We report a rapid and efficient synthesis of A-ring modified 17α-picolyl and 17(E)-picolinylidene androstane derivatives from dehydroepiandrosterone. Compounds were validated spectroscopically and structurally characterized by X-ray crystallography. Virtual screening by molecular docking against clinical targets of steroidal anticancer drugs (ERα, AR, Aromatase and CYP17A1) suggests that 17(E)-picolinylidene, but not 17α-picolyl androstanes could specifically interact with CYP17A1 (17α-hydroxylase) with similar geometry and affinity as Abiraterone, a 17-pyridinyl androstane drug clinically used in the treatment of prostate cancer. In addition, several 17(E)-picolinylidene androstanes demonstrated selective antiproliferative activity against PC3 prostate cancer cells, which correlates with Abiraterone antiproliferative activity and predicted CYP17A1 binding affinities. Based on these preliminary results, 17(E)-picolinylidene androstane derivatives could be a promising starting point for the development of new compounds for the treatment of prostate cancer. 相似文献

Rhododendron shoot regeneration was accomplished using either flower explants (each consisting of ovary with pedicel) of Rhododendron cvs. Nova Zembla and Irina or leaves isolated from in vitro grown Rhododendron catawbiense Michx. Multiple shoot tip clumps were obtained on Anderson's medium containing 0.5 to 1.5 mg dm−3 thidiazuron (TDZ) in combination with 12 to 15 mg dm−3 N6-[2-isopentenyl]adenine (2iP) and 1 to 3 mg dm−3 indole-3-butyric acid (IBA). After 16 weeks on the regeneration media, explants with shoot tip clumps were transferred for
shoot elongation to Anderson's medium with 3 mg dm−3 2iP. Two months later, the shoots have reached 5 to 40 mm in length and were fit for subcultivation.

An Agrobacterium rhizogenes-mediated transformation system for Aesculus hippocastanum L. has been developed. Wounded androgenic embryos of A. hippocastanum were inoculated with bacteria containing the pRiA4 plasmid, with the uid A sequence as a reporter gene. The hairy roots emerging from the wounded sites of androgenic embryos were isolated and maintained in Murashige and Skoog's (MS) liquid hormone-free medium. Five hairy root lines have been maintained in vitro for 4 years with unchanged growth rate and might be a suitable source for secondary metabolite production. The transformation events have been confirmed by a polymerase chain reaction specific to the rol A, B, C and D genes. The absence of residual contaminating bacteria has been shown by a polymerase chain reaction specific to the vir D1 sequence. 相似文献

Stipa crassiculmis subsp. picentina Martinovský, Moraldo & Caputo (Poaceae, Pooideae, Stipae) was found for the first time in the Balkan Peninsula. It was recorded in the northern part of the Republic of Macedonia, in the P?inja River gorge (Badar Gorge), between Veles and Katlanovo on ultramafic substrates. To date, S. crassiculmis subsp. picentina was known only for Italy where it occurs in the Lombardy and Campania administrative regions. This record for the Balkan Peninsula is a new example of Amphi-Adriatic Balkan-Apennine disjunction. In this paper, the morphological and anatomical characters of the specimens collected in the study area were reported together with the phytosociological characteristics of the habitat. The threatened status of the species for Macedonia was also assessed on the basis of the IUCN criteria. 相似文献

Epigenetic modifications may play an important role in invasion and adaptation of clonal and invasive populations to different environments. The aim of this study was to analyse epigenetic diversity and structure within and among populations of invasive pathogenic fungus Cryphonectria parasitica from south-eastern Europe, where one haplotype S12 dominates. The highest level of epigenetic diversity was found in haplotype S1, followed by S2, while the lowest level of epigenetic diversity was found in haplotype S12. Similar pattern of epigenetic diversity was detected in the control, genetically diverse Croatian population where S1 haplotype dominates. In four south-eastern European populations, the highest level of epigenetic diversity was observed in the Italian population, the oldest population in the studied area, while the lowest diversity was found in most recently established Bulgarian population. This relationship between epigenetic diversity and population age implies the important role of epigenetic modifications on the process of invasion. Our data suggest that epigenetic differences might affect the success of expansion of certain haplotype into new regions. Understanding the role of epigenetic processes in expansion and (pre)adaptation of fungal plant pathogens, besides fundamental knowledge, can contribute to development of strategies for control of fungal spread and pathogenesis. 相似文献
