NIGHT BLUE AND VICTORIA BLUE AS INDICATORS IN LIPOLYSIS MEDIA   总被引:4,自引:4,他引:0  
SUMMARY: Optimum conditions for the use of night blue and victoria blue as lipolysis indicators in fat emulsion agar medium required a dye strenth of 1: 15,000 in a medium of pH 8·0 containing 5% fat dispersed by hand shaking. Pour plates should contain 6 ml. and streak plates 10 ml. of the medium in a standard Petri dish. Incubation should be for 5 days at 30°. The pinkish-mauve medium with clear blue-zoned lipolytic colonies gave the same results as butter fat agar without dye but treated with CuSo4, when tested with 962 pure cultures. The inhibitory powers of the dye were assessed and although strongly toxic in the aqueous phase to Gram-positive bacteria, victoria blue appears to have none to slight inhibitory power in the fat agar medium: night blue suppressed growth to about the same extent as tributyrin. The lipolytic flora of butter and to some extent milk shows a remarkable dominance of micorcocci. Organisms lipolytic on fat agar media are able to produce appreciable acid in a fat emulsion in a liquid medium.  相似文献

Procedures are described for the extraction of unmodified andalkylated barley glutelin fractions and their subsequent separationby sodium dodecyl sulphate-polyacrylamide gel electrophoresis.The resulting separations are considerably clearer and sharperthan those previously published. One major polypeptide is presentin the salt-soluble and glutelin fractions in varying proportionsdepending on whether or not the salt-soluble fraction is extractedin the presence of a reducing agent. There is also variationin the amount of contaminating hordein polypeptides in the glutelinsand this appears to depend not only on the conditions used toextract hordein but also those used to extract the salt-solublefraction. The effects of forest fragmentation on ecological interactions and particularly on food webs have scarcely been analysed. There is usually less herbivory in forest fragments than in continuous forests. Here we hypothesize that forest fragmentation enhances top‐down control of herbivory through an increase in insectivorous birds and a decrease in herbivorous insects, with a consequent decrease in plant reproductive success in small forest fragments. In the Maulino forest in central Chile, we experimentally excluded birds from Aristotelia chilensis (Elaeocarpaceae) trees in both forest fragments and continuous forest, and analysed herbivore insect abundance, herbivory and plant reproductive success during two consecutive growing seasons. We expected that insect abundance and herbivory would increase, and reproductive success would decrease in A. chilensis from which birds have been excluded, particularly in forest fragments where bird abundance and predation pressure on insects is higher. The abundance of herbivorous insects was lower in the forest fragments than in the continuous forest only in the first season, and herbivory was lower in forest fragments than in the continuous forest throughout the study. Moreover, during the second growing season herbivory was greater in the excluded trees than in the control trees, and as expected, there was a greater difference in the fragments than in the continuous forest, but this was not statistically significant. Exclusion of birds did not affect the reproductive success of A. chilensis. Our results, after 2 years of study, demonstrate that birds affect the levels of herbivory on A. chilensis in the Maulino forest, but do not support our hypothesis of enhanced top‐down control in fragmented forests, as the strength of the effect of excluding birds did not vary with fragmentation.  相似文献

1. Many insects are expanding their distribution range polewards as a result of climate change, which has been shown to be associated with founder effects leading to a reduction in genetic diversity and an increase in genetic differentiation. These spatial genetic patterns may arise from colonisation from a broad expansion front or a limited neighbourhood after a stepping stone model of dispersal. The temporal persistence of such founder effects are poorly understood, mainly because studies looking at the fine‐scale initial temporal dynamics of the genetic signature of a range expansion are rare. 2. Using microsatellite markers, we performed a detailed spatiotemporal genetic analysis of the range expanding damselfly Coenagrion scitulum (Rambur) along a coastal axis during the first years after colonisation. 3. A decrease was in (private) allelic richness when going northwards along the coastline, which is consistent with a scenario of cumulative founder events. In spite of the spatiotemporal dynamics in the observation records of the species along the coastline, the spatial genetic data indicated a major contribution from the broad expansion front during the colonisation of the coastline rather than a stepping‐stone colonisation process. 4. The fine‐scale temporal dynamics of the range expansion indicated the absence of persistent founder effects and instead showed considerable temporal instability in genetic indices at the more northern edge populations. This may be explained by genetic immigration and admixture from the broad expansion front in this active disperser.  相似文献

The evolution and maintenance of conspicuous animal traits and communication signals have long fascinated biologists. Many yellow–red conspicuous traits are coloured by carotenoid pigments, and in some species they are displayed at a very young age. In nestling birds, the functions and proximate mechanisms of carotenoid‐pigmented traits are probably different and not as well known as those of adults. Here we investigated how Montagu's harrier (Circus pygargus) nestlings within structured families used a limited resource, carotenoid pigments, and whether they used these for increasing coloration (deposition in integuments) or for mounting a response to a phytohaemagglutinin (PHA) challenge, which measures pro‐inflammatory potential and aspects of cellular immune responsiveness. We manipulated carotenoid availability, using dietary carotenoid supplementations, and show that when supplemented, nestlings primarily allocated supplemental carotenoids to increase their coloration, irrespective of their sex, but depending of their position within the brood. Responses to PHA challenge were condition‐dependent, but depending on carotenoid availability. Moreover, how nestlings allocated carotenoids depended on their rank within the brood, which in turn influenced their level of carotenoid limitation (first‐hatched nestlings being less constrained than later‐hatched nestlings). We discuss why nestlings would use supplemental carotenoids for increasing bare parts coloration rather than for responding to a PHA challenge, and the potential benefits for doing so in a parent–offspring communication context. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, 105 , 13–24.  相似文献

Abstract.  1. Habitat loss and fragmentation are the main causes of changes in the distribution and abundance of organisms, and are usually considered to negatively affect the abundance and species richness of organisms in a landscape. Nevertheless, habitat loss and fragmentation have often been confused, and the reported negative effects may only be the result of habitat loss alone, with habitat fragmentation having nil or even positive effects on abundance and species richness.
2. Manipulated alfalfa micro-landscapes and coccinellids (Coleoptera: Coccinellidae) are used to test the effects habitat loss (0% or 84%), fragmentation (4 or 16 fragments), and isolation (2 or 6 m between fragments) on the density, species richness, and distribution of native and exotic species of coccinellids.
3. Generally, when considering only the individuals in the remaining fragments, habitat loss had variable effects while habitat fragmentation had a positive effect on the density of two species of coccinellids and on species richness, but did not affect two other species. Isolation usually had no effect. When individuals in the whole landscape were considered, negative effects of habitat loss became apparent for most species, but the positive effects of fragmentation remained only for one species.
4. Native and exotic species of coccinellids did not segregate in the different landscapes, and strong positive associations were found most often in landscapes with higher fragmentation and isolation.
5. The opposing effects of habitat loss and fragmentation may result in a nil global effect; therefore it is important to separate their effects when studying populations in fragmented landscapes.  相似文献
