorphic Gram-negative bacilli of characteristics similar to Actinobacillus seminis, which is also found in ram epididymitis lesions. The objective of this work was to determine if H. somni (H. somnus) is involved in cases of sheep epididymitis. A clinical examination was performed in 160 rams, extracting semen by electro-ejaculation of 28 of them, which had epididymal lesions. The penis was exteriorized in order to avoid prepuce contamination. The semen samples were cultivated in chocolate agar in a 10% CO2 environment. Two strains were isolated in pure culture with a colony morphology and microscopy similar to H. somni (H. somnus). These were identified using the API 20 E system, using as a control the reference strain of H. somnus (2336ATCC). One of the isolates (129H) resulted identical to the reference strain and the other (827) presented differences in the arginine decarboxylase, H2S, catalase and inositol reactions, although these differences have been reported (in strains isolated from different geographic origins, animal species and anatomical region). To characterize the isolates, an electrophoretic analysis of total proteins was performed (PAGE–SDS) finding identical profiles between the reference strain of H. somnus and isolate 129H and similar in relation to isolate 827. The amplification of a fragment of approximately 407 bp was observed in the 129H isolate and the ATCC strain, but not in 827. In other samples, isolations were made of Brucella ovis, Corynebacterium spp., Staphylococcus and other pleomorphic Gram-negative bacilli similar to A. seminis. Therefore, it has been confirmed that H. somni is present in the reproductive tract of rams and it could be involved in the presentation of ovine epididymitis. It is important that we underline that this is the first report of H. somni isolation in Mexico from ram semen samples.  相似文献

Different traces occur on fossil bones and teeth coming from the Early Miocene Gaiman Formation (Patagonia, Argentina). Most traces were attributed to the action of terrestrial and marine predators and scavengers. However, other traces on bones and teeth from this unit and one tooth from the Eocene La Meseta Formation (Antarctica) are attributed to chemical corrosion by lichens in recent times, that is, in a very late diagenetic time. The living lichens and calcium oxalate deposits occurring on the traces and their particular pattern indicates that they were not produced by vegetal roots. The lichens include reproductive structures which allowed a proper determination. A kind of corrosion pattern (Type 1) on bones and teeth from Patagonia is associated to Sarcogyne orbicularis Körber, Verrucaria sp. Schrad, and Buellia aff. punctiformis (Hoff.) Massal. The lichen Aspicilia aff. aquatica produced rounded holes on an Antarctic tooth (Type 2). On the same tooth, the epilithic lichen Caloplaca sp. Th. Fries did not leave any kind of mark on the enameloid.  相似文献

In women, ovarian hormone loss at menopause has been related to cognitive decline, and some studies suggest that estrogen-containing hormone therapy (HT) can mitigate these effects. Recently, the Women's Health Initiative study found that conjugated equine estrogens, the most commonly prescribed HT, do not benefit cognition. Isolated components of conjugated equine estrogens (tradename Premarin®) have been evaluated in vitro, with delta8,9-dehydroestrone (?8E1) and equilin showing the strongest neuroprotective profiles. It has not been evaluated whether ?8E1 or equilin impact cognition or the cholinergic system, which is affected by other estrogens and known to modulate cognition. Here, in middle-aged, ovariectomized rats, we evaluated the effects of ?8E1 and equilin treatments on a cognitive battery and cholinergic nicotinic receptors (nAChR). Specifically, we used 125I-labeled epibatidine binding to assay brain nicotinic receptor containing 4α and 2β subunits (α4β2-nAChR), since this nicotinic receptor subtype has been shown previously to be sensitive to other estrogens. ?8E1 enhanced spatial working, recent and reference memory. ?8E1 also decreased hippocampal and entorhinal cortex α4β2-nAChR expression, which was related to spatial reference memory performance. Equilin treatment did not affect spatial memory or rat α4β2-nAChR expression, and neither estrogen impacted 86Rb+ efflux, indicating lack of direct action on human α4β2 nAChR function. Both estrogens influenced vaginal smear profiles, uterine weights, and serum luteinizing hormone levels, analogous to classic estrogens. The findings indicate that specific isolated Premarin® components differ in their ability to affect cognition and nAChR expression. Taken with the works of others showing ?8E1-induced benefits on several dimensions of health-related concerns associated with menopause, this body of research identifies ?8E1 as a new avenue to be investigated as a potential component of HT that may benefit brain health and function during aging.  相似文献

A four-year study on the interactions between plants and seed-harvesting ants in a Mediterranean pasture is reviewed in this paper. As previously reported in many studies on plant–herbivore interactions, ant–plant relationships are also asymmetric; plants had a larger impact on herbivore dynamics than vice versa. However, the asymmetry did not refer to population dynamics but rather to animal foraging strategies. Ants did not exert a significant influence on vegetation dynamics in terms of plant abundance. The main constraints underlying vegetation change were self-regulation and rainfall. In contrast, the structural characteristics and abundance of vegetation had a significant impact on several important features of food harvesting by ants. This influence was not only associated with their feeding requirements but also with their foraging activities.  相似文献

Venoms of Colubridae snakes are a rich source of novel compounds, which may have applications in medicine and biochemistry. In the present study, we describe the purification and characterization of a metalloproteinase (patagonfibrase), the first protein to be isolated from Philodryas patagoniensis (Colubridae) snake venom. Patagonfibrase is a single-chain protein, showing a molecular mass of 53,224 Da and an acidic isoelectric point (5.8). It hydrolyzed selectively the Aα-chain of fibrinogen and when incubated with fibrinogen or plasma, the thrombin clotting time was prolonged. Prominent hemorrhage developed in mouse skin after intradermal injection of patagonfibrase. When administered into mouse gastrocnemius muscle, it induced local hemorrhage and necrosis, and systemic bleeding in lungs. Patagonfibrase showed proteolytic activity toward azocasein, which was enhanced by Ca2+ and inhibited by Zn2+, cysteine, dithiothreitol and Na2EDTA. Patagonfibrase impaired platelet aggregation induced by collagen and ADP. Thus, patagonfibrase may play a key role in the pathogenesis of disturbances that occur in P. patagoniensis envenomation, and may be used as a biological tool to explore many facets of hemostasis.  相似文献
