Telomeric chromatin has different features with respect to bulk chromatin, since nucleosomal repeat along the chain is unusually short. We studied the role of telomeric DNA sequences on nucleosomal spacing in a model system. Nucleosomal arrays, assembled on a 1500-bp-long human telomeric DNA and on a DNA fragment containing 8 copies of the 601 strong nucleosome positioning sequence, have been studied at the single molecule level, by atomic force microscopy imaging. Random nucleosome positioning was found in the case of human telomeric DNA. On the contrary, nucleosome positioning on 601 DNA is characterized by preferential positions of nucleosome dyad axis each 200 bp. The AFM-derived nucleosome organization is in satisfactory agreement with that predicted by theoretical modeling, based on sequence-dependent DNA curvature and flexibility. The reported results show that DNA sequence has a main role, not only in mononucleosome thermodynamic stability, but also in the organization of nucleosomal arrays. 相似文献

Serotonin is a signal molecule with a wide range of functions in vertebrates. In Antarctic fishes, the serotonergic system
has been studied in the brain, revealing differences from temperate fishes related to the long-term cold adaptation. To date,
little is known regarding the peripheral nervous system, and no information is available for the stomach. In the present work,
we contribute to fill the gaps by investigating the presence and the immunohistochemical distribution of serotonin in the
stomach of the Antarctic silverfish Pleuragramma antarcticum, a cold-adapted key species of the Southern Ocean shelf food web. The main aim was to investigate the serotonergic system
at the gastric level, in order to reveal possible peculiarities related to long-term cold adaptation, similar to the ones
seen in the central nervous system of Antarctic fishes. Serotonin immunoreactivity was detected in the pyloric and cardiac
mucosa of P. antarcticum stomach with immunopositive cells in the pyloric and cardiac surface epithelia and in the tubular glands. No immunopositive
fibers and neuronal cell bodies were found. Our results highlight that the serotonin distribution pattern at the gastric level
is similar to that described in temperate teleosts. This finding suggests that in P. antarcticum, long-term adaptations to the Antarctic condition do not affect the serotonergic system at the gastric level. In addition,
our data constitute the baseline information for further investigations aimed at clarifying the effects of short-term temperature

TRF1 and TRF2 are key proteins in human telomeres, which, despite their similarities, have different behaviors upon DNA binding. Previous work has shown that unlike TRF1, TRF2 condenses telomeric, thus creating consequential negative torsion on the adjacent DNA, a property that is thought to lead to the stimulation of single-strand invasion and was proposed to favor telomeric DNA looping. In this report, we show that these activities, originating from the central TRFH domain of TRF2, are also displayed by the TRFH domain of TRF1 but are repressed in the full-length protein by the presence of an acidic domain at the N-terminus. Strikingly, a similar repression is observed on TRF2 through the binding of a TERRA-like RNA molecule to the N-terminus of TRF2. Phylogenetic and biochemical studies suggest that the N-terminal domains of TRF proteins originate from a gradual extension of the coding sequences of a duplicated ancestral gene with a consequential progressive alteration of the biochemical properties of these proteins. Overall, these data suggest that the N-termini of TRF1 and TRF2 have evolved to finely regulate their ability to condense DNA. 相似文献

Adarotene belongs to the so-called class of atypical retinoids. The presence of the phenolic hydroxyl group on Adarotene structure allows a rapid O-glucuronidation as a major mechanism of elimination of the drug, favoring a fast excretion of its glucuronide metabolite in the urines. A series of ether, carbamate and ester derivatives was synthesized. All of them were studied and evaluated for their stability at different pH. The cytotoxic activity in vitro on NCI-H460 non-small cell lung carcinoma and A2780 ovarian tumor cell lines was also tested. In the framework of the I.C.E.FISH (International Collaborative Effort on Antarctic Fish Adaptive Evolution) project, during the 1998-1999 season at the Italian shore-based Terra Nova Bay Station, the broad fields of adaptation and evolution of the fish suborder Notothenioidei were tackled through the integration of many disciplines. As a representative contribution of I.C.E.FISH, a cytogenetic study of the bathydraconid fish Gymnodraco acuticeps is reported. The Bathydraconidae is a heterogeneous notothenioid taxon whose intra-family relationships are very uncertain. The conventional karyotype and the location of nuclear ribosomal genes, by means of fluorescence in situ hybridisation, contributed to species-specific characterisation and provided the basis for comparative analyses. The karyotype of G. acuticeps consists of 48 chromosomes (2 metacentric, 2 submetacentric and 44 acrocentric); major ribosomal cistrons correspond to a large region in a single chromosome pair. Mapping of the available karyotypic data on a molecular phylogenetic tree provided information on chromosomal diversification during the cladogenesis of the bathydraconids. 相似文献

The culture filtrates of ten species of Cephalosporium, which had been grown under conditions of submerged culture, were tested for enzymatic activity against each of seven substrates. The latter included casein, gelatin, milk, hemoglobin, human plasma clots, starch, and N-acetyl-β-D-glucosaminide. All organisms tested were active, but to varying degrees. The most pronounced activities were obtained against the proteinaceous substrates. Two unidentified species of Cephalosporium exhibited the highest over-all activities, but no one organism predominated for all enzymatic functions. The ability of a filtrate to degrade a specific substrate was not always correlated with its ability to attack other substrates. The fibrinolytic properties of three of the cephalosporia were of particular interest. α-Amylase activity was not significant. The results obtained suggest the possible use of selected species of Cephalosporium as sources of a variety of microbial enzymes. 相似文献

Telomere integrity is essential to maintain genome stability, and telomeric dysfunctions are associated with cancer and aging pathologies. In human, the shelterin complex binds TTAGGG DNA repeats and provides capping to chromosome ends. Within shelterin, RAP1 is recruited through its interaction with TRF2, and TRF2 is required for telomere protection through a network of nucleic acid and protein interactions. RAP1 is one of the most conserved shelterin proteins although one unresolved question is how its interaction may influence TRF2 properties and regulate its capacity to bind multiple proteins. Through a combination of biochemical, biophysical and structural approaches, we unveiled a unique mode of assembly between RAP1 and TRF2. The complete interaction scheme between the full-length proteins involves a complex biphasic interaction of RAP1 that directly affects the binding properties of the assembly. These results reveal how a non-DNA binding protein can influence the properties of a DNA-binding partner by mutual conformational adjustments. 相似文献

The COVID-19 pandemic has presented significant challenges and implications for the sports community. Thus, this study aimed to describe the prevalence of COVID-19 in Brazilian athletes and identify the epidemiological, clinical, athletic, life and health factors associated with the disease in these individuals. A cross-sectional study was performed involving 414 athletes from 22 different sports using an online questionnaire from August to November 2020. The association between the athletes’ characteristics and COVID-19 was evaluated using a logistic regression model. The prevalence of COVID-19 was 8.5%, although only 40% of athletes reported having been tested. Being under 27 years of age (3-fold), having children (~5-fold), having a teammate test positive for COVID-19 (2.5-fold), and smoking (14-fold) were associated with a possible higher risk of disease. Almost 20% of athletes self-reported musculoskeletal injuries during the period of the pandemic that was studied. Athletes with a university education (P = 0.02), a profession other than sports (P < 0.001), those from a low-income family (P = 0.01), and public health system users (P = 0.04) were significantly less frequently tested for COVID-19, whereas international competitors, athletes who received a wage, and athletes who had a teammate who tested positive for COVID-19 were 2-, 3-, and 15-fold more likely to be tested for COVID-19, respectively. Approximately 26% of the athletes who tested negative or were untested reported more than three characteristic COVID-19 symptoms, and 11% of athletes who tested positive for COVID-19 were asymptomatic. The identification of modifiable (have children, smoking, and teammates positively tested) and non-modifiable (age under 27 years) factors related to COVID-19 in athletes can contribute to implementing surveillance programmes to decrease the incidence of COVID-19 in athletes and its negative impacts in sports. 相似文献
