1. Bacteria can be an important resource for zooplankton production in aquatic food webs, although the degree to which bacteria sustain zooplankton growth and reproduction is not clear. We performed a growth experiment with Daphnia galeata feeding on different ratios of P‐replete Pseudomonas and Rhodomonas, ranging from a 100% bacterial to a 100% algal diet. 2. A pure bacterial diet did not support survival, growth or reproduction of D. galeata. While a 20% share of Rhodomonas in the food allowed survival of daphniids, the occurrence of offspring on a 50% algal diet indicated that the threshold for successful reproduction was between those two proportions of algal food. Increasing the proportion of the alga further increased growth and reproductive output, indicating that Rhodomonas was a higher‐quality food than Pseudomonas. 3. A subsequent labelling experiment demonstrated that D. galeata incorporated phosphorus from Pseudomonas and Rhodomonas with similar efficiency, whereas carbon was incorporated more efficiently from Pseudomonas than from Rhodomonas. 4. we hypothesise that inadequate levels of essential biochemicals in pure bacterial diets led to decreased Daphnia performance. Concentrations of fatty acids in general, and especially of polyunsaturated fatty acids, were much lower in Pseudomonas than in Rhodomonas. This difference could explain the different growth and reproduction responses, although limitation by other essential biochemicals (e.g. sterols) cannot be ruled out. 5. Hence, where they dominate, bacteria may provide a significant part of the elemental flux to species feeding higher in the food web on the short term. However, the performance of consumers may be constrained by essential biochemicals.  相似文献

Morphometrical univariate analyses of otter skulls collected over the past hundred years in European countries from presumed healthy populations were compared with skulls from presumed endangered populations. The average degree of sexual dimorphism of die European populations was found to be directly correlated to die skull size of die male otters. Fluctuating asymmetry (FA) in metric skull traits was analysed as an estimator of developmental stability. There was evidence for increased FA in different traits over time in some of the presumed endangered populations, and for a reduction in size of skull traits. In contrast, the healthy populations did not show any significant changes in the same traits during the same period. The reduced sexual dimorphism of the endangered populations is suggested to be a product of relaxed sexual selection and deteriorated habitat conditions. Environmental and genetic forces that may have shaped these patterns are discussed.  相似文献

Clones from a Sau 3A family of eight highly repetitive sequences previously isolated from a genomic DNA library of Alstroemeria ligtu ssp. ligtu were sequenced and found to be highly conserved. A trinucleotide microsatellite repeat [GCA]3?4 was present. A second, unrelated, Sau 3A repeat was also characterised. Southern analysis proved that the isolated repeats were specific for the A. ligtu subspecies and could not be detected in other Chilean or Brazilean Alstroemeria species. As shown by in situ hybridisation, the Sau 3A family and the unrelated Sau 3A repeat co‐localised at distinct sites along most chromosomes of Alstroemeria ligtu ssp. ligtu and Alstroemeria ligtu ssp. simsii. The present set of species‐specific repetitive sequences enables the identification of A. ligtu chromosomes, and thus the tracking of chromosome transmission to interspecific hybrids and their progeny.  相似文献

In photosynthetic organisms, excess light is a stress that induces production of reactive oxygen species inside the chloroplasts. As a response, the capacity of antioxidative defence mechanisms increases. However, when cells of Chlamydomonas reinhardtii were shifted from dark to high light, a reversible partial inactivation of catalase activity was observed, which correlated with a transient increase in the level of H2O2 in the 10 μm range. This concentration range seems to be necessary to activate H2O2‐dependent signalling pathways stimulating the expression of H2O2 responsive genes, such as the heat shock protein HSP22C. Catalase knock‐down mutants had lost the transient accumulation of H2O2, suggesting that a decrease in catalase activity was the key element for establishing a transient H2O2 burst. Catalase was inactivated by a one‐electron event consistent with the reduction of a single cysteine. We propose that under high light intensity, the redox state of the photosynthetic electron transport chain is sensed and transmitted to the cytosol to regulate the catalase activity. This allows a transient accumulation of H2O2, inducing a signalling event that is transmitted to the nucleus to modulate the expression of chloroplast‐directed protection enzymes.  相似文献

1. Growth defines the major life‐history traits such as size, weight, and age at maturity that determine an organism's fitness. Different models have been developed to describe growth by means of geometric progressions (e.g. Dyar's rule). However, growth forced along a geometric trajectory might constrain a plastic response to variable environmental conditions (e.g. food availability). 2. The present study investigated growth patterns under varying food conditions in the bridge spider, Larinioides sclopetarius, an extremely successful species in colonising urban habitats. 3. In L. sclopetarius growth ratios of successive instars were not constant but decreased over development. Instead, these spiders' growth is well described by a developmental growth rate (weight gain per moult) and a growth coefficient (weight gain per development time), both of which are based on a geometric progression. All developmental parameters, including developmental growth rate and growth coefficient as well as the intermoult duration and the number of instars, highly depend on food availability in L. sclopetarius and thus show plasticity. 4. Our study shows that geometric growth patterns do not necessarily preclude plasticity and that the parameters of geometric growth are affected by developmental plasticity. We suggest that their high developmental plasticity may facilitate bridge spiders' success in invading urban habitats.  相似文献

Fluorescence-labeled neuropeptide Y (NPY) has been used in flow cytometric binding assays for the determination of affinity constants of NPY Y1, Y2, and Y5 receptor ligands. Because the binding of fluorescent NPY is insufficient for competition studies at the human Y4 receptor (hY4R), we replaced Glu-4 in hPP with Lys for the derivatization with cyanine-5. Because cy5-[K4]hPP has high affinity (Kd 5.6 nM) to the hY4R, it was used as a probe in a flow cytometric binding assay. Specific binding of cy5-[K4]hPP to hY4R was visualized by confocal microscopy. The hY4R, the chimeric G protein Gqi5 and mitochondrially targeted apoaequorin were stably coexpressed in CHO cells. Aequorin luminescence was quantified in a microplate reader and by a CCD camera. By application of these methods 3-cyclohexyl-N-[(3-1H-imidazol-4-ylpropylamino)(imino)methyl]propanamide (UR-AK49) was discovered as the first nonpeptidic Y4R antagonist (pKi 4.17), a lead to be optimized in terms of potency and selectivity.  相似文献
