Hemolysis or extensive cell damage can lead to high concentrations of free heme, causing oxidative stress and inflammation. Considering that heme induces neutrophil chemotaxis, we hypothesize that heme activates a G protein-coupled receptor. Here we show that similar to heme, several heme analogs were able to induce neutrophil migration in vitro and in vivo. Mesoporphyrins, molecules lacking the vinyl groups in their rings, were not chemotactic for neutrophils and selectively inhibited heme-induced migration. Moreover, migration of neutrophils induced by heme was abolished by pretreatment with pertussis toxin, an inhibitor of Galpha inhibitory protein, and with inhibitors of phosphoinositide 3-kinase, phospholipase Cbeta, mitogen-activated protein kinases, or Rho kinase. The induction of reactive oxygen species by heme was dependent of Galpha inhibitory protein and phosphoinositide 3-kinase and partially dependent of phospholipase Cbeta, protein kinase C, mitogen-activated protein kinases, and Rho kinase. Together, our results indicate that heme activates neutrophils through signaling pathways that are characteristic of chemoattractant molecules and suggest that mesoporphyrins might prove valuable in the treatment of the inflammatory consequences of hemorrhagic and hemolytic disorders. 相似文献
Genome integrity and cell proliferation and survival are regulated by an intricate network of pathways that includes cell cycle checkpoints, DNA repair and recombination, and programmed cell death. It makes sense that there should be a coordinated regulation of these different processes, but the components of such mechanisms remain unknown. In this report, we demonstrate that p19INK4d expression enhances cell survival under genotoxic conditions. By using p19INK4d-overexpressing clones, we demonstrated that p19INK4d expression correlates with the cellular resistance to UV treatment with increased DNA repair activity against UV-induced lesions. On the contrary, cells transfected with p19INK4d antisense cDNA show reduced ability to repair DNA damage and increased sensitivity to genotoxic insult when compared with their p19INK4d-overexpressing counterparts. Consistent with these findings, our studies also show that p19INK4d-overexpressing cells present not only a minor accumulation of UV-induced chromosomal aberrations but a lower frequency of spontaneous chromosome abnormalities than p19INK4d-antisense cells. Lastly, we suggest that p19INK4d effects are dissociated from its role as CDK4/6 inhibitor. The results presented herein support a crucial role for p19INK4d in regulating genomic stability and overall cell viability under conditions of genotoxic stress. We propose that p19INK4d would belong to a protein network that would integrate DNA repair, apoptotic and checkpoint mechanisms in order to maintain the genomic integrity. 相似文献
Microbial invasions can compromise ecosystem services and spur dysbiosis and disease in hosts. Nevertheless, the mechanisms determining invasion outcomes often remain unclear. Here, we examine the role of iron-scavenging siderophores in driving invasions of Pseudomonas aeruginosa into resident communities of environmental pseudomonads. Siderophores can be ‘public goods’ by delivering iron to individuals possessing matching receptors; but they can also be ‘public bads’ by withholding iron from competitors lacking these receptors. Accordingly, siderophores should either promote or impede invasion, depending on their effects on invader and resident growth. Using supernatant feeding and invasion assays, we show that invasion success indeed increased when the invader could use its siderophores to inhibit (public bad) rather than stimulate (public good) resident growth. Conversely, invasion success decreased the more the invader was inhibited by the residents’ siderophores. Our findings identify siderophores as a major driver of invasion dynamics in bacterial communities under iron-limited conditions. 相似文献
Industrial Control Systems and Supervisory Control and Data Acquisition (ICS/SCADA) systems are profound backbones of the national critical infrastructures and are essential to the sustainability of society since they help monitoring and controlling the cyber-enable services, such as energy, transportation, healthcare, etc. Modern SCADA systems continue to use the legacy communication protocols that lack adequate security mechanisms to provide trusted device authentication and ensure data flow integrity. Furthermore, advent of state-of-the-art network-capable sensor technology exposes many unique vulnerabilities to the adversaries. Thus, integrity of the data originated from field sensors along with their identity must be managed carefully in order to enhance reliability of ICS/SCADA ecosystems. In this paper, we present a blockchain-based SRAM PUF Authentication and Integrity (BloSPAI) protocol that aims to ensure a continuous authentication of field sensors and provide a robust data flow integrity process by leveraging distributed ledger and hardware security primitives. The prototype of the protocol has been implemented in a sensor-integrated Raspberry PI testbed that is interfaced with a permissioned blockchain network. We discuss the performance and overhead aspects of the proposed BloSPAI protocol and compare with state-of-art cybersecurity solutions. Through experimental evaluation demonstrates the relationship between the size of the blockchain network impacts the throughput in terms of time to commit transactions and overall systems setup time.
Pomacea flagellata is a gastropod conspicuous in freshwater environments, and represents a fishing resource. To assess their abundance, distribution, and secondary production, monthly samplings were carried out in Bacalar Lake from June 2012 to May 2013 at 12 sampling sites. In each site, three random transects were marked parallel to the shore. All snails on transect were collected and shell length and wet weight measured. The highest density occurred in September (1.27 ind.m?2), lowest in October (0.47 ind.m?2). Shell lengths ranged from 2 to 56 mm, with recruitment in January–March. Growth parameters were L∞ 59.50 mm, K 0.65.year?1; the lifetime span was 3 years. Average biomass reached 5.57 wet g.m?2 and secondary production was 6.025 wet g.m?2.year-1; annual renewal rate P/B 1.08. Highest abundance and secondary production was contributed by individuals between 31 and 41 mm in length. A potential biomass of 25.06 tons of snails was estimated in the lake. Snail densities, secondary production, and turnover were very low during the year, indicating that it is not viable to consider a commercial catch without affecting the population. A ban of 10 years is proposed, and aquaculture practices of snails are recommended to recover the resource. 相似文献
