Embryos of the sea urchin, Arbacia punctulata, were shown to utilize carboxyl labeled acetate and glycine for the synthesis of nucleic acids and protein.The hypothesis that DNA must arise exclusively from the pre-formed RNA of the virgin egg is unlikely since the specific activities of DNA and its isolated purines from the blastula were at least 10 times greater than the corresponding values for RNA.The specific activities of adenine and guanine of RNA formed from glycine-1-C14 were identical, whereas in DNA the specific activity of adenine was less than half that of guanine.Comparison of the specific activities of the DNA purines with the glycine in the medium indicated that a large part of the newly formed DNA arose from unlabeled endogenous precursor.  相似文献

Summary This article investigates the mean abundances of trophic levels in simple models of two- and three-level food chains as a function of the rate of input of nutrients. The analysis concentrates on cases in which the equilibrium point with all species present is unstable. In most of the models, the instability arises because the consumer species become satiated when food density is high. In unstable two-level systems, bottom level abundance generally increases with increased nutrient input. The abundance of the second level may decrease with increased input. Changes in the intrinsic rate of increase and carrying capacity of the bottom level can have qualitatively opposite effects on trophic level abundances. Refuges for or immigration of the bottom level usually cause both levels to increase in mean abundance with an increased carrying capacity. A variety of different predator—prey models are discussed briefly and the results suggest that increased nutrient input will often increase the abundance of both levels; however, several circumstances can cause the top level to decrease. In three-level systems, an increased carrying capacity can cause extinction of the top level. Extinction may or may not be conditional on the initial densities of the three levels. These results may help explain the observed lack of correlation between productivity and the number of trophic levels in natural food webs, as well as the lack of very long food chains. The results suggest that patterns of abundances across productivity gradients cannot be used to assess the importance of top-down vs bottom-up effects.  相似文献

Simian immunodeficiency virus (SIV) was used as a model to study the protective efficacy of an immunization regimen currently being evaluated as candidate vaccines against HIV in human subjects. Four Macaca fascicularis were first immunized with recombinant vaccinia virus expressing the envelope glycoprotein gp160 of SIVmne and then boosted with subunit gp160. Both cell-mediated and humoral immune responses against SIV, including neutralizing antibodies, were elicited. The macaques were shown to be protected from a homologous virus infection as determined by serology, lymphocyte cocultivation, polymerase chain reactions and in vivo transmission analyses. Four unimmunized control animals were readily infected. However, viremia in infected control animals could decrease substantially following the initial phase of infection so that persistent infection might not be readily detectable.  相似文献

We have identified a cDNA for pleckstrin 2 that is 39% identical and 65% homologous to the original pleckstrin. Like the original pleckstrin 1, this protein contains a pleckstrin homology (PH) domain at each end of the molecule as well as a DEP (Dishevelled, Egl-10, and pleckstrin) domain in the intervening sequence. A Northern blot probed with the full-length cDNA reveals that this homolog is ubiquitously expressed and is most abundant in the thymus, large bowel, small bowel, stomach, and prostate. Unlike pleckstrin 1, this newly discovered protein does not contain obvious sites of PKC phosphorylation, and in transfected Cos-7 cells, it is a poor substrate for phosphorylation, even after PMA stimulation. Cells expressing pleckstrin 2 undergo a dramatic shape change associated with actin rearrangement, including a loss of central F-actin and a redistribution of actin toward the cell cortex. Overexpression of pleckstrin 2 causes large lamellipodia and peripheral ruffle formation. A variant of pleckstrin 2 lacking both PH domains still had some membrane binding but did not efficiently induce lamellipodia, suggesting that the PH domains of pleckstrin 2 contribute to lamellipodia formation. This work describes a novel, widely expressed, membrane-associating protein and suggests that pleckstrin 2 may help orchestrate cytoskeletal arrangement.  相似文献
