To determine differences in microbial community structure, phospholipid fatty acids (PL-FA) from rhizosphere bacteria of two different wheat cultivars Triticum aestivum L. (cv. Bohouth-6 and cv. Salamouni) were extracted and analyzed by gas chromatography. This approach was used to overcome the methodological underestimation of microbial densities obtained with isolation, culture techniques and microscopic observations. Our objective was to verify differences in PL-FA profiles from two wheat cultivars grown under controlled environmental conditions. Principal component analysis (PCA) and cluster analysis were used to detect dissimilarities between rhizosphere microbial communities of the two wheat cultivars and signature fatty acids (FA) were used to determine specific differences in the community structures. PCA of the two cultivars explained 79.18% of the variance on principal component 1 (PC1), which accounted for Bohouth-6 rhizosphere soil. The rhizosphere soil of Salamouni accounted for 11.66% of the variance on principal component 2 (PC2). The results demonstrated repeatedly the clustering of the samples into two distinct groups; each group belonging specifically to one of the two wheat cultivars. Profiles of Bohouth-6 showed higher amounts of cyclopropane acid 19:0cy and Sif 7 (Sum in feature 7) than Salamouni. Those FA are known as signature molecules for Gram-negative bacteria. This was also reflected by the higher bacterial counts (cfu g–1 fresh root weight) of Gram-negative bacteria from the rhizosphere of the former than the latter. The results indicated that under controlled environmental conditions, wheat cultivars of different genotypes exhibit distinct microbial colonization in their rhizosphere.  相似文献

We present data that summarize our findings on the role of taurine in the central nervous system and in particular taurine's interaction with the inhibitory and excitatory systems. In taurine-fed mice, the expression level of glutamic acid decarboxylase (GAD), the enzyme responsible for GABA synthesis, is elevated. Increased expression of GAD was accompanied by increased levels of GABA. We also found in vitro, that taurine regulates neuronal calcium homeostasis and calcium-dependent processes, such as protein kinase C (PKC) activity. This calcium-dependent kinase was regulated by taurine, whereas the activity of protein kinase A (PKA), a cAMP-dependent, calcium-independent kinase, was not affected. Furthermore, as a consequence of calcium regulation, taurine counteracted glutamate-induced mitochondrial damage and cell death.  相似文献

For butterflies, tolerance to the matrix may be an important criterion of habitat occurrence in fragmented landscapes. Here we examine the relative effects of habitat fragmentation and the surrounding agricultural matrix on the functional composition of fruit-feeding butterflies of the Atlantic rain forest in southeastern Brazil. Generalized linear models were used to detect the effects of landscape metrics on butterfly richness and abundance of the total assemblage and functional groups. Circular statistics were used to analyze the patterns of monthly abundance of the total assemblage and functional groups in the forest remnants and the surrounding matrices. In total, 650 butterflies representing 57 species were captured; species composition differed significantly between the forest fragments and the surrounding matrices. We recorded 22 forest specialists, 18 matrix specialists, 11 common species with matrix preference and six common species with forest preference. Forest connectivity favored the richness of forest specialists, while habitat fragmentation enhances the richness and abundance of matrix-tolerant species. Circular analysis revealed that forest specialists were more abundant in the rainy season while matrix-tolerant species proliferated in the dry season. Although maintaining connectivity of forest fragments may increase the mobility and dispersion of forest species, our results showed that landscape fragmentation modify butterfly assemblage by promoting an increase of matrix tolerant species with detriment of forest specialists.  相似文献

Autism is a severe neurodevelopmental disorder, which typically emerges in early childhood. We have used real-time PCR to investigate changes in expression levels of three genes: WNT2, MECP2, and FMR1 in different brain regions of Fagile X mice and litter mate controls. We found major changes in the expression pattern for the three genes examined. FMR1, MECP2, and WNT2 expression were drastically down regulated in the Fragile X mouse brain.  相似文献

Knowledge-based mean force potentials are certainly the most popular type of interaction function for protein threading. They are derived from statistical analyses of interacting groups in experimentally determined protein structures. These potentials are developed at the atom or the amino acid level. While numerous studies have examined the effects of increased primary production on higher trophic levels, most studies have focused primarily on the grazing food web and have not considered the importance of alternate prey channels. This has happened despite the fact that fertilization not only increases grazing herbivore abundance, but other types of consumers such as detritivores that serve as alternate prey for generalist predators. Alternate prey channels can sustain generalist predators at times when prey abundance in the grazing food web is low, thus increasing predator densities and the potential for trophic cascades. Using arthropod data from a fertilization experiment, we constructed a hierarchical Bayesian model to examine the direct and indirect effects of plant production and alternate prey channels on predators in a salt marsh. We found that increased plant production positively affected the density of top predators via effects on lower trophic level herbivores and mesopredators. Additionally, while the abundance of algivores and detritivores positively affected mesopredators and top predators, respectively, the effects of alternate prey were relatively weak. Because previous studies in the same system have found that mesopredators and top predators rely on alternate prey such as algivores and detritivores, future studies should examine whether fertilization shifts patterns of prey use by predators from alternate channels to the grazing channel. Finally, the hierarchical Bayesian model used in this study provided a useful method for exploring trophic relationships in the salt marsh food web, especially where causal relationships among trophic groups were unknown.  相似文献

A total of 250 endophytic fungal isolates, representing 30 morphotaxa, were isolated and characterised, they were collected from the different living symptomless parts of date palm trees of orchards of six Egyptian governorates. Colonisation was greater in samples from the midrib than in those from laminar tissue and slightly greater at the tip of the lamina compared with the base of the leaf. Acremonium spp. were frequently isolated as date palm root endophytes. Acremonium isolates were screened in Petri dishes to select the highest antagonistic one against an Algerian isolate of Fusarium oxysporum f.sp. albedinis. Two-week-old axenically reared date palm seedlings grown in Petri dishes were directly injected with spore suspension (1.5?×?107 spores/ml) of a pure culture of the virulent antagonistic isolate of Acremonium sp. One week after endophytic colonisation, date palm seedlings were then challenged with the pathogen, Fusarium albedinis. The challenged seedlings exhibited a significant reduction in wilt symptom percentage (by 87.0%), while the seedlings exposed to Fusarial toxin without pathogen exhibited the wilt disease symptoms. This indicates that the endophyte ably depresses any toxic action of F. albedinis. The endophytic fungus was recovered from sites distant from the point of inoculation after six?months from the application, indicating that the Acremonium sp. has the potential to move throughout the tissue plant, even the end time of trial. The Acremonium mode of action, as a biocontrol agent, was discussed.  相似文献

Thalassemia intermedia is a clinical definition applied to patients whose clinical phenotype is milder than thalassemia major. To characterize different common mechanisms involving in pathogenesis of moderate to severe β-thalassemia intermedia, we have studied four factors in 38 Iranian patients with thalassemia intermedia: β-globin gene mutation, deletion in α-globin genes, presence of XmnI polymprphism and RFLP haplotype at β-globin gene cluster. The results showed that 84.4% of patients were associated with severe mutations in β-globin gene, mainly IVSII-1(G to A) (56.4%). The positive XmnI polymorphism was seen in 76.9% of the studied alleles which showed strong linkage to β° mutations and high level of fetal hemoglobin. Co-existence of α-globin gene deletions, β+ mutation and the most frequent of RFLP haplotype (−/−, +/+, −/+, +/+, +/+, +/+, −/−) were seen in 7.7, 12.8 and 17.9%, respectively. In this group of our study it seems the main ameliorating factor in the patients was co-inheritance of a positive XmnI polymorphism with β° mutation especially IVSII-1, which were associated with increased production of fetal hemoglobin. However, the other probable genetic factors should be investigated to describe genotype-phenotype correlation in thalassemia intermedia patients.  相似文献
