Human B-cell glucokinase displays sigmoidal kinetics towards D-glucose or D-mannose, but fails to do so towards D-fructose. Yet, D-glucose, D-mannose and 2-deoxy-D-glucose confer to the enzyme positive cooperativity towards D-fructose. For instance, in the presence of 5 mM D-[U-14C]fructose, its rate of phosphorylation is increased to 214.3 ± 11.0%, 134.0 ± 4.3% and 116.5 ± 3.0% of paired control value by D-glucose, D-mannose and 2-deoxy-D-glucose (each 6 mM), respectively. D-glucose and, to a lesser extent, D-mannose also display reciprocal kinetic cooperativity. D-fructose, however, fails to affect D-glucose or D-mannose phosphorylation under conditions in which positive cooperativity is otherwise observed. These findings are relevant to the reciprocal effects of distinct hexoses upon their phosphorylation by B-cell glucokinase and, as such, to the metabolic and functional response evoked in pancreatic islet B-cells by these sugars, when tested either separately or in combination. The hexavalent chromium Cr(VI) poses a threat as a hazardous metal and its removal from aquatic environments through biosorption has gained attention as a viable technology of bioremediation. We evaluated the potential use of three green algae (Cladophora glomerata, Enteromorpha intestinalis and Microspora amoena) dry biomass as a biosorbent to remove Cr(VI) from aqueous solutions. The adsorption capacity of the biomass was determined using batch experiments. The adsorption capacity appeared to depend on the pH. The optimum pH with the acid-treated biomass for Cr(VI) biosorption was found to be 2.0 at a constant temperature, 45?°C. Among the three genera studied, C. glomerata recorded a maximum of 66.6% removal from the batch process using 1.0?g dried algal cells/100?ml aqueous solution containing an initial concentration of 20?mg/L chromium at 45?°C and pH 2.0 for 60?min of contact time. Langmuir and Freundlich isotherm equations fitted to the equilibrium data, Freundlich was the better model. Our study showed that C. glomerata dry biomass is a suitable candidate to remove Cr(VI) from aqueous solutions.  相似文献

GDSL-type esterase/lipase (GELP) is mainly characterized by a conserved GDSL domain at N terminus, and is widely found in all living species, both prokaryotes and eukaryotes. GELP gene family consists of a wide range of members playing important roles in plant physiological processes, such as development, stress responses, and functional divergences. In our study, 597 GELP genes were identified from six Rosaceae genomes (i.e., Fragaria vesca, Prunus persica, Prunus avium, Prunus mume, Pyrus bretschneideri, and Malus domestica) by a comprehensive analysis. All GELP genes were further divided into ten subfamilies based on phylogenetic tree analysis. Subfamily D and subfamily E are the two largest subfamilies. Microcollinearity analysis suggested that WGD/segmental events contribute to the expansion of the GELP gene family in M. domestica and P. bretschneideri compared to F. vesca, P. persica, P. avium, and P. mume. Some PbGELPs were expressed during the fruit development of P. bretschneideri and pollen tubes, indicating their activity in these tissues. The expression divergence of PbGELP duplication gene pairs suggests that many mutations were allowed during evolution, although the structure of GELP genes was highly conserved. The current study results provided the feasibility to understand the expansion and evolution patterns of GELP in Rosaceae genomes, and highlight the function during P. bretschneideri fruits and pollen tubes development.  相似文献

Mushrooms have been highly regarded as possessing enormous nutritive and medicinal values. In the present study, we evaluated the anti-oxidative and anti-atherosclerotic potential of shiitake mushroom (Lentinula edodes) using its solvent–solvent partitioned fractions that consisted of methanol:dichloromethane (M:DCM), hexane (HEX), dichloromethane (DCM), ethyl acetate (EA) and aqueous residue (AQ). The hexane fraction (1 mg/mL) mostly scavenged (67.38%, IC50 0.55 mg/mL) the 2,2-diphenyl-1-picryl hydrazyl (DPPH) free radical, contained the highest reducing capacity (60.16 mg gallic acid equivalents/g fraction), and most potently inhibited lipid peroxidation (67.07%), low density lipo-protein oxidation and the activity of 3-hydroxy 3-methyl glutaryl co-enzyme A reductase (HMGR). GC–MS analyses of the hexane fraction identified α-tocopherol (vitamin E), oleic acid, linoleic acid, ergosterol and butyric acid as the bio-functional components present in L. edodes. Our findings suggest that L. edodes possesses anti-atherosclerotic bio-functionality that can be applied as functional food-based therapeutics against cardiovascular diseases.  相似文献

The intention of the present wok is to provide an account on the pollen morphological features of Lamiaceae in Saudi Arabia as a basis for future studies of Lamiaceae pollens in the region. Pollen morphology of 20 species belong to 16 genera of the Lamiaceae has been investigated using Light Microscope (LM) and Scanning Electron Microscope (SEM). The study revealed that the pollen grains were characterized by 3-zonocolpate or 6-zonocolpate. Size of the pollen is variable between the genera but not among the species of the same genus. The surface pattern of the exine varies from fine reticulate, rough reticulate, mega-reticulate, reticulate-perforate, bireticulate-perforate or granulate, leading to 6 types of pollen grains. These variations revealed by this study implies that pollen Morphology may be of significant value sharing in solving problems in the classification of Lamiaceae members. A Key to the species, based on the morphological features of pollen grains, is also provided.  相似文献

In this study, the removal of arsenic (As) by plant, Ludwigia octovalvis, in a pilot reed bed was optimized. A Box-Behnken design was employed including a comparative analysis of both Response Surface Methodology (RSM) and an Artificial Neural Network (ANN) for the prediction of maximum arsenic removal. The predicted optimum condition using the desirability function of both models was 39 mg kg?1 for the arsenic concentration in soil, an elapsed time of 42 days (the sampling day) and an aeration rate of 0.22 L/min, with the predicted values of arsenic removal by RSM and ANN being 72.6% and 71.4%, respectively. The validation of the predicted optimum point showed an actual arsenic removal of 70.6%. This was achieved with the deviation between the validation value and the predicted values being within 3.49% (RSM) and 1.87% (ANN). The performance evaluation of the RSM and ANN models showed that ANN performs better than RSM with a higher R2 (0.97) close to 1.0 and very small Average Absolute Deviation (AAD) (0.02) and Root Mean Square Error (RMSE) (0.004) values close to zero. Both models were appropriate for the optimization of arsenic removal with ANN demonstrating significantly higher predictive and fitting ability than RSM.  相似文献
