ABSTRACT. Trypanosoma cruzi epimastigote motility can be enhanced by addition of L-arginine, to the culture. This effect is blocked by N-methyl-L-arginine, a competitive inhibitor of the nitric oxide synthase. N-methyl-D-aspartate and L-glutamate, two agonists of the NMDALglutamate receptor, also enhanced motility. This stimulation is blocked by MK-801 a noncompetitive antagonist of the NMDA receptor. In addition, sodium nitroprusside, a guanylyl cyclase stimulator and 8-Br-cyclic GMP, an analog of cyclic GMP, also stimulated epimastigote motility. It is suggested that an increase of intracellular cyclic GMP levels mediated by nitric oxide may be responsible for the increase in epimastigote motility.  相似文献

Abstract.  The New World screwworm, Cochliomyia hominivorax (Coquerel) (Diptera: Calliphoridae), is one of the most important myiasis-causing flies in South America. It is responsible for severe economic losses to livestock producers, mainly because it causes mortality in newborn calves and reductions in the quality of leather and in the production of milk and meat. The economic losses caused by myiasis, along with those caused by other internal and external parasites, are the main factors limiting meat production. In Brazil, C. hominivorax has been controlled by applying insecticides, particularly organophosphate (OP)-based compounds. However, the improper and continuous use of these chemicals can lead to the selection of OP-resistant strains. This, associated with the fast development of OP resistance in other myiasis-causing flies, shows the importance of investigating resistance in C. hominivorax. Based on the findings of previous studies, the objective of the current work was to isolate and sequence the E3 gene in C. hominivorax. Mutations at the positions (Gly137 and Trp251) responsible for conferring OP resistance in Lucilia cuprina and Musca domestica L. (Muscidae) were identified in C. hominivorax . In addition, the orthologous region in C. hominivorax contained motifs that are highly conserved among carboxyl/cholinesterases and contribute to the catalytic mechanism of the active site. The characterization of this gene in natural populations of New World screwworm can be an important tool for monitoring resistance to insecticides throughout its current geographic distribution. This will provide information for the selection and implementation of more effective pest management programmes.  相似文献

ABSTRACT We developed predictive habitat models for a bighorn sheep (Ovis Canadensis) population in the Peninsular Ranges of southern California, USA, using 2 Geographic Information System modeling techniques, Ecological Niche Factor Analysis (ENFA) and Genetic Algorithm for Rule-set Production (GARP). We used >16,000 Global Positioning System locations from 34 animals in 5 subpopulations to develop and test ENFA and GARP models, and we then compared these models to each other and to the expert-based model presented in the United States Fish and Wildlife Service's Recovery Plan for this population. Based on a suite of evaluation methods, we found both ENFA and GARP to provide useful predictions of habitat; however, models developed with GARP appeared to have higher predictive power. Habitat delineations resulting from GARP models were similar to the expert-based model, affirming that the expert-based model provided a useful delineation of bighorn sheep habitat in the Peninsular Ranges. In addition, all 3 models identified continuous bighorn sheep habitat from the northern to southern extent of our study area, indicating that the Recovery Plan's recommendation of maintaining habitat connectivity throughout the range is an appropriate goal.  相似文献

Hydrocleys martii (Limnocharitaceae) is an annual aquatic herb common in ephemeral isolated ponds in semi‐arid northe‐astern Brazil. We studied pollination of H. martii, emphasizing reproductive success and association with oligolectic pollinators. The yellow flowers bear a central cone of staminodes that encloses the fertile stamens and four free carpels. The self‐incompatible species depends on pollinators to set fruits. In 25 temporary water bodies in five Brazilian states, Protodiscelis palpalis bees (Colletidae, Paracolletinae) were the unique effective pollinators of H. martii and, in 18 of these, the sole flower visitors. Females of this narrowly oligolectic species show adapted behaviour to access the pollen chamber in the flower centre. Females removed more than 80% of the 480 000 pollen grains in only 2 h from the flowers but maintained a high visitation frequency almost until flower senescence. In this highly specialized plant‐pollinator system, on average, 1.6% of the pollen grains reached the stigmatic surface and 9.6% remained uncollectible in the flowers. In the absence of P. palpalis, flowers set almost no seeds, indicating reproductive dependence on the oligolectic species. This pioneer pollination study of a species of Limnocharitaceae provides evidence of a close relationship of the family to species of Protodiscelis, their specific pollinators. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 102 , 355–368.  相似文献

1. The evolutionary advantages that have driven the evolution of sex are still very much debated, and a number of benefits of parthenogenesis over sexual reproduction have been proposed. In particular, parthenogenetic individuals are thought to exhibit higher probabilities of establishment following arrival in new, isolated habitats such as islands. 2. One notable example of parthenogenesis occurring in islands is the damselfly Ischnura hastata, an American species that has colonised the Azores archipelago, where the populations consist only of females. This is the only known example of parthenogenesis within the insect order Odonata. 3. Here, two island populations of I. hastata were studied, one in the Galapagos and one in Cuba, to test whether island colonisation is consistently associated with parthenogenesis in this species. Field capture–mark–recapture studies and laboratory rearing of field‐collected eggs were undertaken in both areas. 4. Sex ratios in the field were found to be heavily female‐biased among mature individuals; however, fertility rates of field‐collected eggs were high, and the sex ratios in the laboratory did not differ from 1 : 1. Data from laboratory rearing showed that shorter larval development times and shorter adult life spans in males result in protandry, which might explain the skewed sex ratios in the field. 5. These findings are consistent with sex differences in key demographic parameters which could predispose I. hastata to parthenogenesis. However, the Azores population of I. hastata remains the only documented case of asexual reproduction in this insect group.  相似文献

ABSTRACT. Trypanosoma cruzi trypomastigotes survive inside macrophages by promoting fusion between the parasitophorous vacuole and mature host lysosomes upon internalization. Since trypomastigotes can evade the lytic pathway, the earliest steps of endocytosis, such as early endosome fusion, may be affected. To test this hypothesis, we used an in vitro early endosome fusion assay. Our results show that trypomastigote-infected macrophage cytosols cannot promote fusion between early endosomes, compared to mock-infected cytosols (heat-killed trypomastigotes were used in the parasite-macrophage interaction assay). GTPγS addition potentiates the fusogenic activity driven by trypomastigote-infected macrophage cytosol-mediated assays, unlike the biphasic fusogenic effect obtained with GTPγS treatment of macrophage cytosol controls. Calcium-stimulated early endosome fusogenic processes are not affected in the assays mediated by infected macrophage cytosol. We conclude that GTP-regulated factors, and not calcium-regulated elements, are involved in the inhibition of the early endosome fusogenic process by the trypomastigote-infected macrophage cytosol. This primary impediment to the progress of a normal endocytosis may be a relevant step required for the lysosomal recruitment-fusion of the host lysosmes upon trypomastigote infection and further survival of the parasite within its host.  相似文献

1. This work investigated the consumption of a microalgal (Anabaena spiroides) exopolysaccharide by the cladoceran Simocephalus serrulatus (Cladocera, Daphnidae) and its effect on copper toxicity. 2. Total organic carbon concentration was used to quantify the microalgal exopolysaccharide. Both copper‐complexed exopolysaccharides and copper‐free exopolysaccharides were taken up by S. serrulatus. 3. A reduction of free copper ions was obtained in the presence of A. spiroides exopolysaccharide. Copper toxicity (EC50) and zooplankton concentrations were inversely correlated to exopolysaccharide concentration in experimental media. 4. An increase of free copper ions in experimental media was obtained after exopolysaccharide consumption, suggesting that S. serrulatus excreted copper to the surrounding environment.  相似文献

This study evaluated the seasonal dynamics of Rhipicephalus sanguineus (Latreille) (Acari: Ixodidae) on naturally infested dogs in a private shelter in southern Italy. From March to May 2008, 39 autochthonous mixed‐breed young dogs and 10 beagles were enrolled in the study. From March 2008 until March 2009, every 21 ± 2 days, 11 body sites of each dog were checked for ticks. At each follow‐up, the number of ticks, their developmental stage, sex and location on the dog's body were recorded. Adult ticks were found throughout the year, but immatures were absent in January and February. The adult tick population increased from July to August, whereas the load of immatures increased in early July and peaked in September, which suggests that R. sanguineus develops one generation per year in this area. The mean number of immature ticks per infested dog was higher than that of adults from March to October 2008. Ears, interdigital areas and armpits were the most frequent attachment sites of adult ticks. At the last follow‐up, a total of 2266 ticks were collected and identified as R. sanguineus. The results suggest that R. sanguineus develops one generation per year in the study area, but that it infests dogs in all seasons. This information should be taken into account when planning control programmes against this tick species and the pathogens it transmits.  相似文献

The pattern of cytospindle assembly and the modifications of the microtubular cytoplasmic network during division of Paramecium are studied by means of indirect immunofluorescence. The assembly of cytospindle starts at two independent areas placed respectively around the proter's and opisthe's buccal overture. The moment of the microtubule bundles’appearance depends on their distance from the buccal opening, with those closest appearing 1st. The existence of microtubule organizing centers that act transiently during division of Paramecium is discussed.  相似文献

Abstract. A high level of DDT resistance and low levels of resistance to organophosphorus, carbamate and pyrethroid insecticides were detected by discriminating dose assays in field populations of Anopheles albimanus in Chiapas, southern Mexico, prior to a large-scale resistance management project described by Hemingway et al. (1997) . Biochemical assays showed that the DDT resistance was caused by elevated levels of glutathione S-transferase (GST) activity leading to increased rates of metabolism of DDT to DDE. The numbers of individuals with elevated GST and DDT resistance were well correlated, suggesting that this is the only major DDT resistance mechanism in this population. The carbamate resistance in this population is conferred by an altered acetylcholinesterase (AChE) -based resistance mechanism. The level of resistance observed in the bioassays correlates with the frequency of individuals homozygous for the altered AChE allele. This suggests that the level of resistance conferred by this mechanism in its heterozygous state is below the level of detection by the WHO carbamate discriminating dosage bioassay. The low levels of organophosphate (OP) and pyrethroid resistance could be conferred by either the elevated esterase or monooxygenase enzymes. The esterases were elevated only with the substrate pNPA, and are unlikely to be causing broad spectrum OP resistance. The altered AChE mechanism may also be contributing to the OP but not the pyrethroid resistance. Significant differences in resistance gene frequencies were obtained from the F1 mosquitoes resulting from adults obtained by different collection methods. This may be caused by different insecticide selection pressures on the insects immediately prior to collection, or may be an indication that the indoor- and outdoor-resting A. albimanus collections are not from a randomly mating single population. The underlying genetic variability of the populations is currently being investigated by molecular methods.  相似文献
