Despite the historical relationship between embryology and evolutionarybiology, during most of the 20th century developmental biologyand evolutionary biology have existed as two separate subdisciplinesof biology. Recently a reunion of these subdisciplines beganto take place. Inherent differences in the responses of stomata on abaxialand adaxial epidermal surfaces of leaves of Commelina communishave previously been suggested to be due to differences in theconcentrations of apoplastic Ca. Adaxial stomata have also beenreported to be more sensitive than abaxial stomata to appliedabscisic acid (ABA). The aims of these experiments were to determinethe validity of these conclusions and to see if xylem sap Cahas a role in determining the response of stomata to ABA. It can be shown from measurements of relative stomatal resistance(determined with a viscous flow porometer) and stomatal conductancethat stomata were more open in plants grown on 8-0 mol m3Ca, than with those grown on 2-0 mol m3 Ca. When attachedleaves were fed with ABA via the transpiration stream neitherthe extent nor the rate with which conductance declined wasdependent on Ca nutrition. The extent of Ca accumulation within both epidermes was relatedto the concentration of Ca in the rhizosphere and in the xylemsap. It did not, however, appear to reflect the apparent differencesin the flux of the transpiration stream between the two epidermes.Plants growing at the lower Ca concentration accumulated proportionallymore epidermal Ca relative to Ca in xylem sap. The evidencepresented suggests that Ca movement from the xylem to the epidermiscannot be simply described by a mass flow model, and that thedistribution of Ca is not an adequate explanation of the differencesin the behaviour of adaxial and abaxial stomata. Energy dispersive X-ray microanalysis of 83 eggs of G. rostochiensis for calcium content showed that juveniles from eggs exposed to active hatching factor in potato root diffusate for 48 h contained significantly more calcium than those exposed for this time to the same diffusate inactivated by autoclaving, or to water. This occurred despite a slightly greater calcium concentration in the autoclaved than in the untreated diffusate; both contained more calcium than the water. Eggshells removed from stimulated eggs also contained more calcium than those from unstimulated eggs. The calcium content of juveniles and eggshells exposed to inactivated diffusate was similar to their corresponding values for eggs soaked in water. A similar analysis was made of freeze-dried samples of fluid taken from the matrix surrounding the eggs in cysts exposed to water or to active root diffusate. This showed a significantly greater calcium concentration in the stimulated cysts after 48 h exposure. The concentration subsequently decreased over the succeeding 72 h however, suggesting that the rate of calcium uptake by the stimulated eggs exceeded that of diffusion into the cyst. This uptake of calcium by eggs of G. rostochiensis exposed to a hatching stimulus seems pertinent to recent evidence that the active factor in potato root diffusate may initiate hatching through a calcium-mediated process. 相似文献

In order to identify proteinaceous secretions involved in feeding of the soya bean cyst nematode, Heterodera glycines, over 1800 monoclonal antibodies (MAbs) were raised in four experiments using extracts of second stage juveniles and adult females as the immunogens. A rapid, indirect immunofluorescent screening procedure is described that enabled the antigens for 100 MAbs/day to be localised in juveniles. A similar immunofluorescent screening procedure for adult females was less successful due to the masking of the fluorophore by autofluorescence; additional approaches, including enzyme linked immunosorbent assays (ELISA), were necessary to aid in the screening process. The immunosuppressive drug, cyclophosphamide, had an effect on the range of antibodies obtained in two experiments. One was designed to favour antibodies specific to the hatched rather than the unhatched juvenile, whereas the second aimed to select those for the anterior rather than the posterior part of the adult female. A large number of MAbs showed well defined specificities and numbers are given in parentheses for those that recognised the following potential sites of secretion into the plant: the subventral pharyngeal glands (84), the dorsal pharyngeal gland (5), the amphidial pouches (10) and the excretory system (15). These MAbs will aid further work to define the role of nematode secretions in host invasion and the feeding process of this cyst nematode. 相似文献
