Primary hyperoxaluria type I (PH1) is an autosomal recessive metabolic disorder caused by inherited mutations in the AGXT gene encoding liver peroxisomal alanine : glyoxylate aminotransferase (AGT) which is deficient or mistargeted to mitochondria. PH1 shows considerable phenotypic and genotypic heterogeneity. The incidence and severity of PH1 varies in different geographic regions. DNA samples of the affected members from two unrelated Tunisian families were tested by amplifying and sequencing each of the AGXT exons and intron–exon junctions. We identified a novel frameshift mutation in the AGXT gene, the c.406_410dupACTGC resulting in a truncated protein (p.Gln137Hisfs*19). It is found in homozygous state in two nonconsanguineous unrelated families from Tunisia. These molecular findings provide genotype/phenotype correlations in the intrafamilial phenotypic and permit accurate carrier detection, and prenatal diagnosis. The novel p.Gln137Hisfs*19 mutation detected in our study extend the spectrum of known AGXT gene mutations in Tunisia. 相似文献

Inoculation of seed potato tubes with Verticillium nubilum produced infection on the underground stem bases to subsequent growth in the form of brown lesions, often accompanied by longitudinal or transverse cracking. Some inoculation tests provided further evidence of a causal relationship between infection with V. nubilum and coiled sprout, but it was not possible to define the conditions in which the fungus gave rise to coiling. Seed tubers sprouted in light before planting produced more coiling than unsprouted or desprouted tubers. Sprouting had this effect whether V. numbilum was present or not and coiling induced by sprouting and that induced by V. nubilum occurred independently. The increased coiling of stem bases from sprouted tubers was often accompanied by fasciation, whereas this symptom was not usually associated with the coiling from unsprouted or desprouted seed tubers inoculated with the fungus. Brown lesions associated with V. nubilum were readily distinguishable from those of Rhizoctonia solani but not from those caused by Oospora pustulans. Neither of these fungi was found to cause coiling. Different varieties of potatoes showed differences in the incidence of coiling in response to sprouting treatment. Bread-making quality in hexaploid wheats is a complex trait. It has been shown that the amount and composition of protein can influence dough rheological properties. The high-molecular-weight (HMW) glutenins are encoded by a complex locus, Glu-1, on the long arm of group-1 homoeologus chromosome of the A, B and D genomes. In this work we used PCR-based DNA markers as a substitution tool to distinguish wheat bread-making quality. We detected PCR-based DNA markers for coding sequence of Glu-A1x, Glu-B1x and Glu-D1x to be 2300 bp, 2400 bp and 2500 bp respectively. DNA markers related to coding sequence of Glu-A1y, Glu-B1y and Glu-D1y were; 1800 bp, 2100 bp and 1950 bp, however, the repetitive region of their coding sequence were shown to be about 1300 bp, 1500 bp and 1600 bp. The results demonstrate that the size variation was due to different lengths of the central repetitive domain. Good or poor bread-making quality in wheat is associated with two allelic pairs of Glu-D1, designated 1Dx5-1Dy10 and 1Dx2-1Dy12. The 1Bx7 allele has moderate-to-good quality score. The specific DNA markers, of 450 bp, 576 bp, 612 bp and 2400 bp respectively were characterized for 1Dx5, 1Dy10, 1Dy12 and 1Bx7 alleles. Abstract: Among the new dental remains from the late Early Eocene of Chambi (Kasserine area, Tunisia) is a large‐sized upper molar of a new bat species, Witwatia sigei nov. sp. (Chiroptera, Vespertilionoidea, Philisidae), described herein. The locality of Chambi has revealed evidence for an early appearance of two modern microchiropteran superfamilies in Africa: Dizzya exsultans, a Philisidae, which is considered to be an archaic Vespertilionoidea, and an indeterminate Rhinolophoidea. In addition to D. exsultans, the new species, W. sigei, is the second representative of the Philisidae in this locality. W. sigei extends back to the late Early Eocene the occurrence of the genus Witwatia, which was previously only reported from the early Late Eocene of the Fayum (BQ‐2, Egypt). By analogy with the largest extant microbats, the large size of Witwatia suggests a tendency to the opportunistic diet of this taxon, thereby contrasting with the strict insectivory characterizing primitive bats found in other continents in the same epoch. 相似文献

Maternal-environmental effects on subsequent progeny life-history traits were evaluated in squash ( Cucurbita pepo L.) in terms of the amount of time available for seed development, and the timing of fruit production. Progeny arising from three kinds of fruit were compared. Plants from which fruits were removed 3 d post-pollination throughout the growing season developed only 'late' fruits (during 10–15 d) at the end of the growing season; on control plants both 'early' and 'late' fruits developed (both types allowed to ripen fully). Seed from each type of fruit was weighed individually and categorized into three size classes, then germinated and raised to maturity, including regular harvesting of all fruits 3 d post-pollination. Maternal effects were evident for both vegetative and reproductive traits and carried over to later stages. In contrast, effects due simply to seed size disappeared by day 30 for leaf variables and day 60 for male flower production. Within a seed-size class, progeny arising from fruit of treated plants produced significantly more leaves, with greater size, and more male flowers than those arising from fruit of control plants, while the reverse was true for fruit number and fruit mass. This result is discussed in terms of possible gibberellic acid involvement. In control plants, progeny arising from seeds in the large, fully mature 'early' fruits produced significantly more, and larger leaves by day 30 than did those from late fruits (suggesting differential provisioning in seeds during development). Male flower production had a highly significant positive correlation with vegetative mass and a significant negative correlation with fruit production. 相似文献
