The aim of this study was to assess genome size variation and multivariate morphometric analyses to ascertain cytotype distribution patterns and the morphological differentiation within the Ranunculus parnassifolius group in the Pyrenees and the Alps. Although divergences in nuclear DNA content among different species within a genus are widely acknowledged, intraspecific variation is still a somewhat controversial issue. Holoploid and monoploid genome sizes (C‐ and Cx‐values) were determined using propidium iodide flow cytometry in 125 plants of R. parnassifolius s.l. distributed across four European countries. Three different DNA ploidy levels were revealed in the study area: diploid (2n ~ 2x, 57.14%), triploid (2n ~ 3x, 1.19%), and tetraploid (2n ~ 4x, 41.67%). The mean population 2C‐values ranged from 8.15 pg in diploids to 14.80 pg in tetraploids, representing a ratio of 1 : 1.8. Marked intraspecific/interpopulation differences in nuclear DNA content were found. Diploid populations prevail in the Pyrenees, although tetraploid cytotypes were reported throughout the distribution area. In general, mixed‐cytotype populations were not found. The Spearman correlation coefficient did not reveal significant correlations between genome size and altitude, longitude, or latitude. Morphometric analyses and cluster analyses based on genome size variation revealed the presence of three major groups, which exhibited a particular biogeographical pattern. A new cytotype, DNA triploid, was found for the first time. Tetraploid populations showed constant nuclear DNA levels, whereas diploid populations from the Pyrenees, in which introgressive hybridization is suggested as a presumable trigger for genome size variation, did not. Scenarios for the evolution of geographical parthenogenesis in R. parnassifolius s.l. are discussed. The evolutionary potential in the timing of recruitment and reproduction may be crucial for the ability of populations to buffer against environmental changes, allowing them to avoid unfavourable breeding conditions. The evolution of a trait in a local population is determined by its heritability and selection. In the present study, we performed pedigree‐based quantitative genetic analyses for two life‐history traits (recruiting age and laying date) using population data of the storm petrel over an 18‐year period in two adjacent breeding colonies (only 150 m apart) that share the same environmental conditions. In both traits, natal colony effect was the main source of the phenotypic variation among individuals, and cohort variance for recruitment age and additive genetic variance for laying date were natal colony‐specific. We found significant heritability only in laying date and, more specifically, only in birds born in one of the colonies. The difference in genetic variance between the colonies was statistically significant. Interestingly, selection on earlier breeding birds was detected only in the colony in which heritable variation in laying date was found. Therefore, local evolvability for a life‐history trait may vary within a unexpectedly small spatial scale, through the diversifying natural selection and insulating gene flow. © 2012 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, 106 , 439–446. 相似文献

This article documents the addition of 83 microsatellite marker loci and 96 pairs of single‐nucleotide polymorphism (SNP) sequencing primers to the Molecular Ecology Resources Database. Loci were developed for the following species: Bembidion lampros, Inimicus japonicus, Lymnaea stagnalis, Panopea abbreviata, Pentadesma butyracea, Sycoscapter hirticola and Thanatephorus cucumeris (anamorph: Rhizoctonia solani). These loci were cross‐tested on the following species: Pentadesma grandifolia and Pentadesma reyndersii. A combination of founder effects and local adaptation – the Monopolization hypothesis – has been proposed to reconcile the strong population differentiation of zooplankton dwelling in ponds and lakes and their high dispersal abilities. The role genetic drift plays in genetic differentiation of zooplankton is well documented, but the impact of natural selection has received less attention. Here, we compare differentiation in neutral genetic markers (FST) and in quantitative traits (QST) in six natural populations of the rotifer Brachionus plicatilis to assess the importance of natural selection in explaining genetic differentiation of life‐history traits. Five life‐history traits were measured in four temperature × salinity combinations in common‐garden experiments. Population differentiation for neutral genetic markers – 11 microsatellite loci – was very high (FST = 0.482). Differentiation in life‐history traits was higher in traits related to sexual reproduction than in those related to asexual reproduction. QST values for diapausing egg production (a trait related to sexual reproduction) were higher than their corresponding FST in some pairs of populations. Our results indicate the importance of divergent natural selection in these populations and suggest local adaptation to the unpredictability of B. plicatilis habitats. 相似文献

Scophthalmus maximus is an important commercially aquaculture fish species. We tackle the search for new microsatellites using two different approaches: an enriched partial genomic library and a screening of all turbot DNA sequences deposited in GenBank. Out of 15 genomic library derived loci, five gave working primer pairs, with expected heterozygosities (HE) ranging from 0.13 to 0.91. Out of seven loci derived from database sequences, two amplified successfully with an HE ranging from 0.56 to 0.63. The average allele number of these microsatellites was 5.7 per locus with a range between two and 15. 相似文献

The braconid subfamily Doryctinae (Hymenoptera) is a cosmopolitan, species‐rich group of parasitoid wasps whose known species richness and morphological diversity is mainly concentrated in the Neotropical region. Among the doryctine taxa that are endemic to this region, there is a group of six genera (the Pedinotus genus group) that are mainly characterized by having lateral, posteriorly converging or subparallel grooves on the second metasomal tergite (including Gymnobracon Szépligeti, Hybodoryctes Szépligeti, Lamquetia Braet et van Achterberg, Osmophila Szépligeti, Pedinotus Szépligeti and Trigonophasmus Enderlein). Most of these genera were described more than a century ago, and thus their limits and diagnostic morphological features are not well defined. We present an analysis of the phylogenetic relationships among various representative species of five of the six genera belonging to the Pedinotus genus group based on one mitochondrial (cytochrome oxidase I) and two nuclear (28S, Wingless) gene markers. We also estimated the timing of origin and diversification within the group and investigated the external morphology of the genera involved in order to determine reliable identification characters. The group as a whole, whilst being recovered as monophyletic with the inclusion of Acanthorhogas Szépligeti, was not statistically supported. However, the respective monophyly of all of the morphologically supported genera are confirmed, as are the intergeneric relationships (Gymnobracon, Trigonophasmus) ((Acanthorhogas, Osmophila) (Lamquetia, Pedinotus)). We propose that the pattern of sublateral grooves or depressions appears to be a useful systematic character for recognition of an endemic Neotropical radiation that could have originated c. 41–27 Ma. We redescribe six of the seven genera that we recognize for the Pedinotus genus group and provide a key to enable their identification. 相似文献

Deserts shrubs are well known to facilitate vegetation aggregation, mostly through seed trapping, and stress amelioration during and after plant establishment. Because vegetation aggregation effects are a by‐product of shrub presence, beneficiary species may not only be native, but also exotic. However, despite the high risk that exotic invasive species pose to ecosystem services, little is known of the role of desert shrubs on plant invasions. We assessed the influence of two shrub species on the non‐dormant soil seed bank (i.e. the number of seeds that readily germinate with sufficient water availability) of an invasive annual grass (Schismus barbatus) and of coexisting native species in a central‐northern Monte Desert (Argentina). Soil samples were collected beneath the canopies of two dominant shrub species (Bulnesia retama and Larrea divaricata) and in open spaces (i.e. intercanopies) in May 2001. Overall, the density of germinated seedlings of Schismus and that of the native species were negatively associated across microsite types. Schismus density was similar to that of all native species pooled together (mostly annuals), and was highest in Larrea samples (with no significant differences between Bulnesia and intercanopies). On the contrary, the density of all native species pooled together was highest in Bulnesia samples. Our results suggest that shrubs may contribute to plant invasions in our study system but, most importantly, they further illustrate that this influence can be species specific. Further research is needed to assess the relative importance of in situ seed production (and survival) and seed redistribution on soil seed bank spatial patterns. 相似文献

Mitotic activity does not stop for different meristematic cells of the root apex at the same distance from the initials. The differences are connected with the functional heterogeneity of the apical meristem of the root. The arrangement of vascular bundles,i.e. the alternation of independent xylem and phloem groups, is of major importance. In broad bean roots, the protophloem sieve elements stop dividing first. The centre of the stelei. e. late metaxylem elements stop dividing next. Division in the stele gradually ceases centrifugally, while it ceases centripetally in the peripheral part of the root. The cylindrical region with prolonged cell division includes internal layers of the cortex including endodermis, pericycle and adjoining cells of the stele. Proximally apical meristem is reduced to isolated strands of cells adjacent to the protoxylem poles. Pericycle cells stop dividing last at a distance of approx. 9–10 mm from the initials. The number of the division cycles is limited and is specific for individual cell types. Epidermal and cortical cells divide in broad bean roots transversely approximately seven times, cells of late metaxylem approximately five times. Root apical meristem is an asynchronous cell population with a different duration of the mitotic cycle. We determined local variations in the duration of the mitotic cycle in the apical meristem of broad bean root by means of colchicine-induced polyploidy. The cells of the quiescent centre had the longest mitotic cycle after colchicine treatment. The region of the proper root adjacent to the quiescent centre was mixoploid (2n and 4n). Isolated cells with a long cycle occurred also in the cortex and in the central cylinder. Cells with a division cycle of 18h were found in the root cap, in the epidermis, in the cortex and in the central cylinder. Relatively numerous cells with the shortest division cycle, approx. 12 h, occurred farther of the quiescent centre in the epidermis, in the cortex, in the pericycle, and in adjacent layers of the stele through-out the entire meristematic region. The results derived from the analysis of the apical meristem are discussed in connection with the ontogenesis of different types of cells taking part in the primary structure of the root. 相似文献

TRANSFORMATION of allogenic lymphocytes in mixed cultures depends chiefly on an incompatibility between the lymphocyte donors at the major histocompatibility locus in man (HL-A), mouse (H-2) and rat (H-l)1. Although the mouse H-2 locus can be divided into several regions each of which controls one or more antigenic specificities2 and two or more subloci control HL-A antigens in man3, it is not known whether all parts of the major histocompatibility locus are equally important in eliciting transformation in mixed lymphocyte cultures. Intraspecific variation in morphology has often been related to fitness differences through its effects on performance. In lizards, variation in hind limb length can be shaped by natural selection for increased locomotor performance, sexual selection on the number or size of femoral pores involved in chemical signalling, or both. Here, we analyse the selective forces involved in sexual dimorphism and differences in hind limb length between two populations of Psammodromus algirus living at different elevation. Males were more robust and had longer hind limbs and limb segments than females, and low‐elevation lizards had longer limbs than high‐elevation lizards. However, differences in locomotor performance were small and non‐significant, making natural selection for faster runs an unlikely explanation for the observed pattern. On the other hand, males had more femoral pores than females, and lizards had more pores at lower elevation, although the difference was significant only for males (which invest more in chemical signalling). In males, the number of pores, which remains constant along a lizard's life, was not correlated with hind limb length. However, femur length was positively correlated with mean pore size, allowing low‐elevation males to have larger than expected pores, which could increase the effectiveness with which they spread their signals in a dry and warm habitat where chemicals become volatile rapidly. Also, saturation of the sexual coloration of the head was higher for low‐elevation males, suggesting that sexual selection pressures may be more intense.
