A high-performance liquid chromatographic method for the determination of trimetazidine dihydrochloride (TMZ) in spiked human plasma is described. The method is based on the pre-column derivatization with 9-fluorenylmethyl chloroformate (FMOC-Cl) using the fluorimetric detection technique. Fluoxetine HCl (FLX) was used as internal standard. Both, TMZ and FLX were completely derivatized after heating at 50 degrees C for 20 min in borate buffer pH 8.0. Samples were analyzed by high performance liquid chromatography (HPLC) using Zorbax-TMS column (250 mm x 4.6 mm, i.d., 5 microm) and mobile phase consist of acetonitrile, methanol and 20 mM sodium acetate pH 4.7 (44:6:50; v/v/v). Fluorescence detector (FLD) was adjusted at excitation and emission wavelengths; 265 and 311 nm, respectively. The linearity of the method was in the range of 4.5-200 ng/ml. Limits of detection (LOD) and quantification (LOQ) were 1.5 and 4.5 ng/ml, respectively. Trimetazidine recovery was 96.5+/-1.3% (n=6; RSD=2.1%).  相似文献

This study aims at determining relationships between water quality parameters and radiance data from Landsat-5 Thematic Mapper (TM). Fortunately, water samples and the total biomass of aquatic plants were collected at the same time of acquired satellite image (September 1995). TM bands 1, 2, 3, 4, 5 and 7 and band ratios 3/1, 2/1, 2/4, and 3/4 were compared with in situ measurements and laboratory analysis of water samples. The water quality of interest included K+, Na+, total phosphorus (TP), total nitrogen (TN), dissolved oxygen (DO), pH, Salinity, depth, Secchi disc, and biomass of the aquatic plants (submerged, floating, and emergent). Correlation and regression models were developed between each of the water quality parameter measurements and the radiance of image data. Some water quality parameters were significantly correlated with TM radiance data. Subsequently, the regression models were used to prepare digital cartographic products depicting the water quality over the entire study area. This study constitutes the first effort to use Landsat TM data for mapping water quality parameters in all over the Egyptian lagoons.  相似文献

A sensitive high-performance liquid chromatographic method for determination of ranitidine (RAN) in rabbit plasma is described. The method is based on liquid-liquid extraction, labeling with dansyl chloride and monitoring with fluorescence detector at 338nm (ex)/523nm (em). Plasma samples were extracted with diethyl ether alkalinized with 1M sodium hydroxide. Ephedrine HCl (EPH-HCl) was used as internal standard. Both, RAN and EPH were completely derivatized after heating at 60 degrees C for 10min in sodium bicarbonate solution (pH 9.5). The derivatized samples were analyzed by HPLC using Agilent Zorbax Extended C18 column (150mmx4.6mm i.d.) and mobile phase consists of 48% acetonitrile and 52% sodium acetate solution (0.02M, pH 4.6). The linearity of the method was in the range of 0.025-10microg/ml. The limits of detection (LOD) and quantification (LOQ) were 7.5+/-0.18 and 22.5+/-0.12ng/ml, respectively. Ranitidine recovery was 97.5+/-1.1% (n=6; R.S.D.=1.8%). The method was applied on plasma collected from rabbits at different time intervals after oral administration of 5mg/kg ranitidine HCl.  相似文献

A stereoselective high‐performance liquid chromatographic (HPLC) method was developed and validated to determine S‐(?)‐ and R‐(+)‐propranolol in rat serum. Enantiomeric resolution was achieved on cellulose tris(3,5‐dimethylphenylcarbamate) immobilized onto spherical porous silica chiral stationary phase (CSP) known as Chiralpak IB. A simple analytical method was validated using a mobile phase consisted of n‐hexane‐ethanol‐triethylamine (95:5:0.4%, v/v/v) at a flow rate of 0.6 mL min‐1 and fluorescence detection set at excitation/emission wavelengths 290/375 nm. The calibration curves were linear over the range of 10–400 ng mL‐1 (R = 0.999) for each enantiomer with a detection limit of 3 ng mL‐1. The proposed method was validated in compliance with ICH guidelines in terms of linearity, accuracy, precision, limits of detection and quantitation, and other aspects of analytical validation. Actual quantification could be made for propranolol isomers in serum obtained from rats that had been intraperitoneally (i.p.) administered a single dose of the drug. The proposed method established in this study is simple and sensitive enough to be adopted in the fields of clinical and forensic toxicology. Molecular modeling studies including energy minimization and docking studies were first performed to illustrate the mechanism by which the active enantiomer binds to the β‐adrenergic receptor and second to find a suitable interpretation of how both enantiomers are interacting with cellulose tris(3,5‐dimethylphenylcarbamate) CSP during the process of resolution. The latter interaction was demonstrated by calculating the binding affinities and interaction distances between propranolol enantiomers and chiral selector. Lake Bardawil is the only oligotrophic hypersaline lagoon along theMediterranean coast of Egypt. Its ecological significance is increasing due tothe progressive degradation of comparable wetlands in the region. The aim ofthis study is to analyse the structure and life forms of the vegetation alongthe lake before the execution of the North Sinai Agricultural DevelopmentProject (NSADP) which will threaten the ecosystem of the lake. A data set of150 stands was analysed using multivariate procedures(TWINSPAN, DCA and CCA), to classify the lake's vegetation, and to determinetherelationship between the plant community structure and the environmentalfactors. The classification and ordination resulted in a clear demonstration ofnine vegetation groups associated with four habitat types: submerged seagrasses, salt marshes and sabkhahs, eastern and middle calcareous dunes, andwestern non-calcareous dunes. The first axis of the CCA-ordination separatesthe salt marshes andsabkhahs species from those of the sand dunes along the soil salinity, watertable depth, cations, and pH gradients. CaCO3 and soil textureshowedhighly significant correlation with the second axis of CCA which was animportant predictor for the psammophytic species distribution. The life formsranged from hydrophytes (sea grasses) to phanerophyteswith the dominance of therophytes and chamaephytes. Geophytes and chamaephytesdominate the saline habitats, while therophytes and hemicryptophytes dominatedthe sandy dunes. Eventhough the eastern section of the lake (Lake Zaraniq) wasdeclared as a RAMSAR site, Lake Bardawil needs urgent management to prevent itspollution by the new land use system.  相似文献

Moringa peregrina (Moringaceae), an economically important tree that occurs in arid desert mountains, is currently threatened in large part of its distribution range in the Middle East and the Arabian Peninsula. We assessed the impact of topographic factors on population structure and performance of the species at its northwest range edge on the Hafit Mountain, UAE, where it faces harsh climatic conditions and human disturbances. Size structure, fruit production, and vitality were assessed on all 165 individual plants encountered in 12 gorges. The size distribution indicates that the population of M. peregrina is young; the proportion of small and medium-size individuals is high, but with few recorded juveniles. The frequency of plant occurrence and their performance were highest at middle altitudes. Large trees produced significantly more fruits than small and medium plants, with the larger trees of the population at middle altitude producing most fruits. M. peregrina facilitated the growth of grasses and small shrubs, but limited the presence of other big shrubs and trees beneath their canopies, compared to areas beyond. The study indicates that gorges play an important role in maintaining the M. peregrina population at its hyper-arid northwest range edge.  相似文献

Of the approximately 34 identified Biomphalaria species,Biomphalaria alexandrina represents the intermediate host of Schistosoma mansoni in Egypt. Using parasitological and SOD1 enzyme assay, this study aimed to elucidate the impact of the age of B. alexandrina snails on their genetic variability and internal defence against S. mansoni infection. Susceptible and resistant snails were reared individually for self-reproduction; four subgroups of their progeny were used in experiment. The young susceptible subgroup showed the highest infection rate, the shortest pre-patent period, the highest total cercarial production, the highest mortality rate and the lowest SOD1 activity. Among the young and adult susceptible subgroups, 8% and 26% were found to be resistant, indicating the inheritance of resistance alleles from parents. The adult resistant subgroup, however, contained only resistant snails and showed the highest enzyme activity. The complex interaction between snail age, genetic background and internal defence resulted in great variability in compatibility patterns, with the highest significant difference between young susceptible and adult resistant snails. The results demonstrate that resistance alleles function to a greater degree in adults, with higher SOD1 activity and provide potential implications for Biomphalaria control. The identification of the most susceptible snail age enables determination of the best timing for applying molluscicides. Moreover, adult resistant snails could be beneficial in biological snail control.  相似文献

Forty-day-old seedlings of Atriplex halimus were treated either with NaCl (50, 300 and 550 mM) for the subsequent 30 days or with 15% PEG for the subsequent 10 days. As much as 50 mM of NaCl significantly increased shoot fresh and dry weight and height; nevertheless, 300 or 550 mM NaCl seemed to have no effect. On the other hand, these growth parameters were not affected by drought after 3 or 6 days, but were reduced after 10 days. The gas exchange parameters (photosynthetic rate, stomatal conductance and transpiration rate) were increased by 50 mM NaCl, but decreased by 300 and 550 mM. These parameters were decreased in response to drought only after 10 days of withholding water. In contrast to Na+, K+ was significantly decreased by NaCl but not by drought. The time course effect revealed that phosphoenol pyruvate carboxylase (PEPC) protein was doubled in response to NaCl after 1 and 5 h and continued thereafter, higher than control, while drought had no significant effect. Rubisco seemed unchanged by NaCl or drought. It could be concluded that the decrease in fresh weight might be attributed to the decrease in water content. Moreover, the decrease in photosynthesis could result from a decrease in stomatal conductance, a protective mechanism against water loss to improve water use efficiency. These findings indicate that Atriplex halimus tolerates NaCl and drought through decreasing growth, reducing gas exchange parameters to improve water use efficiency, uptake Na+ and saving, if any, the photosynthetic enzyme particularly PEPC.  相似文献

Introduction and aimBlastocystis is a common enteric parasite, having a worldwide distribution. Many antimicrobial agents are effective against it, yet side effects and drug resistance have been reported. Thus, ongoing trials are being conducted for exploring anti-Blastocystis alternatives. Proteases are attractive anti-protozoal drug targets, having documented roles in Blastocystis. Serine proteases are present in both hepatitis C virus and Blastocystis. Since drug repositioning is quite trendy, the in vitro efficacy of simeprevir (SMV), an anti-hepatitis serine protease inhibitor, against Blastocystis was investigated in the current study.MethodsStool samples were collected from patients, Alexandria, Egypt. Concentrated stools were screened using direct smears, trichrome, and modified Ziehl-Neelsen stains to exclude parasitic co-infections. Positive stool isolates were cultivated, molecularly subtyped for assessing the efficacy of three SMV doses (100,150, and 200 μg/ml) along 72 hours (h), on the most common subtype, through monitoring parasite growth, viability, re-culture, and also via ultrastructure verification. The most efficient dose and duration were later tested on other subtypes.ResultsResults revealed that Blastocystis was detected in 54.17% of examined samples. Molecularly, ST3 predominated (62%), followed by ST1 (8.6%) and ST2 (3.4%). Ascending concentrations of SMV progressively inhibited growth, viability, and re-culture of treated Blastocystis, with a non-statistically significant difference when compared to the therapeutic control metronidazole (MTZ). The most efficient dose and duration against ST3 was 150 µg/ml for 72 h. This dose inhibited the growth of ST3, ST1, and ST2 with percentages of 95.19%, 94.83%, and 94.74%, successively and viability with percentages of 98.30%, 98.09%, and 97.96%, successively. This dose abolished Blastocystis upon re-culturing. Ultra-structurally, SMV induced rupture of Blastocystis cell membrane leading to necrotic death, versus the reported apoptotic death caused by MTZ. In conclusion, 150 µg/ml SMV for 72 h proved its efficacy against ST1, ST2, and ST3 Blastocystis, thus sparing the need for pre-treatment molecular subtyping in developing countries.  相似文献
