It is crucial to examine the genetic diversity and structure of small, isolated populations, especially those at the edge of their distribution range, because they are vulnerable to stochastic processes if genetic diversity is low and the isolation level is high, and because such populations provide insight into the consequences of population declines in a broader conservation context. The harbour seal (Phoca vitulina) population in Svalbard is the world's northernmost P. vitulina population. Nothing is known about the genetic diversity, distinctiveness, or origin of this small, marginalized mammalian population. Thus, the present study investigated its genetic status in the context of nearby P. vitulina in Iceland, south‐east Greenland, and northern Norway: this species is depleted/threatened in all of these regions. A total of 174 samples distributed between the four locations were analysed using 15 polymorphic microsatellites and variation in the displacement loop region (D‐loop). Each of the four locations was a genetically distinct population. The Svalbard population was highly genetically distinct, had reduced genetic diversity, received limited gene flow, had a rather low effective population size, and showed an indication of having experienced a bottleneck resulting from a recent population decline. The significant heterozygote excess observed in the Svalbard sample might be attributed to the low effective population size, which could initiate future population inbreeding effects. This phenomenon has not been reported earlier from other P. vitulina populations, but if the Svalbard population is experiencing inbreeding, this could reduce its resilience to climate change, disease outbreaks, or other perturbations. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 102 , 420–439.  相似文献

Colonization of the roots of tobacco by Pseudomonas chlororaphis O6 induces systemic resistance to the soft-rot pathogen, Erwinia carotovora ssp. carotovara SCC1. A screen of the transposon mutants of P. chlororaphis O6 showed mutants with about a fivefold reduction in ability to induce systemic resistance to the soft-rot disease. These mutations disrupted genes involved in diverse functions: a methyl-accepting chemotaxis protein, biosynthesis of purines, phospholipase C, transport of branched-chain amino acids and an ABC transporter. Additional mutations were detected in the intergenic spacer regions between genes encoding a GGDEF protein and fumarate dehydratase, and in genes of unknown function. The mutants in the ABC transporters did not display reduced root colonization. However, the other mutants had up to 100-fold reduced colonization levels. Generally the production of metabolites important for interactions in the rhizosphere, phenazines and siderophores, was not altered by the mutations. A reduced induction of systemic resistance by a purine biosynthesis mutant with a disrupted purM gene correlated with poor growth rate, lesser production of phenazines and siderophore and low levels of root colonization. These studies showed that multiple determinants are involved in the induction of systemic resistance, with there being a requirement for strong root colonization.  相似文献

To determine how elevated night temperature interacts with carbon dioxide concentration ([CO2]) to affect methane (CH4) emission from rice paddy soil, we conducted a pot experiment using four controlled‐environment chambers and imposed a combination of two [CO2] levels (ambient: 380 ppm; elevated: 680 ppm) and two night temperatures (22 and 32 °C). The day temperature was maintained at 32 °C. Rice (cv. IR72) plants were grown outside until the early‐reproductive growth stage and then transferred to the chambers. After onset of the treatment, day and night CH4 fluxes were measured every week. The CH4 fluxes changed significantly with the growth stage, with the largest fluxes occurring around the heading stage in all treatments. The total CH4 emission during the treatment period was significantly increased by both elevated [CO2] (P=0.03) and elevated night temperature (P<0.01). Elevated [CO2] increased CH4 emission by 3.5% and 32.2% under high and low night temperature conditions, respectively. Elevated [CO2] increased the net dry weight of rice plants by 12.7% and 38.4% under high and low night temperature conditions, respectively. These results imply that increasing night temperature reduces the stimulatory effect of elevated [CO2] on both CH4 emission and rice growth. The CH4 emission during the day was larger than at night even under the high‐night‐temperature treatment (i.e. a constant temperature all day). This difference became larger after the heading stage. We observed significant correlations between the night respiration and daily CH4 flux (P<0.01). These results suggest that net plant photosynthesis contributes greatly to CH4 emission and that increasing night temperature reduces the stimulatory effect of elevated [CO2] on CH4 emission from rice paddy soil.  相似文献

The permeability to water of the fibre cell wall of birch (Betulapubescens Ehrh.) and lime (Tilia x vulgaris Hayne) wood, wasmeasured in wood slices in which most of the void space wasfilled with paraffin wax or a polymerizing silicone elastomer.An osmotic technique was used with solutes of molecular weightgreat enough to prevent their molecules from penetrating thepores in the water-swollen cell wall. The solutes used weredextran and polyethylene glycol with molecular weights of 40000 and 6000 respectively. Values for permeability k x 1021, as defined by the Darcy equation,ranged between 3.6 m2 for the tangential direction in birch,to 27 m2 for the longitudinal direction in birch. These resultsare in good agreement with previously measured values but areat least ten times less than theoretical values. It is calculatedthat total emptying or filling of a wood cell through the wallmight be possible in as little as 5 min under a pressure differenceof 0.1 MPa if other flow pathways were blocked. We investigate the role of parasite-mediated sexual selection in the divergence of two species of Lake Victoria cichlids. Pundamilia pundamilia and Pundamilia nyererei represent a common pattern of male nuptial colour divergence between haplochromine sister species: metallic grey–blue in P. pundamilia and bright yellow and red in P. nyererei . Female mating preferences for different male colours maintain the genetic and phenotypic differentiation of the two species in clear water. Previous work indicated that the red coloration of P. nyererei males, which is subject to directional sexual selection, may be a carotenoid-dependent signal of parasite infestation rate. In the present study, we find a parallel result for P. pundamilia : bright blue males are infected with fewer species of parasites. We also find that parasite infestation rates differ quantitatively between the two species in a way that is consistent with species differences in diet and microhabitat. We conclude that parasite-mediated sexual selection may have contributed to the divergence of female mating preferences between P. pundamilia and P. nyererei , and may currently strengthen reproductive isolation between these species.  © 2008 The Linnean Society of London, Biological Journal of the Linnean Society, 2008, 94 , 53–60.  相似文献

Using a model system of 3mM NaCl and purified water, predictions were made for subjects’ability to discriminate the odd sample in a set of nine triadic tasting sequences associated with the triangle test. With such individual triads rather than triangular testing protocols which involve more than one triad, it was found necessary to modify the regular Sequential Sensitivity Analysis. The modification required a consideration of sensory adaptation effects caused by repeated tasting of the same stimulus in some of the sequences. Further confirmation was also obtained for the order of magnitude of signal strengths of water and NaCl stimuli tasted after water or NaCl prerinses.  相似文献

1. Cyclical parthenogens, organisms switching between sexual and asexual mode of reproduction, are common in aquatic habitats. In permanent waterbodies with year‐round persistence of populations, the relevance of sexually produced resting eggs for population dynamics and structure is still largely unexplored. 2. The aim of this study was to investigate the importance of recruitment from resting eggs to Daphnia spp. in a deep dimictic reservoir with overwintering populations (Saidenbach Reservoir, Germany). We linked field studies on Daphnia population dynamics, hatching rates and diapausing egg abundances in the sediment with allozyme analysis of the genetic variability of both the pelagic population and hatchlings from resting eggs. 3. We found higher hatching rates at intermediate depths (10–18 m), although highest diapausing egg abundances were found in the deeper parts of the reservoir. From the extensive field data, we calculated a population budget for the main basin of the reservoir, which showed that hatched daphnids accounted for <1% of the total abundance and explained only a tiny fraction of population growth. 4. The genetic composition (MultiLocus Genotypes, MLG) of the pelagic population was relatively stable throughout the stratified period with up to five among 39 detected MLGs attaining frequencies >5% per date. From 11 MLGs differentiated among hatched daphnids, three were recovered in the pelagic population in perceptible, albeit low frequencies (1–4%). 5. We conclude that the contribution of hatched animals to the spring population dynamics is negligible in situations with an overwintering population present and will only become relevant if densities are extremely low. In this type of system, the significance of the resting egg bank is presumably mainly related to the maintenance of genetic diversity.  相似文献

Male sexual displays often include components detected across long distances, and those perceived only at close range. Understanding what information females gain from each component of a complex display and how they use these signals to make decisions are questions of major interest in sexual selection research. We evaluated content‐based hypotheses (‘redundant signals’ and ‘multiple messages’) for the courtship displays of field crickets (Gryllus integer) by measuring female responses to males' long‐distance calling song (calls) and close‐range chemical cues. Females' responses to a male's calls and chemical cues were uncorrelated, supporting the ‘multiple messages’ hypothesis. We also tested the ‘inter‐signal interaction’ hypothesis by investigating how long‐distance calls influence evaluation of close‐range courtship. The relationship between long‐ and close‐range signals was complex and conditional: females accepted close‐range courtship more quickly after exposure to attractive calling song than they did after exposure to either unattractive calling song or silence, and unattractive calls were no more or less effective than silence. This inter‐signal interaction could affect our understanding of mate choice in species with multiple mating signals because it implies that females may save time and energy by not assessing the close‐range signals of attractive long‐distance signalers. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2010, 100 , 856–865.  相似文献

  • 2 Field trials show that the scouring sampler is significantly more efficient at removing and retaining periphyton from stones in streams for biomass estimation than other commonly used techniques.
  • 3 The scouring sampler requires only one operator. Its small size and speed of use facilitate replication of samples. Samples can be collected from stones as small as 30mm diameter.
