The detailed structure of histograms constructed from the results of synchronous measurements of the alpha-activity of 239Pu microsamples, conducted in Pushchino (Moscow Region, Russia) and on board the ship "Academician Fedorov" during the Arctic and Antarctic expeditions (2000 and 2001) was analyzed. It was shown that, if the histograms were constructed over a total period of 15 min and more, the "local zone effect" and the circadian periodicity in the similarity of histogram structure, observed during measurements in Pushchino, are not found in measurements on board the ship near the North Pole (latitude 82 degrees). If the histograms were constructed over a total time of 1 min, a strict periodicity ("stellar day" 23 h 56 min) in the appearance of similar histograms in Pushchino and the Arctic and a strict synchronicity of the appearance of similar histograms of measurements in Pushchino and on board the ship at the same local geographical time were observed. During the Antarctic expedition, the effect of synchronicity of the appearance of similar histograms in Pushchino and on board the ship decreased as the ship moved towards the South Pole and the difference in latitude between the points of measurements increased.  相似文献

The role of cAMP/protein kinase A (PKA)- and tyrosine kinase (TK)-dependent intracellular mechanisms in mediating the action of porcine growth hormone (GH) on insulin-like growth factor I (IGF-I) secretion by porcine ovarian granulosa cells was studied. It was observed that GH-induced stimulation of IGF-I secretion was accompanied by an increase in cAMP production. The stimulation of PKA by the addition of either a cAMP agonist or a phosphodiesterase inhibitor to the medium increased IGF-I release by the cells, indicating a direct stimulation of IGF-I release by cyclic nucleotides. Moreover, the stimulatory effect of GH on IGF-I was completely suppressed by the addition of the PKA blocker Rp-cAMPS. Neither TK blocker altered the basal IGF-I level, but both strongly suppressed the GH-induced increase in IGF-I accumulation. Taken together, these findings suggest that cAMP/PKA- and/or TK-dependent pathways may be involved in the mediation of GH action on IGF-I release by porcine granulosa cells.  相似文献

The probability of a series of substituted 1,2,4-tri- and tetrazole compounds and by these modified polymer film materials to inhibit the process of microbiological corrosion of metals has been investigated. Fungi-toxicity of the studied compounds and materials has been observed for Aspergillus, Penicillium and Trichoderma fungi whose metabolites initiate corrosion of ferrous and nonferrous materials. For Thiobacillus ferrooxidans as an example, the bactericidal properties have been studied and azoles have been proven to suppress test-culture growth in culture medium. A comparative analysis of fungi and bactericidal activity of the studied compounds has been carried out. According to experimental results of kinetics of modifier desorption from the polymer matrix, the microbicidal effect of modified films is determined along with the corrosion inhibitor (CI) biocidal properties by its volatility and the intensity of the liquid phase (plasticizer + CI) syneresis from the material bulk. It has been concluded that there are fair prospects of application of azoles and by azoles modified materials as means of protection against both microbiological and electrochemical corrosion.  相似文献

The aim of our study was to examine whether: (1) the exposure of bovine embryos to the BHV-1 virus in vitro can compromise their further development and alter the ultrastructural morphology of cellular organelles; (2) whether the zona pellucida (ZP) can be a barrier protecting embryos against infection; and (3) whether washing with trypsin after viral exposure can prevent virus penetration inside the embryo and subsequent virus-induced damages. The embryos were recovered from superovulated Holstein-Friesian donor cows on day 6 of the estrous cycle. Only compact morulas or early blastocysts were selected for experiments with virus incubation. We used the embryos either with intact ZP (either with or without trypsin washing) or embryos in which the ZP barrier was avoided by using the microinjection of a BHV-1 suspension under the ZP. ZP-intact embryos (n = 153) were exposed to BHV-1 at 10(6.16) TCID(50)/ml for 60 min, then washed in trypsin according to IETS guidelines and postincubated in synthetic oviduct fluid (SOF) medium for 48 h. Some of the embryos (n = 36) were microinjected with 20 pl of BHV-1 suspension under the ZP, the embryos were washed in SOF medium and cultured for 48 h. Embryo development was evaluated by morphological inspection, the presence of viral particles was determined both immunocytochemically, using fluorescent anti-IBR-FITC conjugate and by transmission electron microscopy (TEM) on the basis of the ultrastructure of the cellular organelles. It was found that BHV-1 exposure impairs embryo development to higher preimplantation stages independent of the presence of the ZP or the trypsin treatment step, as most of the embryos were arrested at the morula stage when compared with the control. Immunofluorescence analysis confirmed the presence of BHV-1 particles in about 75% of embryos that were passed through the trypsin treatment and in all the BHV-1-microinjected embryos. Ultrastructural analysis, using TEM, revealed the presence of virus-like particles inside the BHV-1-exposed embryos, where the trypsin washing step was omitted. Conversely, in trypsin-treated BHV-1-exposed embryos, TEM detected only the envelope-free virus-like particles adhered to pores of the ZP. The embryos that were microinjected with BHV-1 suspension showed the presence of BHV-1 particles, as well as ultrastructural alterations in cell organelles. Taken together these findings may suggest that BHV-1 infection compromises preimplantation development of bovine embryos in vitro and therefore the ZP may not be enough on its own to prevent virus-induced damage, unless it is not accompanied with trypsin washing.  相似文献

The influence of foreign transgene integration on the reproductive capabilities of rabbit males is not known. Therefore, we analyzed their ejaculate characteristics, reproductive capabilities, occurrence of pathological spermatozoa and histological structure of the testis. We have generated transgenic rabbits by microinjection of WAP-hFVIII gene into pronucleus of fertilized egg. We observed that the libido, volume and pH value of the ejaculate did not differ significantly between transgenic and non-transgenic male lines. The motility, concentration, osmolarity, thermoresistant test of spermatozoa (at 1 or 6 h) and the percentage of alive spermatozoa were significantly different (p < 0.001) among transgenic and non-transgenic males. No significant differences were found between transgenic and non-transgenic male lines in the occurrence of pathological spermatozoa and histology of the testis. The ability of spermatozoa from transgenic and non-transgenic males to fertilize eggs was ranged within 96 and 100%; while the yield of transgenic embryos ranged from 43 to 57%. Our results show that mammary gland specific over-expression mWAP-hFVIII gene construct does not affect reproductive traits of transgenic rabbit males.  相似文献

This paper analyzes data on the state of the pelagic microalgae community of the Kara Sea for all hydrological seasons. The data were obtained during complex in situ observations in 1996–2006. Four phases in the annual succession cycle of the phytoplankton of the nearshore continental area of the Kara Sea were identified: a prevernal phase (cryoflora bloom), vernal phase (ice-edge bloom), summer-fall phase (mixed synthesis phase), and winter phase (dormant phase). These periods are clearly distinguished from each other in their composition of dominating species complexes and quantitative characteristics, i.e., the numbers and biomasses of microalgae. In the investigated region, which is completely covered by ice during most of the year, the primary production processes begin at the same time as in ice-free coastal areas. Growth and blooming of cryoflora occur under the ice cover long before it breaks down. The peak development of the pelagic microalgae community, which is comparable to the spring peak of microalgae in ice-free coastal areas, is observed in the summer, when freshwater runoff from large rivers increases sharply.  相似文献

AIMS: The aim of our in vitro studies was to understand the role of leptin and the insulin-like growth factor I/insulin-like growth factor protein (IGF/IGFBP) system in controlling human ovarian function. METHODS: We studied the action of leptin (0, 1, 10, or 100 ng/ml) and immunoneutralization of IGF-I using specific antiserum (0.1%) on the release of progesterone (P), estradiol (E), oxytocin (OT), IGF-I, IGFBP-3, and prostaglandins F (PGF) by these cells using radioimmunoassay/immunoradiometric assay. RESULTS: It was found that leptin stimulated the secretion of OT, IGFBP-3, and PGF. It suppressed the secretion of E and IGF-I, but not P, into the medium. The addition of antiserum against IGF-I decreased IGF-I output, increased P, OT, IGFBP-3, and PGF secretion, and had no effect on E release. Immunoneutralization of IGF-I also prevented or reversed the effects of leptin on P, E, IGF-I, IGFBP-3, PGF, but not on OT. CONCLUSIONS: These observations (1) demonstrate that leptin directly controls the secretory activity of human ovarian cells, (2) confirm the involvement of IGF-I in the regulation of ovarian cells, and (3) suggest an inter-relationship between leptin and the IGF/IGFBP system in the control of these functions and the involvement of IGF/IGFBP system in mediating leptin action on the ovary.  相似文献

Seed development in flowering plants is initiated by the fusion of two male gametes with two female gametes--the egg cell and the central cell--which leads to the formation of an embryo and an endosperm, respectively. Fertilization-independent seed formation is actively repressed by the FERTILIZATION-INDEPENDENT SEED (FIS) Polycomb group (PcG) proteins, an evolutionarily conserved class of proteins that ensures the stable transmission of developmental decisions. The FIS proteins act together in a complex and modify their target genes by applying repressive methylation on histone H3 lysine 27. In addition to its function before fertilization, the FIS complex restricts endosperm proliferation. This function is likely to be achieved by imprinting the maternal alleles of FIS target genes. However, imprinting in the endosperm is controlled not only by the FIS complex but also by DNA methylation, and the interconnections between these two processes are now being investigated.  相似文献

The aim of the study was to define interrelationships between histopathological alterations in ovarian antral follicles and body condition in dairy cows with a tendency to emaciation (BCS 1 and 2) compared with dairy cows with normal body condition (BCS 3). The ovaries were recovered from slaughtered cyclic dairy cows (at the luteal phase of the cycle) of Czech Fleckvieh and Holstein breeds at different times of the post-partum period. The animals were estimated as belonging to certain grade of body condition score (BCS) according to a 5-point scale. Only dairy cows with BCS1 (emaciation; n=6), BCS2 (tendency to emaciation; n=5) and BCS3 (optimal body condition status; n=6) were available for the experiment. The ovarian samples were embedded into Technovit 7100 resin; the tissue sections were stained with buffered basic fuchsine with toluidine blue. For acidic mucopolysaccharides (aMPS) a combination of PAS-technique with Alcian blue was used. Histological analysis showed that emaciation was associated with an increased occurrence of late (cystic) and luteinization-related atresia in granulosa and theca cells and increased levels of aMPS in small atretic follicles. Our observations indicate that dairy cows with a tendency to emaciation (BCS 2) or emaciated (BCS 1) have elevated occurrence of late atresia and atresia with luteinization, while initial atresia is less. This expands our basic knowledge of ovarian histopathology providing new insight into the association of antral follicle atresia and body condition status in dairy cows.  相似文献
