Fibroblast growth factor 2 (FGF2) has been assigned a role in melanocyte proliferation and in development of human cutaneous melanoma. We have used a transgenic mouse melanoma model in combination with mice lacking mouse FGF2 to analyse the possible implication of FGF2 in melanomagenesis. Tyr::N‐rasQ61K transgenic mice which are deficient for FGF2 and the tumor suppressors p16INK4a and p19ARF are hyperpigmented and develop cutaneous metastasizing melanoma, with no difference to mice wildtype for FGF2. We conclude from our data, that FGF2 is not essential for melanoma progression and metastasis.  相似文献

The use of listeriaphages as a means of disinfecting contaminated stainless-steel and polypropylene surfaces was investigated. Surfaces artificially contaminated with L. monocytogenes 10401 and 8427 were sanitized with suspensions of listeriaphages (H387, H387-A, and 2671), all belonging to the Siphoviridae family. Phage suspensions at concentrations of up to 3.5 x 10(8) PFU/ml were at least as efficient as a 20 ppm solution of a quaternary ammonium compound (QUATAL) in reducing L. monocytogenes populations. A synergistic activity was observed when two or more phages were used in combination and when phages were suspended in QUATAL. The biological activity of the three phages was not affected by QUATAL concentrations of 50 ppm and a contact time of 4 h.  相似文献

Algae are capable of accumulating nutrients from aqueous waste, which makes them a potential fertilizer. The ability of the fast growing Chlorella vulgaris strain IPPAS C1 to accumulate phosphorus (P) was probed in V-shaped plastic foil photobioreactors. The P uptake was 0.13–0.53 g(P)·m?2·day?1 when the algal culture densities were kept between 0.1 and 1.0 g(DW)·L?1 in a typical summer irradiance of Central Europe. The algal biomass can be effectively utilized for soil fertilization only if the algal cells release nutrients into the soil in a form that would be available to roots and at a rate sufficient to support plant growth. To examine this, we compared the growth of wheat, Triticum aestivum L., in two nutrient-deficient substrates: “Null Erde” and sand, with and without fertilization by wet and spray-dried algae. Plants grown in the two nutrient-deficient substrates supplemented by mineral fertilizer served as a control representing optimal nutrient supply. Plants grown in a high-nutrient substrate (SoMi 513) were used as an additional reference representing the maximum growth potential of wheat. Wheat growth was monitored for 8 weeks and measured, including the increase of the leaf area as well as shoot and root dry weight in 10 randomized replicates for each substrate and fertilization variant. After harvest, the biomass and N, P, and C contents of the plant shoots and roots were recorded. Algae fertilization of “Null Erde” led to wheat growth, including root hair production, which was similar to mineral-fertilized “Null Erde” and only slightly less vigorous than in the nutrient-rich SoMi 513 substrate. The plants grown in sand were smaller than the plants in “Null Erde” but fertilization by algae nevertheless led to growth that was comparable to mineral fertilizer. These results unambiguously demonstrate that algal biomass is a viable option for delivering nutrients to support agriculture on marginal soils.  相似文献

长江航运业的快速发展导致长江中船舶数量激增,相应的水体噪声污染可能对同水域的长江江豚(Neophocaena asiaeorientalis asiaeorientalis)产生一定的负面影响,本研究采用宽频录音设备对长江和畅洲北汊非正式通航江段的各类常见大型船舶(长>15m且宽>5m)的航行噪声进行了记录,并分析其峰值-峰值声压级强度(SPLp-p)和功率谱密度(PSD)等。结果表明,大型船舶的航行噪声能量分布频率范围较广(>100kHz),但主要集中于中低频(<10kHz)部分,各频率(20Hz~144kHz)处的均方根声压级(SPLrms)对环境背景噪声在该频率处的噪声增量范围为3.7~66.5dB。接收到的1/3倍频程声压级(TOL)在各频率处都大于70dB,在8~140kHz频段内都高于长江江豚的听觉阈值。说明大型船舶的航行噪声可能会对长江江豚个体间的声通讯及听觉带来不利影响,如听觉掩盖。  相似文献

To investigate the effects of factors secreted by different cell lines on human monoclonal antibody (MAb) integrity, 600 mg of a human MAb, which specifically binds to human erythrocytes, were produced in a perfusion process. After purification by protein A affinity chromatography, the MAb was used for integrity testing in supernatants of several cell lines to investigate their potential to degrade the antibody in the extracellular environment. One insect cell line (IPLB-SF-21 AE) and four mammalian cell lines [CHO K1, BHK-21 (C13), C1271, P3-X63-Ag8.653], all of them commonly used for the production of recombinant proteins, and the human-human-mouse heterohybridoma cell line itself (H-CB-hahE), were adapted to serum-free culture media. For integrity testing all cell lines were cultivated in spinner flasks using serum-free media supplemented with 30 mug mL(-1) of purified MAb. MAb integrity was assayed by SDS polyacrylamide gel electrophoresis (SDS-PAGE), isoelectric focusing, both followed by Western blotting, and an antigen binding assay. None of the mammalian cells showed any detectable effects on antibody stability and integrity during exponential growth, whereas isoelectric focusing of monoclonal antibody taken from IPLB-SF-21 AE culture supernatants revealed a new band indicating a partial modification of the MAb by secreted factors of these cells. This observation did not correlate with the total proteolytic activity, which was measured in all supernatants and found to be lowest in the insest cell cultures. For mammalian cell cultures, it could be concluded from these findings that shifts of the antibody microheterogeneity pattern, which can be found normally as a result of variations in different production parameters, are not caused by extracellular factors once the product has been secreted into the supernatant. In addition to their well-known advantages in posttranslational modifications (e.g., formation of complex type N-glycans), mammalian cells appear to be more suitable as expression systems for human monoclonal antibodies to be used in vivo when compared with baculovirus-infected insect cells. 用蛋白质内源荧光、疏水荧光探针TNS及蛋白酶K限制性酶解等方法研究了二氢叶酸还原酶在盐酸胍变性过程中的构象变化及动力学,并与活力变化进行了比较.TNS可以监测到与激活同步的构象变化;盐酸胍浓度大于0.75mol/L时,二氢叶酸还原酶被蛋白酶K水解速度增大;当盐酸胍浓度大于1.2mol/L时,才能监测到酶分子整体构象的变化.以上结果表明二氢叶酸还原酶在盐酸胍溶液中的变性并不符合标准的二态模型,而是先经历构象逐步松散的序变过程,然后发生协同的构象伸展.二氢叶酸还原酶在低浓度盐酸胍溶液中的激活是由于酶活性部位构象的微小变化引起的.酶活性部位构象的变化虽然降低了酶与废物的结合能力,但加快了酶促反应限速步骤,即底物解离速度而使酶活力升高.  相似文献

Full grown oocytes derived from Bufo Bufo gargarizans rearing at high temperature environment (24℃), never underwent GVBD after progesterone treatment.No p34^cdc2 Hl kinase activity was detected in the oocytes after progesterone stimulation or OA microinjection;Western blotting analysis showed that the level of p34^cdc2 and p33 in the oocytes are significantly lower than those in the oocytes derived from the hibernating toads (below 10℃).^35S-Met incorporation analysis showed that when the oocytes were incubated at 6℃,synthesis of about thirty defferent polypeptides was promoted or induced,including p34^cdc2 and some other p13^suc1-binding proteins.All these results indicated that a low temperature environment is essential for the oocytes of Bufo Bufo gargarizans to express and stord some cell cycle drivers and its regulators,and to gain the maturation competence.These results will also provide a nwe clue for explaining the molecular mechanisms why gametogenesis of some organisms depends on a relative low temperature and how to maintain the geographical distribution of some animals.  相似文献

Six phages were isolated from sewage and from a lysogenic strain. Three of them, belonging to a new morphological group, had contractile tails and elongated heads with axial ratios of 2.4:1. Two phages, possessing short tails and very long heads with axial ratios of 3.5:1, were new isolates of an extremely rare group. Phages of both groups formed polyheads of various sizes and shapes. The last phage, 61 nm in diameter, seemed to have a tail-like appendage. All phages had double-stranded DNA, were active on enterobacteria only, and differed in their host range. The first five phages seemed to be Salmonella specific.  相似文献

Inhibition of HeLa cell deoxyribonucleic acid (DNA) synthesis, which occurred by the 4th to 5th hr after infection with poliovirus, could be blocked completely by guanidine only when it was present before the 2nd hr. At the 2nd hr, there was no significant ribonucleic acid (RNA)-replicase activity, and addition of guanidine inhibited all production of virus but allowed 57% of maximal DNA inhibition to develop. Maximum DNA inhibition developed in cells infected for 4 hr in the presence of guanidine when the guanidine was removed for a 10-min interval. RNA-replicase activity was not enzymatically detectable and viral multiplication did not develop in these cells unless the interval without guanidine was extended to 60 min. The interpretation of the data was that the effect of guanidine on viral-induced inhibition of DNA synthesis was distinct and not a consequence of the inhibition of RNA-replicase.  相似文献
