Background aimsThe aim was to investigate the therapeutic effect of granulocyte–colony-stimulating factor (G-CSF) administration following implantation of autologous bone marrow mononuclear cells (BM MNC) for patients with lower limb ischemia.MethodsThe design was a randomized controlled trial. Fifteen patients with severe chronic limb ischemia were treated with autologous BM MNC [without G-CSF (MNC–G-CSF) or combined with G-CSF administration for 5 days following transplantation (MNC+G-CSF)].ResultsAll clinical parameters, including ankle brachial index, visual analog scale and pain-free walking distance, showed a mean improvement from baseline, which was measured at 4 and 24 weeks after transplantation in both groups. However, in three (20%) patients, the clinical course did not improve and limb salvage was not achieved. No significant difference was observed among the patients treated in the MNC–G-CSF and MNC+G-CSF groups. No severe adverse reactions were reported during the study period. No relationship was observed between both the numbers of viable MNC or CD34+ cells and the clinical outcome.ConclusionsAutologous transplantation of BM MNC into ischemic lower limbs is safe, feasible and efficient for patients with severe peripheral artery disease. However, the administration of G-CSF following cell transplantation does not improve the effect of BM MNC implantation and therefore would not have any beneficial value in clinical applications of such cases.  相似文献

A renewable three-dimensional chemically modified carbon ceramic electrode (CCE) containing nickel powder and K4[Mo(CN)8] was constructed by sol-gel technique. The electrochemical properties and stability of modified electrode was evaluated by cyclic voltammetry in pH range 4-10. The redox couple of [Mo(CN)8] (4-/3-) was shown both as a solute in electrolyte solution and as a component of a carbon based conducting composite electrode. The apparent electron transfer rate constant (ks) and transfer coefficient (alpha) were determined by cyclic voltammetry and they were about 17.1 and 0.57 s(-1), respectively. The catalytic activity of the modified CCE toward insulin oxidation was investigated at pH range of 3-8 by cyclic votammetry. The modified electrode showed excellent electrocatalytic activity toward insulin electroxidation at physiological pH value. The modified electrode was used for insulin detection chronoamperometrically at pH 7. Under optimized condition in amperometry method, the concentration calibration range, detection limit and sensitivity were 0.5-500 nM, 0.45 nM and 6140 nA/microM, respectively. Flow injection amperometric determination of insulin at pH 7.4, at this modified electrode yielded a calibration curve with the following characteristics, linear dynamic range 100-500 pM; sensitivity 8.1 nA/nM and detection limit 40 pM (based on S/N = 3). The inherent stability at wide pH range, high sensitivity, low detection limit, low cost and ease of preparation are of advantageous of this insulin sensor. This sensor indicates great promise for monitory insulin in chromatographic effluents.  相似文献

Considered to be the "holy grail" of dentistry, regeneration of the periodontal ligament in humans remains a major clinical problem. Removal of bacterial biofilms is commonly achieved using EDTA gels or lasers. One side effect of these treatment regimens is the etching of nanotopographies on the surface of the tooth. However, the response of periodontal ligament fibroblasts to such features has received very little attention. Using laser interference lithography, we fabricated precisely defined topographies with continuous or discontinuous nanogrooves to assess the adhesion, spreading and migration of PDL fibroblasts. PDL fibroblasts adhered to and spread on all tested surfaces, with initial spreading and focal adhesion formation slower on discontinuous nanogrooves. Cells had a significantly smaller planar area on both continuous and discontinuous nanogrooves in comparison with cells on non-patterned controls. At 24 h post seeding, cells on both types of nanogrooves were highly elongated parallel to the groove long axis. Time-lapse video microscopy revealed that PDL fibroblast movement was guided on both types of grooves, but migration velocity was not significantly different from cells cultured on non-patterned controls. Analysis of filopodia formation using time-lapse video microscopy and labeling of vinculin and F-actin revealed that on nanogrooves, filopodia were highly aligned at both ends of the cell, but with increasing time filopodia and membrane protrusions developed at the side of the cell perpendicular to the cell long axis. We conclude that periodontal ligament fibroblasts are sensitive to nanotopographical depths of 85-100 μm, which could be utilized in regeneration of the periodontal ligament.  相似文献

The vibration characteristics of shell structures such as eyes have been shown to vary with intraocular pressure (IOP). Therefore, vibration characteristics of the eye have the potential to provide improved correlation to IOP over traditional IOP measurements. As background to examine an improved IOP correlation, this paper develops a finite element model of an eye subject to vibration. The eye is modeled as a shell structure filled with inviscid pressurized fluid in which there is no mean flow. This model solves a problem of a fluid with coupled structural interactions of a generally spherically shaped shell system. The model is verified by comparing its vibrational characteristics with an experimental modal analysis of an elastic spherical shell filled with water. The structural dynamic effects due to change in pressure of the fluid are examined. It is shown that the frequency response of this fluid-solid coupled system has a clear increase in natural frequency as the fluid pressure rises. The fluid and structure interaction is important for accurate prediction of system dynamics. This model is then extended to improve its accuracy in modeling the eye by including the effect of the lens to study corneal vibration. The effect of biomechanical parameters such as the thicknesses of different parts of the eye and eye dimensions in altering measured natural frequencies is investigated and compared to the influence of biomechanical parameters in Goldmann applanation tonometry models. The dynamic response of the eye is found to be less sensitive to biomechanical parameters than the applanation tonometry model.  相似文献

Bakhtiarian savory (Satureja bachtiarica Bunge) is an endemic herb growing wild in Iran with interesting pharmacological and biological properties. In this research, the variability of essential oil content and phytochemical compositions among seven populations of this species was studied in both natural and field conditions. There was a wide interpopulation variability in phytochemical variation of studied populations under both natural and field conditions, indicating the existence of different chemotypes. Six chemical compositions showed high value in studied plants so that carvacrol and thymol were known as major compounds and confirmed by principal component analysis. Ilam population had the highest thymol in both field (77.10 %) and natural (42.32 %) conditions and was known as a thymol chemotype. Beside, Yazd and Shahrekord populations, respectively, showed the highest carvacrol percentages in both field (84.83 and 63.81 %) and natural (72.50 and 58.05 %) conditions and were known as carvacrol chemotypes. In other populations, carvacrol and thymol were the main components and showed partly equal percentage and these populations were known as carvacrol/thymol chemotypes. According to Pearson correlation analysis, carvacrol was negatively correlated with p-cymene, γ-terpinene and thymol (r = ?0.61, ?0.70 and ?0.98, respectively), indicating very high reverse correlation between carvacrol and thymol. In conclusion, the high value of the essential oil compositions in studied S. bachtiarica populations could provide useful information for conservation and selection of cross parents in breeding programs to develop improved cultivars.  相似文献

Salicylic acid (SA) may reduce the negative impact of water deficit on growth and metabolite yield of Thymus daenensis Celak subsp. daenensis Celak. The effect of foliar application of SA and reduced irrigation on growth, oil yield, chemical components, and antibacterial and antioxidant activities of T. daenensis in field condition were investigated. Treatments comprised 0.0, 1.5 and 3.0 M SA applied to plants under normal irrigation and stressed conditions. Results indicated that irrigation regime had a significant effect on growing degree days (GDD) required to reach early and full flowering. Foliar application of SA influenced GDD from early growing stage to 50 % and full flowering, minimum radius and canopy diameter. The highest values of oil content (3.2 % v/w) and yield (14.9 g m?2) were obtained from application of 3.0 M SA. Percentage of some chemical constituents in the essential oil extracted from the plants under stress was higher than non-stressed plants. Thymol content was significantly reduced under stressed conditions. Foliar application of SA significantly improved carvacrol, α-thujene, α-pinene and p-cymene contents in the oils, but reduced thymol and, β-caryophyllene amounts. Our results showed that foliar application of SA reduced the negative effect of water deficit on thymol content in the essential oil of T. daenensis. The essential oils of T. daenensis exhibited antioxidant and antibacterial activities when plants were sprayed with 1.5 and 3.0 M SA, respectively.  相似文献

Key message

This work is an important step in the conservation of genetic cherry resources, which showed distinctive and interesting agronomical characters. Also it introduces suitable genotypes for cultivation and breeding studies.

Abstract

The purpose of this study was to characterize cherry germplasm that is cultivated in Iran. Thirty-three morphopomological parameters were studied in this germplasm, consisting of 70 cherry genotypes (41 sweet cherry, 24 sour cherry and 5 duke cherry genotypes). A wide variation was found in blooming time, ripening time, fruit weight, fruit color, anthocyanin, total soluble solids (TSS), titratable acidity (TA), fruit dimensions and flesh firmness and stone size. There were close positive correlations between fruit weight and fruit dimensions, and between fruit weight and fruit stalk weight, fruit flesh firmness and cracking and also a negative correlation between pH and TA. Dendrogram gave a clear separation between the sour, duke and sweet cherry species and also showed existing intraspecific morphological variation. Based on fruit size and organoleptic properties, the sweet cherry genotypes ‘Siah-Mashhad’, ‘Takdaneh-Mashhad’, ‘Shabestar’, ‘Siah-Daneshkade’, ‘Ghazvin’ and ‘Droongezna’ are recommended for fresh consumption. Good fruit chemical composition and late-ripening time stands out genotypes ‘Dirres-Italia’, ‘Dirres-Pardis’, ‘Maremoot’, ‘Abardeh’ and ‘Rorshon’ and make them suitable for processing. Also, ‘Gilas46’ and ‘Gilas49’ were substantially late-ripening, a characteristic that makes these genotypes highly suitable for breeding studies in case of ripening time. Furthermore, sour cherries ‘Hashtgerd2’ and ‘Hashtgerd3’ and duke cherries ‘Pardis1’ and ‘Pardis3’ were the best genotypes. This work is an important step in the conservation of genetic cherry resources in Iran, which showed distinctive and interesting agronomical characters such as low susceptibility to fruit cracking, high levels of total soluble solids, early fruit maturity and high fruit quality.  相似文献

Thymus caramanicus is an endemic species grown in Iran with interesting pharmacological and biological properties. In the present work, essential oil compositions and inter-simple sequences repeat (ISSR) markers were used to estimate the relationships among and within seven populations of T. caramanicus, belonging to three provinces in Iran. The studied individuals were distinguished on the basis of ISSR markers and constituents of essential oil. A total of 127 band positions were produced by 12 ISSR primers, of which 105 were found polymorphic with 82.68 % polymorphism. Genetic similarity values among individuals ranged between 0.15 and 0.82 which was indicative of a high level of genetic variation. On the basis of their genetic similarities, ISSR analysis allowed to group the samples into two main clusters. One of these included populations originated from Kerman and Isfahan provinces, and the other cluster consists of populations from Semnan province. Chemical compounds of essential oils were found variable in the various individuals and all samples were principally composed of phenolic constituents (carvacrol and/or thymol). As a consequence, the plants were classified into two major chemotypes including carvacrol and thymol/carvacrol. A relationship between genetic and chemical variability and geographic distribution has been observed in studied populations of T. caramanicus.  相似文献

The morphological variation and essential oil content of 150 individuals of motherwort (Leonurus cardiaca) from six natural regions of Iran were examined biometrically based on multivariate analysis. The highest variation was observed in plant height, length of main stems, main inflorescence length, floral cycles of main stem, lateral branches length, basal leaf length, floral leaf length width, flower length, and calyx color. Results of simple correlation analysis showed the existence of significant positive and negative correlations among some important parameters. The highest correlation was observed between plant height and length of main stems and between lateral branch length and lower surface color of leaf. Populations were clustered in two groups representing subspecies cardiaca and persicus. The subspecies cardiaca populations were closely related with each other and differentiated from the subspecies persicus by pubescence stems and leaves, more number of floral cycles, higher compaction of floral cycles, lower number of lateral branches, shorter length of petioles and leaves and smaller floral leaves. Essential oil yields varied from 0.02 (for Taleghan population) to 0.053 mg/100 g dry mater (for Kerman population). The conservation of the highly diverse native populations of Iranian motherwort germplasm is recommended.  相似文献
