A combined cytochemical and electronmicroscopic study of feeding Tokophrya revealed that it has 2 sources of acid phosphatase. One is from the prey, Tetrahymena, supplying newly formed food vacuoles with large amounts of enzyme. The other source is in Tokophrya itself, the enzyme being found in small vesicles, small dense elongate bodies surrounded by a membrane, or in residue vacuoles. It seems that the 2 former small structures contain insignificantly small amounts of phosphatase; however, large deposits of lead phosphate are present in residue vacuoles, former food vacuoles. Since Tokophrya has no cytopyge these vacuoles are not excreted. On the contrary, when feeding is resumed, they merge with food vacuoles, presumably supplying them with acid phosphatase. Whether this enzyme ultimately is derived from the prey Tetrahymena and persists undegraded in the residue vacuoles, or whether it is synthesized by Tokophrya cannot be determined from present work.  相似文献

The effect of water stress in three Quercus species belongingto different habitats is investigated. Chlorophyll concentrationdecreased in all Quercus species with the greatest decreaseoccurring in Quercus robur. An increase of total carotenoidcontent was observed in Quercus coccifera and Quercus ilex,while in Q. robur water stress effected a carotenoid contentreduction. Anthocyanins increased with decreasing relative watercontent in the three Quercus species, the largest increase occurringin Q. coccifera. A correlation between xerophytic characterand pigment concentration is discussed.  相似文献

SYNOPSIS. Most of the known mutants of the axolotl were uncoveredby R. R. Humphrey through inbreeding laboratory stock or wildanimals imported from Mexico. All are spontaneous in origin.We believe that chemical mutagenesis may be useful for increasingboth the frequency and variety of new mutations. For our experiments,we have chosen the alkylating agent ethyl methanesulfonate (EMS),because its mode of action is reasonably well understood. Thoughwe were unable to establish a lethal dose of EMS by intraperitonealinjection, we were successful when we administered the compounddirectly in the water. In the weeks immediately after treatment,females administered sublethal doses did not ovulate, exceptin response to large injections of follicle stimulating hormone.Recovery took several months. Treated males showed more normalperformance, in terms of courtship and the number of spermatophoresproduced. At low doses, and short intervals after treatment,embryos from spawnings with treated males survived nearly aswell as controls. Survival reached a minimum about a month afterthe male had been treated, then improved again. This depressionand recovery may be correlated with the stage of sperm maturationat the time of treatment. Treatment of mature sperm from spermatophoresaffected only their ability to fertilize eggs in an artificialinsemination; fertile eggs developed normally. The most promisingprocedure would, therefore, seem to be treatment of the male.Following mutagenesis, we propose that gynogenesis be used toaid in the rapid identification of new recessive mutations.  相似文献

Crithidia deanei from the reduviid hemipteron, Zelus leucogrammus, unlike most lower trypanosomatids cultivated in defined medium, required only 2 amino acids, methionine and tyrosine; only 4 vitamins, folic acid, thiamine, biotin, and nicotinamide; and neither hemin nor a purine source. Electron microscopy reveals an endosymbiont, probably bacterial, which presumably provides the other basic trypanosomatid essential nutrients.  相似文献

Age was determined in three anuran species inhabiting a temperate oceanic climate ( Rana temporaria, Bufo calamita and Bufo bufo ) by means of the histological structure of the phalanges. Narrow, haematoxylinophilic lines were visible in these, and evidence from the examination of subadults and frog ( R. temporaria ) recaptures showed the lines to be annual. The validity of using the lines as age indicators was tested most extensively on natural frog populations and the effects of endosteal resorption were not found to be significant. The evidence for this conclusion was provided directly by recapture data and indirectly by an analysis of histological measurements made on the phalanges.  相似文献

Pigtailed macaques (Macaca nemestrina) provide an important model for biomedical research on human disease and for studying the evolution of primate behavior. The genetic structure of captive populations of pigtailed macaques is not as well described as that of captive rhesus (M. mulatta) or cynomolgus (M. fascicularis) macaques. The Washington National Primate Research Center houses the largest captive colony of pigtailed macaques located in several different housing facilities. Based on genotypes of 18 microsatellite (short tandem repeat [STR]) loci, these pigtailed macaques are more genetically diverse than captive rhesus macaques and exhibit relatively low levels of inbreeding. Colony genetic management facilitates the maintenance of genetic variability without compromising production goals of a breeding facility. The periodic introduction of new founders from specific sources to separate housing facilities at different times influenced the colony's genetic structure over time and space markedly but did not alter its genetic diversity significantly. Changes in genetic structure over time were predominantly due to the inclusion of animals from the Yerkes National Primate Research Center in the original colony and after 2005. Strategies to equalize founder representation in the colony have maximized the representation of the founders’ genomes in the extant population. Were exchange of animals among the facilities increased, further differentiation could be avoided. The use of highly differentiated animals may confound interpretations of phenotypic differences due to the inflation of the genetic contribution to phenotypic variance of heritable traits. Am. J. In two heavily infested rural villages of Santiago del Estero, Argentina, where no indoor-spraying with residual insecticides had ever been carried out by official control services, we studied the influence of roof and wall structure, domestic use of insecticide, family size and the number of domestic dogs, on the domiciliary density of Triatoma infestans (Klug). Bug density was significantly associated with (1) the interaction between insecticide use and type of roof, (2) the structure of indoor walls, (3) the number of dogs sharing sleeping areas of people (room-mate dogs), and (4) the number of people plus room-mate dogs, but not with just the number of people resident in the house. The interaction between insecticide use and a roof made of 'simbol', a locally available grass (Pennisetum sp.), also reflected a younger age structure of domestic bug populations. In infested houses, the density of bugs infected with Trypanosoma cruzi Chagas was significantly correlated with overall bug density. Our data suggest that the application of environmental management measures by the affected people, such as plastering of walls and modification of roofs, coupled with keeping dogs away from bedrooms and application of insecticides, should limit the domestic population density of T. infestans and thus reduce the transmission of T. cruzi to people.  相似文献

Abstract Species of didelphid marsupials (Didelphimorphia, Didelphidae) differ in their use of the forest strata, but it is not clear whether these differences are in fundamental or realized niches. The fundamental niche of seven species of didelphids (Caluromys philander, Didelphis aurita, Gracilinanus microtarsus, Marmosops incanus, Metachirus nudicaudatus, Micoureus demerarae, and Philander frenatus) was compared using their performance in arboreal walking. The association between performance and vertical use of the forest also was tested accounting for phylogenetic and allometric effects. Tests consisted of making the animal cross five 3 m long horizontal supports of different diameters, 1 m from the ground. The cycle of maximum speed was chosen to measure stride length, frequency and velocity. Arboreal species performed better than the terrestrial ones, but a major part of the variation in stride length (70.95%) and stride frequency (88.10%) was associated with body size. Part of the variation in stride length independent of body size (14.05%) was associated with the degree of vertical use of the forest, after phylogenetic effects were accounted for. Fundamental niches of six of the seven species were discriminated with the performance tests used. Discrepancies between the realized and fundamental niches can be inferred for two of these species, D. aurita and P. frenatus.  相似文献

Transaminase, amylase and protease activities were comparedin seedlings of normal and Opaque-2 (o2) maize. Transaminaseactivity, greater in normal maize, was highest in the scutellumfrom which it decreased rapidly in activity from day 2 afterimbibition; only low activity was observed in endosperm andaxis tissue. Amylolytic activity, optimal around pH 5, was greater in normalmaize at all stages of endosperm degradation. Activity whichwas low on day 2, rose to a peak on day 6 and declined afterwards.The level of free sugars in the endosperm of normal was higherthan in o2 maize, and in both varieties was highly correlatedwith amylolysis. Protease activity, optimal at pH 3.6, was also greater in normalendosperms and increased up to day 6 and activity was maintainedat this level until around day 14. Although the activity ofall three enzyme systems examined was greater in normal maizethere were no apparent differences in the overall growth ofnormal and o2 seedlings during this period.
