Germinal centres (GCs) are specialised lymphoid microenvironments that form in secondary B-cell follicles upon exposure to T-dependent antigens. In the GC, clonal expansion, selection and differentiation of GC B cells result in the production of high-affinity plasma cells and memory B cells that provide protection against subsequent infection. The GC is carefully regulated to fulfil its critical role in defence against infection and to ensure that immunological tolerance is not broken in the process. The GC response can be controlled by a number of mechanisms, one of which is by forkhead box p3 expressing regulatory T (Treg) cells, a suppressive population of CD4+ T cells. A specialised subset of Treg cells – follicular regulatory T (Tfr) cells – form after immunisation and are able to access the GC, where they control the size and output of the response. Our knowledge of Treg cell control of the GC is expanding. In this review we will discuss recent advances in the field, with a particular emphasis on the differentiation and function of Tfr cells in the GC.  相似文献

1. As long‐term observational lake records continue to lengthen, the historical overlap with lake sediment records grows, providing increasing opportunities for placing the contemporary ecological status of lakes in a temporal perspective. 2. Comparisons between long‐term data sets and sediment records, however, require lake sediments to be accurately dated and for sediment accumulation rates to be sufficiently rapid to allow precise matching with observational data. 3. The critical role of the sediment record in this context is its value in tracking the changing impact of human activity on a lake from a pre‐disturbance reference through to the present day. 4. Here, we use data from a range of lakes across Europe presented as case studies in this Special Section. The seven sites considered all possess both long‐term observational records and high‐quality sediment records. Our objective is to assess whether recent climate change is having an impact on their trophic status and in particular whether that impact can be disentangled from the changes associated with nutrient pollution. 5. The palaeo‐data show clear evidence for the beginning of nutrient pollution varying from the mid‐nineteenth century at Loch Leven to the early and middle twentieth century at other sites. The monitoring data show different degrees of recovery when judged against the palaeo‐reference. 6. The reason for limited recovery is attributed to continuing high nutrient concentrations related to an increase in diffuse nutrient loading or to internal P recycling, but there is some evidence that climate change may be playing a role in offsetting recovery at some sites. If this is the case, then lake ecosystems suffering from eutrophication may not necessarily return to their pre‐eutrophication reference status despite the measures that have been taken to reduce external nutrient loading. 7. The extent to which future warming might further limit such recovery can be evaluated only by continued monitoring combined with the use of palaeo‐records that set the pre‐eutrophication reference.  相似文献

Measurements of whole-organism performance traits have been useful in studies of adaptation and phenotype–environment correlations. Bite force capacities may be tightly linked to both the type and magnitude of the ecological challenges of food acquisition, mate acquisition, and antipredation in vertebrates. In the present study, we present technical details on bite meters and on measuring bite forces. The ability to take reliable measurements depends on specific features of the measuring device and on where in the mouth the bite is applied. Using both previously available and original data, we demonstrate several ecologically and evolutionarily relevant features of bite force measurements. First, maximal bite forces are repeatable among individuals across all vertebrates studied to date. Second, in ectotherms such as lizards, maximal bite forces are affected by body temperature and motivational states. Third, bite forces are strongly correlated with head size and shape. Fourth, bite forces correlate with features of prey of vertebrates. Finally, bite forces are linked to male dominance and correlated with social-display structures. Thus, bite force performance measures can be used as 'traits', and thus be used in integrative studies at multiple levels of organismal biology. Accordingly, bite force data will help our understanding of the functions, capacities, and evolution of jaw–cranial musculoskeletal systems. Moreover, a plethora of opportunities exist for the use of bite force measurements, and if methods are carefully applied, several levels of organismal and ecological organization can be integrated to aid our understanding of the ecology and evolution of vertebrate taxa.  © 2008 The Linnean Society of London, Biological Journal of the Linnean Society , 2008, 93 , 709–720.  相似文献

1. Surface sediment samples of subfossil chironomid head capsules from 47 lakes in southern West Greenland were analysed using multivariate numerical methods in order to explore the relationship between chironomid assemblages and selected environmental variables. The study lakes are located along a climate gradient ranging from coastal maritime conditions near the Davis Strait to a continental climate near the margin of the Greenland ice sheet. 2. High‐resolution surface water temperatures were measured through the summer season using automatic data loggers in 21 of the study lakes. The mean July surface water temperature (1999) ranged from 7.3 to 16.5 °C in the data set. 3. In all lakes, a total of 24 chironomid taxa were recorded; Micropsectra, Psectrocladius, Chironomus and Procladius were the dominant genera. There was a strong correlation between the trophic variables [total nitrogen and total phosphorus (TN, TP)] and temperature, and in redundancy analysis (RDA) the three variables explained almost equal significant amounts of variation in the chironomid data (19.8–22.3%). However, temperature lost significant explanatory power when the effect of TN was partialled out in RDA. 4. The lakes were classified using two‐way indicator species analysis (TWINSPAN ) into eight groups defined by temperature, trophic variables, salinity (conductivity) and lake‐morphometric data. Fourteen chironomid taxa showed significant differences in percentage abundances among groups, with Heterotrissocladius, Micropsectra, Ablabesmyia and Chironomus as the most robust group‐indicator taxa. Forward selection of taxa in multiple discriminant analysis was used to fit chironomid assemblages into lake groups. Using only eight taxa, 95% of lakes were correctly classified at a second TWINSPAN division level (four groups) and 85% of lakes at a third division level (eight groups). 5. This study showed that there is considerable potential in using subfossil chironomid head capsules as paleoenvironmental indicators in both short‐ and long‐term (down‐core) studies of lake ontogeny and palaeoclimate conditions in West Greenland. However, because of the strong correlation between temperature and trophic variables, a quantitative reconstruction of lake‐ and habitat‐type is recommended, in combination with direct reconstruction of single variables such as temperature.  相似文献

Soya Bean Seed Growth and Maturation In vitro without Pods   总被引:2,自引:0,他引:2  
Immature Glycine max (L.) Merrill seeds, initially between 50and 450 mg f. wt, were grown and matured successfully in vitro.Excised seeds were floated in a liquid medium containing 5 percent sucrose, minerals and glutamine in flasks incubated at25 °C under 300 to 350 µE m–2 s–1 fluorescentlight. During 16 to 21 d in culture, seeds grew to a matured. wt of 100 to 600 mg per seed at an average rate of 5 to 25mg d. wt per seed d–1 depending on initial size. Growthrates were maximal during the first 8 to 10 d in vitro but declinedwith loss of green colour in the cotyledons. Seed coats rupturedwith rapid cotyledon expansion during the first 2 d in culture.Embryos were tolerant to desiccation and 80 to 90 per cent germinatedif removed from culture before complete loss of green colour.The growth of excised seeds in vitro exceeded the growth ofseeds in detached pods, but when windows were cut in pods topermit direct exposure of seeds to the medium, seed growth wascomparable. Yellow-brown, algal symbionts varying in diameter from approximately 5 μ m to 20 μ m, associated with solitary Radiolaria with spongiose skeletons (i.e. Spongodrymus sp.), exhibit fine structural features resembling the Prymnesiida (botanical class, Prymnesiophyceae). A large central vacuole is surrounded by a thin layer of cytoplasm containing plastids with lamellae composed of three thylakoids and granular pyrenoids with internal tubules immersed between the thylakoids. The pyrenoids lack internal thylakoid membranes. The nucleus is surrounded by a dilated cisterna of the nuclear envelope that also encloses the plastids and gives rise to saccules of the endoplasmic reticulum. The algal symbionts appear coccoid; hence no flagella nor surface scales were observed. The symbiont fine structure is compared to similar yellow-brown symbionts associated with Acantharia. Thus far, three kinds of algal symbionts have been observed to be associated with solitary Radiolaria: dinoflagellate, prasinomonad, and this apparent prymnesiomonad.  相似文献

A sediment core from the lower Mulgrave River flood plain in north-east Queensland was examined using standard pollen analysis techniques to investigate vegetation and sea level changes during and since the Holocene marine transgression. Saltwater penetrated the Mulgrave River to the core site in the early Holocene, with the consequent development of riverside mangrove forests dominated by Sonneratia lanceolata, a community not previously recorded in Australian pollen sequences. From about 7000 to 6000 years ago, Rhizophora forest was the dominant vegetation community around the site. During this period, vertical sedimentation kept pace with, or at times exceeded, the rate of sea level rise. As sea level stabilized, about 6000 years ago, continued sediment accumulation led to the development of upper-intertidal Ceriops/Bruguiera forest and then freshwater swamp forest. The vegetation changes in this latter part of the sequence reflect the present seaward to landward vegetation zonation in the area.  相似文献
