Starting from the histone mixture obtained from calf thymus, the arginine rich fraction ARE+) was coupled to organomercurial agarose via a mercaptide bond to one of its cysteines. ARE-agarose proved to be useful for a large scale affinity chromatographic separation of whole histone. In 1M NaCl, pH 4.5, highly pure histone fractions could be eluted with an urea gradient revealing increasing affinity towards ARE in the order: KAP < KAS < LAK < (ARE)n(GRK)n < GRK < ARE.  相似文献

1. The effect of tetanic stimulation of ipsilateral group I afferents from the GS muscle on a synchronous stretch of the flexor EDL/TA muscles has been investigated in precollicular decerebrate cats. 2. The stretch-induced tension of the EDL/TA muscles increases remarkably during simultaneous stimulation of the GS nerve with maximal intensities corresponding to 1.5 times the threshold for the group 1 afferents. This increas appears above all in the dynamic part of stretch. 3. Under our experimental conditions there is no activation fo flexor-alpha-motoneurones during tetanic stimulation of the GS afferents without muscle stretch, as measured by the resting tension of the EDL/TA muscles. 4. Desptie an increase in the stretch-induced tension during fusimotor stimulation of antagonistic group I afferents, a transmission loss in the excitation via the psi-loop to the flexor-alpha-motoneurones occurs. This could be demonstrated by the ratio: increase in the tension T /increase in the number of Ia spikes. This is explained by snychronous convergence of the discharges of Ia inhibitory interneurones to the flexor-alpha-motoneurones. 5. The system studied demonstrates an input-output relation of the stretch reflex during conditions in which both reciprocal inhibition and autogenetic excitation via the psi-loop occur. It appears however, that the reciprocal inhibition is partly overwhelmed by the autogenetic excitation which results from the increase in the Ia discharge rate during fusimotor reflex. 6. It is postulated that static rather than dynamic psi-moto-neurones are involved in the investigated reflex arc.  相似文献

1. We have defined conditions that permit quantitative immunoelectrophoresis in agarose gels of dodecyl sulfate-solubilized erythrocyte membrane proteins. 2. Using human serum albumin, transferrin, MN-glycoprotein (glycophorin) and crude spectrin as test proteins, we found that accurate analyses are possible if samples and gels are 1% in non-ionic detergent (Berol EMU-043) or Triton X-100) and if no more than 100 nmol free dodecyl sulfate is applied per sample. 3. Dodecyl sulfate treated membranes analyzed by crossed immunoelectrophoresis using rabbit antibodies against membrane material yielded optimal precipitation patterns in gels containing 1% of non-ionic detergent. 4. Crossed immunoelectrophoresis in the presence of 1% of Berol revealed precipitates when 10 protein bands defined and isolated by preparative dodecyl sulfate-polyacrylamide gel electrophoresis were run against anti-membrane antibodies. Seven of these bands showed more than one precipitation arc, indicating the presence of more than one antigenic component. 5. Crossed-line immunoelectrophoresis showed that dodecyl sulfate-polyacrylamide gel electrophoresis bands 1, 2 and 2.1 shared common antigenic components. The MN-glycoprotein was present in bands 3, 4A, 4B and 5, where antigenic components of the major intrinsic erythrocyte membrane protein, band 3, were also found. 6. After absorption of the anti-membrane antibody with intact erythrocytes, immunoelectrophoresis showed the disappearance of the MN-glycoprotein precipitates. An increase in the area below the precipitate corresponding to the major intrinsic protein (band 3) was also observed, indicating exposure of some antigens of this protein on the outer surface of intact cells. 7. After absorption of the antibody preparation with washed erythrocyte membranes, immunoprecipitates were not seen in any experiments, indicating that all antigenic determinants observed are exposed at one or both surfaces of the membrane. 8. Our analyses indicate that the peptide moieties of serum lipoproteins do not constitute a significant component of erythrocyte membranes.  相似文献

The changes in absorption spectra in the visible region observed on adding different naturally occurring and synthetic DNA duplexes to solutions of 4,5-dibromo-2,7-di-(acetatomercuri)-fluorescein indicate that the mercurial reacts with polynucleotides of this type. The reaction is reversible as proved by adding excess of KCN which restores the original spectra of the free dye. The interaction is characterised also by quenching of the fluorescence of the dye and the induction of optical activity in it. The extent of these spectral effects depends strongly on the (A+T) content of the complexed DNA and decreased in the order: poly [d(A-T)], Clostridium perfringens DAN, Escherichia coli DNA, Micrococcus luteus DNA and poly(dC). From equilibrium-dialysis experiments the same order in affinity is obtained when these poly-nucleotides are at equilibrium with the same concentration of 4,5-dibromo-2,7-di-(acetatomercuri)-fluorescein. From the changes produced by different mercurials in the ORD spectra and viscosity of a DNA solution it has been concluded that 4,5-dibromo-2,7-di(acetatomercuri)-fluorescein does not cause any drastic alteration of the secondary structure of DNA.  相似文献

1. The angle between the Os sphenoidale and the Clivus has been determined on 106 skulls. The mean was 117.68 degrees. 2. The altitude of the Foramen magnum with respect to the Frankfort plane was determined at -1.96 cm on the average. 3. The angle of inclination of the Foramen magnum to the Frankfort plane was determined from the altitude of the Basion and Opisthion and from the length of the Foramen magnum. The mean was -13.71 degrees. 4. In the inner base of the skull the altitudes of following structures have been determined: Planum sphenoidale (mean 2.34 cm), ground of the Fossa hypophysialis (1.32 cm), ground of the middle cranial fossa (-0.03 cm), ground of the posterior cranial fossa (-2.30 cm), upper edge of the Os petrosum (1.12 cm), Tuberculum jugulare (-0.21 cm), ground of the Meatus acusticus internus (0.45 cm). 5. In order to provide numerical data for the step-like building of the inner base of the skull, the differences between the altitudes of important structures from the inner base have been determined: Margo superior partis petrosae - Tuberculum jugulare (mean 1.35 cm). Opisthion - ground of the posterior cranial fossa (0.11 cm), Planum sphenoidale - ground of the middle cranial fossa (2.38 cm), ground of the middle cranial fossa - ground of the posterior cranial fossa (2.27 cm), Margo superior partis petrosae - ground of the posterior cranial fossa (3.35 cm), Margo superior partis petrosae - ground of the middle cranial fossa (1.15 cm).  相似文献

Circular dichroism spectra for C3a anaphylatoxins (protein fragments generated enzymatically in serum during activation of the third component of complement (C3)) from human and porcine sources were compared in the region of 190 to 250 nm. The spectra were indistinguishable in this region, although an estimated difference of approximately 20% exists between the primary structures of human and porcine C3a. Calculations indicated a total of 40 to 45% alpha helical content based on either the 208 or 222 nm extremum of the CD spectra. Addition of either mercaptoethanol or 6 M guanidinium chloride to human C3a produced a marked decrease in the mean residue ellipticity centered at 222 nm without irreversibly affecting the biological activity. Simultaneous addition of mercaptoethanol and guanidinium chloride virtually eliminated the CD contribution at 222 nm and resulted in more than 90% inactivation of the anaphylatoxin. Removal of the denaturant and reducing agent restored both the biological activity and the CD spectrum of C3a. Heat treatment or reduction and alkylation of human C3a produced biologically inactive and conformationally modified anaphylatoxin. In contrast, C3a inactivated by enzymatic removal of the COOH-terminal arginyl residue was structurally unchanged as judged by CD measurements. Consequently, it is proposed that in addition to the essential COOH-terminal arginyl residue, a highly ordered conformation is required for biological functionality of the C3a molecule.  相似文献

Plants homozygous for an interchange in the long arms of chromosomes 3 and 5 were selected from the progeny of translocated heterozygotes through mitotic chromosome analysis. While a deficit of translocated homozygotes was observed in these segregating families, an increase in the proportion of normal plants was obtained. The translocated homozygotes showed a bushy, dwarfed and weak form coupled with nearly complete sterility. Meiosis turned out to be normal except for the presence of anaphase bridges that occurred in about 10 percent of the pollen mother cells. However, vegetative and floral buds degenerated in the majority of cases before pods developed. In mitosis, anaphase bridges were observed. Their frequency was 30 percent and they were formed by only one of the exchanged chromosomes. Such bridges are considered a consequence of the so-called point errors, which are suggested as the main cause of developmental disturbances.The investigations were supported by the Ministry of Education and Sciences of the Federal Republic of Germany and by the Association Euratom-Ital at Wageningen, The Netherland.  相似文献

The suitability of glycine hydrazide as a link between steroids and carrier proteins in the synthesis of antigens was examined. Testosterone was used as hapten; bovine serum albumin as carrier protein. The reaction described here of testosterone with glycine hydrazide to form testosterone glycylhydrazone acetate took place under milk conditions and the yield was nearly quantitative. Rabbits immunized with the new antigen developed specific antibodies against testosterone.  相似文献
