Variation at five polymorphic microsatellite loci was used to investigate genetic diversity and differentiation of two tetraploid Canarian endemics, Bencomia exstipulata and B. caudata. Data were analysed and are discussed in terms of tetrasomic (autotetraploid) and disomic (allotetraploid) inheritance. In both cases, genetic diversity values were similar to those described in other tetraploid plant species. High genetic differentiation between the only two described natural populations of B. exstipulata was detected (FST = 0.411). Bayesian cluster analysis revealed a geographical structure with distinct genetic groups from each island. High genetic differentiation and low genetic diversity of the B. exstipulata population from Tenerife suggest a recent population bottleneck, perhaps caused by the most recent major volcanic eruption, for this natural locality. This may be heightened by possible inbreeding depression and the monoecy of these species. Polymorphic microsatellite loci were also tested across all species in the Bencomia alliance. 1. Elevated allochthonous inputs of nutrients and sediments to aquatic ecosystems are associated with eutrophication and sedimentation. Reservoirs receive substantial subsidies of nutrients and sediments from catchments due to their large catchment : lake area ratios. We examined the effect of elevated subsidies of sediments and/or dissolved nutrients on the success (survival, growth, biomass and condition factor) of larval gizzard shad (Dorosoma cepedianum), a widespread and dominant omnivorous fish in reservoir ecosystems. 2. We simulated allochthonous agricultural subsides by manipulating dissolved nutrients and sediment inputs in a 2 × 2 factorial design in experimental mesocosms. We predicted that larval fish success would be greater under elevated nutrients. However, we propose two alternative hypotheses with respect to the overall effect of allochthonous sediment inputs. If sediment inputs negatively affect larval gizzard feeding success, larval success would be highest when only nutrients are added and lowest when only sediments are added (+N > +N+S ≥ C > +S). If high turbidity enhances larval foraging activity (due to greater contrast between prey and background), we predict that larval success would be highest when both subsidy types (nutrients and sediment) are elevated, intermediate when either nutrients or sediments are added and the lowest when no subsidies are added (+N+S > +N ≥ +S > C). 3. Our results indicate that elevated nutrient and sediment conditions enhanced larval gizzard shad biomass, but the overall nutrient addition effect was greater than the sediment addition effect (+N ~ +N+S > +S > C). We observed differential effects of nutrient and sediment inputs on larval survival, growth and condition factors. 4. The enhancement of fish biomass in elevated nutrients (+N, +N+S) relative to control conditions was associated with improved gizzard shad survival and not greater growth. The enhancement of fish biomass in the elevated sediment treatment (+S) relative to the control conditions was caused by an increase in survival that more than compensated for a negative effect of sediment addition on growth. 5. Our findings support the recommendation that reservoir management practices must consider the links between land use practices and food web dynamics. Our results suggest that reduction of subsidies of nutrients and sediments to productive reservoirs would decrease survival of larval gizzard shad due to lower food availability.  相似文献

The immune system has high costs of maintenance and production and expression of immune function should be dependent on the condition of the individual. The fitness cost that parasites impose on hosts may then be reflected in the investment that hosts make in immunity. Little is known about sources and consequences of intra- and interspecific variation in the presence and size of organs of the immune system in relation to ectoparasitism. Variation in the presence and size of the bursa of Fabricius and spleen size was measured to assess potential relationships between immune defence, nutritional condition and ectoparasites in the Magpie Pica pica. We found three species of lice (Mallophaga), two species of louse flies (Hippoboscidae), and the larvae of a tick (Ixodidae) infecting magpies. Age explained a significant proportion of variation in prevalence and number of parasite species, first-year birds being more parasitized than adults. Spleen size increased with nutritional condition, and the level of ectoparasitism negatively affected nutritional condition. Only one species of lice (Philopterus picae) was involved; the negative effect of this species on nutritional condition was more pronounced in males than females. These results are consistent with the hypotheses of condition-dependent investment in the immune system and sexual differences in susceptibility to parasite effects. Ectoparasites may thus play a role in modulating the relationship between condition and immune defence. We also found evidence for an interaction between the bursa and the spleen. Magpies with bursa had larger spleens than those where it had atrophied. This interaction did not affect ectoparasitism or nutritional condition.  相似文献

During early August to late September 1998 we examined seed dispersal in the tree species Jatropha standleyi (widespread) and J. chamelensis (endemic; Euphorbiaceae) at the Estacion de Biologia Chamela, Jalisco, Mexico. Using general linear models, we found that seed removal from beneath the parental canopy did not differ between the two species and that seed removal was correlated with both seed availability on the ground and rodent predation. We inferred that seed removal was largely mediated by rodents and determined that if a seed was not removed within 4d (approx. 16% chance), it was highly unlikely to be removed at all (approx. 3.5% chance) for both species. We note that although differences in patterns of seed removal may not explain the observed differences in range size, differences between the two species in seed crop size arc similar to patterns found in other endemic/widespread species pairs.  相似文献

In Mexico, mosquito vector‐borne diseases are of public health concern as a result of their impact on human morbidity and mortality. The use of insecticides against adult mosquitoes is one of the most common ways of controlling mosquito population densities. However, the use of these compounds has resulted in the development of insecticide resistance. The aim of this study was to estimate susceptibility to six pyrethroids, two carbamates and two organophosphates in Mexican populations of Stegomyia aegypti (Linnaeus, 1762) (= Aedes aegypti) (Diptera: Culicidae) mosquitoes. Bottle insecticide susceptibility tests, with 1 h exposure, were performed on adult mosquitoes from 75 localities across 28 states. At 30 min of exposure, the proportion of fallen mosquitoes was recorded. After 60 min of exposure, mosquitoes were recovered in non‐treated containers and mortality was determined at 24 h after the set‐up of the experiment. In general, the carbamate insecticides represented the most effective group in terms of the proportion of mosquitoes fallen at 30 min (72–100%) and 24‐h mortality (97–100%). High and widespread resistance to pyrethroids Types I and II and, to a lesser extent, to organophosphates was observed. Insecticide susceptibility among and within states was highly variable.  相似文献

1. Cerambyx welensii (Cw) and Cerambyx cerdo (Cc) are two large saproxylic beetles living on Quercus trees in the Western Palearctic whose current pest and legal status differs markedly. Cw is an emerging pest involved in oak decline while Cc is an internationally protected species. 2. Acquiring knowledge of the ecology, demography and behaviour of Cw and Cc in forests harbouring mixed populations is a demanding task to optimise their management or protection. Here, we report the results of a mark–recapture study on the flight behaviour and dispersal potential of both species in holm oak open woodlands. 3. Average flights, dispersal rates and diffusion models were remarkably similar in Cw and Cc, reflecting a low‐dispersal tendency and sedentary behaviour. However, a subset of adults in both species exhibited a huge propensity to disperse and flew more than 1 km. An aggregated distribution of dispersal distances suggested that a behavioural polymorphism might underlie the flight pattern. 4. Adults moved preferentially in those spatial directions with neighbouring trees, while prevailing winds did not significantly affect dispersal patterns. The main interspecific differences were as follows: (i) Cc performed longer crosswind flights than Cw on windy days; (ii) Cc tended to fly farther than Cw at low temperatures; and (iii) adult feeding improved dispersal but only in small Cw females. Moon phase did not affect flight activity. 5. The results are discussed in an effort to interpret how these ecological and behavioural differences might shape the life history of both congeneric species when they live in sympatry in dehesa woodlands.  相似文献

An electrochemical approach to directly measure the dynamic process of H2O2 release from cultures of Arabidopsis thaliana cells is reported. This approach is based on H2O2 oxidation on a Pt electrode in conjunction with continuous measurement of sample pH. For [H2O2] <1 mm , calibration plots were linear and the amperometric response of the electrode was maximum at pH 6. At higher concentrations ([H2O2] >1 mm ), the amperometric response can be described by Michaelian‐type kinetics and a mathematical expression relating current intensity and pH was obtained to quantitatively determine H2O2 concentration. At pH 5.5, the detection limit of the sensor was 3.1 µm (S/N = 3), with a response sensitivity of 0.16 Am ?1 cm?2 and reproducibility was within 6.1% in the range 1–5 × 10?3 m (n = 5). Cell suspensions under normal physiological conditions had a pH between 5.5–5.7 and H2O2 concentrations in the range 7.0–20.5 µm (n = 5). The addition of exogenous H2O2, as well as other potential stress stimuli, was made to the cells and the change in H2O2 concentration was monitored. This real‐time quantitative H2O2 analysis is a potential marker for the evaluation of oxidative stress in plant cell cultures.  相似文献

SYNOPSIS The surface charge of epimastigote and trypomastigote forms of Trypanosoma cruzi was evaluated by means of binding of cationized ferritin to the cell surface as visualized by electron microscopy, and by direct measurements of the cellular microelectrophoretic mobility (EPM). Epimastigote forms had a mean EPM of -0.52 μm.s-1.V-1.cm and were lightly labeled with cationized ferritin. In contrast, bloodstream trypomastigotes had a much higher EPM (-1.14), and the surface was heavily labeled with cationized ferritin. When trypomastigotes from stationary phase cultures were isolated on DEAE cellulose columns, the mean EPM was found to be significantly lower (-0.63), and labeling with cationized ferritin decreased. With a mixed population containing epimastigote, trypomastigote, and intermediate forms, EPM values ranging between -0.70 to -1.14 were found. From these observations we conclude that there is a definite increase in negative surface charge during development from epi- to trypomastigote forms of T. cruzi.  相似文献

ABSTRACT. Strains of the opportunistic fungal pathogen Candida albicans vary in the presence or absence of a self‐splicing group I intron ribozyme (Ca.LSU) in the 25S rRNA gene on chromosome R. Strains of C. albicans typically either lack or contain this ribozyme. However, some strains have both intron‐containing and intronless rRNA genes (rDNA). Pulsed‐field gel electrophoresis analysis of undigested and restricted DNA showed at least six different karyotypes among eight independent colonies of such a heteroallelic strain. In each case, the variation was in chromosome R, and was due to changes in the number of rDNA units. In strains with only one type of rDNA, chromosome R also varied considerably. Polymerase chain reaction amplification spanning the rDNA unit demonstrated that intron‐containing rDNA units are tandemly arrayed, and are immediately adjacent to intronless units in the same cluster. Both types of units were present in the rDNA clusters of both R chromosomes. Possible explanations of these results are loss of Ca.LSU group I intron through purifying selection and/or a relaxation of the commonly accepted concerted evolution of the rDNA units.  相似文献

The taxonomic status of a very poorly known group of Andean frogs (the Eleutherodactylus discoidalis group) is assessed through acoustic and statistical analyses of differences in temporal parameters of advertisement calls, such as the number of pulses and the call duration, and also in a spectral parameter, dominant frequency. As these species are usually misidentified or ignored because of their taxonomic complexity in both ecological and biodiversity studies, we provide a bioacoustical diagnosis for each species in order to facilitate identification in the field. Differences in acoustic parameters support the specific status of Eleutherodactylus cruralis , E. discoidalis , Eleutherodactylus ibischi , and Eleutherodactylus madidi . The name E. cruralis is probably applied to three different species: the nominal form from Amazonian forests of the Andean slopes and adjacent lowlands, and two cryptic species restricted to inter-Andean dry valleys and cloud forests from central Bolivia. Moreover, the distribution of energy through the call and the aggregation of pulses seem to be useful meristic characters for detecting interspecific differences. Populations from each macrohabitat can be recognized by distinctive advertisement calls, usually corresponding to a recognized species. For the whole group, pulse rate is significantly correlated to latitude, which could indicate a speciation process along the Andes in relation to habitat changes and isolation. © 2008 The Linnean Society of London, Zoological Journal of the Linnean Society , 2008, 152 , 353–365.  相似文献
