Cry1Ab is one of the most studied insecticidal proteins produced by Bacillus thuringiensis during sporulation. Structurally, this protoxin has been divided in two domains: the N-terminal toxin core and the C-terminal portion. Although many studies have addressed the biochemical characteristics of the active toxin that corresponds to the N-terminal portion, there are just few reports studying the importance of the C-terminal part of the protoxin. Herein, we show that Cry1Ab protoxin has a unique natural cryptic endotoxic property that is evident when their halves are expressed individually. This toxic effect of the separate protoxin domains was found against its original host B. thuringiensis, as well as to two other bacteria, Escherichia coli and Agrobacterium tumefaciens. Interestingly, either the fusion of the C-terminal portion with the insecticidal domain-III or the whole N-terminal region reduced or neutralized such a toxic effect, while a non-Cry1A peptide such as maltose binding protein did not neutralize the toxic effect. Furthermore, the C-terminal domain, in addition to being essential for crystal formation and solubility, plays a crucial role in neutralizing the toxicity caused by a separate expression of the insecticidal domain much like a dot/anti-dot system.  相似文献

Human placental stem villi (PSV) present contractile properties. In vitro mechanics were investigated in 40 human PSV. Contraction of PSV was induced by both KCl exposure (n = 20) and electrical tetanic stimulation (n = 20). Isotonic contractions were registered at several load levels ranging from zero-load up to isometric load. The tension-velocity relationship was found to be hyperbolic. This made it possible to apply the A. Huxley formalism for determining the rate constants for myosin cross-bridge (CB) attachment and detachment, CB single force, catalytic constant, myosin content, and maximum myosin ATPase activity. These molecular characteristics of myosin CBs did not differ under either KCl exposure or tetanus. A comparative approach was established from studies previously published in the literature and driven by mean of a similar method. As compared to that described in mammalian striated muscles, we showed that in human PSV, myosin CB rate constants for attachment and detachment were about 103 times lower whereas myosin ATPase activity was 105 times lower. Up to now, CB kinetics of contractile cells arranged along the long axis of the placental sheath appeared to be the slowest ever observed in any mammalian contractile tissue.  相似文献

Attribute-based encryption (ABE) is an access control mechanism that ensures efficient data sharing among dynamic groups of users by setting up access structures indicating who can access what. However, ABE suffers from expensive computation and privacy issues in resource-constrained environments such as IoT devices. In this paper, we present SHARE-ABE, a novel collaborative approach for preserving privacy that is built on top of Ciphertext-Policy Attribute-Based Encryption (CP-ABE). Our approach uses Fog computing to outsource the most laborious decryption operations to Fog nodes. The latter collaborate to partially decrypt the data using an original and efficient chained architecture. Additionally, our approach preserves the privacy of the access policy by introducing false attributes. Furthermore, we introduce a new construction of a collaboration attribute that allows users within the same group to combine their attributes while satisfying the access policy. Experiments and analyses of the security properties demonstrate that the proposed scheme is secure and efficient especially for resource-constrained IoT devices.
