ABSTRACT. Trophozoites of Entamoeba invadens IP-1 can be induced to encyst in simple solutions composed of semipermeable constituents (buffer, salts, or sugars) provided that their osmotic pressure is in the range of 60–160 mosmol/kg. Optimal yield of mature cysts was obtained when the osmotic pressure of the medium was 110 mosmol/kg. Encystation could be obtained in the absence of serum although higher yields were obtained in its presence. No difference in the yield of mature cysts was found when either dialyzed or full serum was used. High yields of encystation were obtained (>70%) in the presence of 5% serum in solutions of NaCl, KCl, or MgSO4, suggesting that the mechanism of encystation is not induced via sodium or potassium channels. Cysts were obtained in the presence of 72 mM glucose, indicating that depletion of a carbon source is not the only requirement for encystation. A rapid change in the density of the Entamoeba cells was observed upon transfer of trophozoites (density 1.061–1.073 g/ml) from growth medium to the low osmotic pressure encystation solutions. Within the first 2 min their density decreased (to 1.050 g/ml), but it soon increased, reaching within 30 min a density higher than 1.120 g/ml. As the encystation process continued to completion, the density of the cells gradually decreased, the mature cysts reaching a density of 1.049–1.061 g/ml.  相似文献

ABSTRACT. Spinal and cranial ganglia of American angler fish, Lophius americanus, are often infected with microsporidia. This protozoon elicits the formation of large, spore-filled, hypertrophied host cells, cysts. Previous reports of microsporidia in European lophiids identify the parasite as Spraguea lophii, a genus which has recently been shown to be dimorphic. The spores from L. americanus are monomorphic (2.8 × 1.5 μm) and uninucleate. Each spore contains a polar tube that forms six to nine coils. Spraguea lophii differs from the microsporidium described in L. americanus in several ways. Spraguea lophii has two spore types: a large spore (4.0 × 1.25 μm) containing a diplokaryon and three to four polar tube coils and a smaller uninucleate spore (3.5 × 1.5 μm) with five to six polar tube coils. Because of these major differences, the microsporidium from L. americanus is removed from the genus Spraguea and returned to its original genus, Glugea, as a new species, G. americanus n. sp. Other ultrastructural characteristics of G. americanus are included: the posterior vacuole encloses two distinct membranous structures; one is tubular and resembles a “glomerular tuft” and the second is lamellar and composed of concentric membrane whorls, additionally, the straight or manubroid portion of the polar tube proceeds beyond the posterior vacuole before it turns anteriorly and begins to coil.  相似文献

It has previously been proposed that sex is selected for in organisms living in antagonistic biotic environments. Here, the contrasting case of mutualistic biotic environments is considered. In these environments, it is argued that there should be selection against sex and an accompanying low rate of genetic change. This proposition is tested on mutualistic symbioses with substantial histories of co-evolution in which one partner, the inhabitant, lives partly or wholly inside the other, the exhabitant. This asymmetry between the partners means that inhabitants should exhibit a reduction in sex in comparison to related free-living taxa and a lower rate of genetic change than exhabitants.
The patterns that emerge from an analysis of 10 kinds of mutualistic symbiosis strongly support these predictions. Where adequate information is available, sex is usually reduced in inhabitants in comparison to related free-living taxa, although it remains widespread in exhabitants. Inhabitants are also represented by a much smaller taxonomic diversity than exhabitants. Various alternative reasons for these patterns are considered but it is concluded that they are best explained by the mutualistic environment in which the inhabitants live.  相似文献

This study demonstrates that Pleistophora schubergi Zwölfer, 1927, a microsporidium originally isolated from the midgut epithelium of Nygmia phaeorrhoea Don (Euproctis chrysorrhoea L.) and Porthetria dispar L., and subsequently reported in several other insects including the spruce budworm, Choristoneura fumiferana (the host used in this investigation), does not belong in the genus Pleistophora Gurley, 1893. Pleistophora schubergi lacks the major features that are characteristic of Pleistophora typicalis, the type species of this genus. A comparison of ultrastructural observations reported for the type species of the genus Pleistophora, P. typicalis, and our observations of P. schubergi revealed significant differences. A thick (0.5 μm) amorphous coat, derived from parasite secretions and deposited external to the parasite plasmalemma, surrounds all developmental stages in P. typicalis. Double membranes, derived from host rough endoplasmic reticulum cisternae encircle the parasite plasmalemma of all developmental stages in P. schubergi. The sporophorous vesicle encases the spores in P. typicalis, and originates from the parasite-secreted coat that is present around meronts. In P. schubergi, the host endoplasmic reticulum cisternae form the envelope that surrounds the meronts. Moreover, the sporophorous vesicle envelope in P. typicalis persists around groups of spores, while in P. schubergi this envelope breaks easily to release the spores in the host cytoplasm. By comparing the characteristics of the microsporidium found in the spruce budworm with those of the recently created polysporous genera that sporulate within a vesicle, we found that P. schubergi does belong in the new genus Endoreticulatus Brooks et al. 1988, and consequently rename it Endoreticulatus schubergi (Zwölfer, 1927) n. comb.  相似文献

Large, agglutinating protozoans belonging to the Foraminiferida(suborder Astrorhizina) and the Xenophyophorea are conspicuous,often dominant faunal elements in the deep sea. A review ofknown and suspected interactions between these forms and metazoansreveals a potentially significant role for the protozoans instructuring deep-sea metazoan assemblages. Direct interactionsinclude provision to metazoans of (a) hard or stable substratum,(b) refuge from predators or physical disturbance, and (c) accessto enhanced dietary resources. In some instances, rhizopod testsmay provide a nursery function. Xenophyophore modification offlow regimes, particle flux, bottom skin friction and sedimentcharacteristics appear likely and are believed to account foraltered composition and abundance of meiofauna and macrofaunain the vicinity of rhizopod tests. Some analogous interactionsare observed between metazoans and biogenic sediment structuresin shallow water. However, metazoan-rhizopod associations arehypothesized to be more highly developed and complex in thedeep sea than are comparable shallow-water associations, dueto rhizopod abilities to enhance scarce food resources and tolow rates of disturbance in much of the deep sea. Agglutinatingrhizopods appear to be a significant source of heterogeneityon the deep-sea floor and large tests often represent "hotspots"of metazoan activity. As such, they are hypothesized to havecontributed to the origin and maintenance of metazoan diversityin the deep sea by providing distinct microenvironments in whichspecies can specialize.  相似文献

A flow-through apparatus for measuring nitrogenase activityin field-grown crops is described. The apparatus was used tomeasure nitrogenase activity in a sainfoin crop throughout agrowth season. A marked seasonal variation was observed whichwas partly the consequence of defoliation. The acetylene-basedestimate of the nitrogen fixed annually by sainfoin was less(146 kg N ha–1) than the nitrogen accumulated in the shoots.Reasons for this difference are discussed with special emphasison the validity of the technique and alternative sources ofnitrogen. There were no treatment differences between the ratesof growth of either sainfoin or lucerne when grown with or withoutfertilizer nitrogen in the field. This result is in marked contrastwith results from a glasshouse experiment where sainfoin plantsutilizing nitrate-nitrogen exhibited faster growth rates thansimilar plants that were solely dependent on symbiotic nitrogenfixation. Climate warming is expected to shift bioclimatic zones and plant species distribution. Yet, few studies have explored whether seedling establishment is a possible bottleneck for future migration and population resilience. We test how warming affects the early stages of seedling establishment in 10 plant species in subarctic tundra. To zoom into the life phases where the effects of warming actually take place, we used a novel approach of breaking down the whole‐season warming effect into full factorial combination of early‐, mid‐, and late‐season warming periods. Seeds were sown in containers placed under field conditions in subarctic heath and were exposed to 3 °C elevation of surface temperature and 30% addition of summer precipitation relative to ambient. Heating was achieved with Free Air Temperature Increase systems. Whole‐season heating reduced germination and establishment, significantly in four out of 10 species. The whole‐season warming effect originated from additive effects of individual periods, although some of the periods had disproportionally stronger influence. Early‐germinating species were susceptible to warming; the critical phases were early summer for germination and mid summer for seedling survival. Graminoids, which emerged later, were less susceptible although some negative effects during late summer were observed. Some species with intermediate germination time were affected by all periods of warming. Addition of water generally could not mitigate the negative effects of whole‐season heating, but at individual species level both strengthening and amelioration of these negative effects were observed. We conclude that summer warming is likely to constrain seedling recruitment in open micro sites, which is a common seed regeneration niche in tundra ecosystem. Importantly, we described both significant temporal and species‐specific variation in the sensitivity of seedling establishment to warming which needs to be taken into consideration when modelling population dynamics and vegetation transitions in a warmer climate.  相似文献

The relationship between uptake by barley roots and translocationto shoots has been examined under different conditions for sixherbicides and a systemic fungicide (four triazines, diuron,2,4-dichlorophenoxyacetic acid and ethirimol). For all thesecompounds a large proportion of the material taken up by rootsdoes not appear to move readily to the shoots. Elution withwater of the roots of intact plants which had previously beenplaced in labelled solutions of these compounds enabled thematerial released to be separated into three fractions differingin their rates of diffusion out of the roots. There was reasonablygood correlation between the concentration of the most readilydiffusible fraction in the roots and the concentration in thetranspiration stream. The results obtained were consistent withthe postulate that lipophilic compounds can diffuse into thevacuoles of the cortical cells where they are available fortransport to the shoots, whereas for lipophobic compounds materialreaching the shoots originates largely from the free space inthe roots.  相似文献

This paper summarises our own accumulated experience from developing community-orientated mental health services in England and Italy over the last 20-30 years. From this we have provisionally concluded that the following issues are central to the development of balanced mental health services: a) services need to reflect the priorities of service users and carers; b) evidence supports the need for both hospital and community services; c) services need to be provided close to home; d) some services need to be mobile rather than static; e) interventions need to address both symptoms and disabilities; and f) treatment has to be specific to individual needs. In this paper we consider ten key challenges that often face those trying to develop community-based mental health services: a) dealing with anxiety and uncertainty; b) compensating for a possible lack of structure in community services; c) learning how to initiate new developments; d) managing opposition to change within the mental health system; e) responding to opposition from neighbours; f) negotiating financial obstacles; g) avoiding system rigidities; h) bridging boundaries and barriers; i) maintaining staff morale; and j) creating locally relevant ser- vices rather than seeking “the right answer” from elsewhere.  相似文献
