以0.25%牛磺胆酸钠-0.25%胰蛋白酶溶液注入大鼠胰导管制备急性(出血坏死性)胰腺炎模型。观察剂量为 3μg/kg的牛胰多肽(BPP)对胰腺炎大鼠胰组织磷脂酶 A(PA)活性、钙含量、胰蛋白酶活性以及血清α_1-抗胰蛋白酶抑制力(STIC)的影响。结果如下:1.在伴注BPP的胰腺炎组,PA 活性、钙含量、胰蛋白酶活性及STIC 分别降为胰腺炎组的51%(P<0.001),75%(P<0.001)、20%(P<0.001)以及24%(P<0.001);2.在伴注碳酰胆碱的胰腺炎组,上述四项指标的水平升高。但在伴注碳酰胆碱及BPP 两者的胰腺炎组却分别降至伴注碳酰胆碱的胰腺炎组的39%(P<0.005)、51%(P<0.001)、40%(P<0.001)以及 59%(P<0.001);3.在伴注Ca~(2 )的胰腺炎组,上述四项指标剧升,注射 BPP对其无影响。结果提示,BPP 能降低大鼠胰组织内磷脂酶A 和胰蛋白酶活性及胰内钙沉积量。机制可能与 BPP打断胰酶活化链有关,是否还影响腺泡细胞膜上M受体的功能,有待探讨。  相似文献

TRANSFORMATION of allogenic lymphocytes in mixed cultures depends chiefly on an incompatibility between the lymphocyte donors at the major histocompatibility locus in man (HL-A), mouse (H-2) and rat (H-l)1. Although the mouse H-2 locus can be divided into several regions each of which controls one or more antigenic specificities2 and two or more subloci control HL-A antigens in man3, it is not known whether all parts of the major histocompatibility locus are equally important in eliciting transformation in mixed lymphocyte cultures. We now show that capacity to elicit lymphocyte transformation is different for different parts of the mouse H-2 locus.  相似文献

A GROUP B herpesvirus is important in the aetiology of Marek's disease, a highly contagious lymphoproliferative disease of chickens1,2. Chicks inoculated with enveloped Marek's disease herpesvirus (MDHV), extracted from feather follicle epithelium of chickens with the disease, developed tumour-like aggregates of lymphoid cells in the viscera and frequently in the peripheral nerves3,4. Cultures of chicken embryo fibroblast (CEF) cells infected with MDHV develop discrete foci of altered cells5. Our data show that MDHV infection of cultures of CEF cells, previously infected with an avian leucosis virus (RAV-2), results in both a reduction in the number of MDHV foci and an increase in the complement fixing avian leucosis antigen (COFAL)6 titre.  相似文献

The responses of the neurons to electrocutaneous stimulation, light flashes, and clicks in the cortical region of the motor representation of the rabbit forelimb were investigated by means of intra- and quasiintracellular recordings. In unanesthetized animals, in only eight out of 65 neurons did postsynaptic potentials (PSP) with a short (10–30 msec) latent period arise in response to light and sound. In 15 neurons, long latency (50–150 msec) responses to one or both of these stimuli were recorded. In most of the cells, short latency stable responses to stimulation of the contralateral forelimb and unstable long latency responses to light and/or sound, frequently of the nature of an increase in the background "synaptic noise," were observed. Under deep chloralose narcosis, the type of convergence was sharply changed: in most of the neurons, short latency responses to all the stimuli used appeared. However, the picture of convergence differed from that described earlier [5,6] for the motor cortex of the cat under chloralose narcosis. The responses to various stimuli were less similar to one another; the somesthetic modality substantially "predominated" (judging by the stability and nature of the interaction of the response).Brain Institute, Academy of Medical Sciences of the USSR. The postinhibitory response of a slowly adapting neuron was investigated in experiments on an isolated preparation of crustacean stretch receptor and abdominal nerve chain. The structural features of this preparation are such that this response can be regarded as the response of the postsynaptic membrane to synaptic inhibition and not the action of synaptic excitation. IPSPs arise in the slowly adapting neuron in response to stimulation of the abdominal nerve chain (direct inhibition) or to excitation of the neuron itself (recurrent inhibition). The postinhibitory response consists of the development of action potentials or an increase in their amplitude and frequency. The magnitude of the response is determined by the duration of the inhibition and the state of the neuron membrane. The postinhibitory response was strongest when IPSPs were superposed on cathodal depression. IPSPs and an intracellular hyperpolarizing current evoke similar postinhibitory responses. Repetitive excitation of an inhibitory neuron may result in the appearance of a regular spike discharge from a previously inactive neuron through the mechanism of the postinhibitory response. Activation of a chain of recurrent inhibition increases the duration of the postinhibitory response evoked by direct inhibition or by a hyperpolarizing current. The existence of a chain of recurrent inhibition prevents the cessation of firing by a neuron during increasing cathodal depression. A mechanism of postinhibitory rebound lies at the basis of this phenomenon.  相似文献

To test the possibility that the theta-rhythm may spread passively from the hippocampus into the neocortex in rabbits and to analyze the properties of these rhythms recorded from the surface of the neocortex, an investigation was made of activity recorded simultaneously from the hippocampus and various points of the neocortex, including from a strip of the sensomotor cortex isolated completely with respect to its nervous connections. The use of special correlation methods showed that an appreciable contribution to the potentials recorded from the isolated strip can be made by the passive conduction of hippocampal activity (provided that it generates a regular high-amplitude theta-rhythm). At the same time, potentials in a theta-rhythm may be generated in the isolated strip itself. The theta-rhythm recorded in the intact rabbit cortex may thus be of combined genesis: generated in the neocortex itself and spreading passively from the hippocampus through the brain tissues as through a volume conductor.  相似文献

N-ras is one of the transforming genes in human hepatic cancer cells.It has been found that N-ras was overexpressed at the mRNA and protein level in hepatoma cells.In order to explore the biological roles of N-ras in human hepatic carcinogenesis and the potential application in control of cancer cell growth,a preudotype retrovirus containing antisense sequence of human N-ras was constructed and packaged.A recombinant retrovirus vector containing antisense or sense sequences of N-ras cDNA was constructed by pZIP-NeoSV(X)1.The pseudotype virus was packaged ang rescued by transfection and infection in PA317 and ψ 2 helper cells.It has been demonstrated that the pseudotype retrovirus containing antisense N-ras sequence did inhibit the growth of human PLC/PRF/5 hepatoma cells accompanied with inhibition of p21 expression,while the retrovirus containing sense sequence had none.The pseudotype virus had no effect on human diploid fibroblasts.  相似文献

In Drosophila pseudoobscura, the amylase (Amy) multigene family is contained within a series of inversions, or gene arrangements, on the third chromosome. The Standard (ST), Santa Cruz (SC), and Tree Line (TL) inversions are central to the phylogeny of arrangements, and have clusters of other arrangements derived from them. The gene arrangements belonging to each of these three clusters have a characteristic number of Amy genes, ranging from three in ST to two in SC to one in TL. This distribution pattern can reflect a history of either duplications or deletions, although the data available in the past did not permit a decision between these alternatives. We provide unambiguous evidence that three Amy genes were present before the divergence of the ST, SC, and TL arrangements. Thus, the current status of the Amy multigene family is the result of deletions in the TL and SC arrangements, which created three new pseudogenes: TL Amy2-psi, TL Amy3-psi, and SC Amy3- psi. Analysis of pseudogene sequences revealed that, in the SC and ST arrangements, pseudogene evolution has been retarded, most likely due to the homogenization effect of gene conversion. Finally, by determining the original copy number, we have reconstructed the evolutionary history of the Amy multigene family and linked it with the evolution of the central gene arrangements.   相似文献

经过茉莉酸甲酯处理的花生幼苗,在形态、解剖和生理上都发生变化,其中以125mg/L处理最显著。处理后的植株幼苗矮化,叶小而厚,叶片贮水细胞大、蒸腾减弱、内源脱落酸和脯氨酸含量增多、过氧化物酶活性加大。由于水分的丧失减少,叶片水分的贮存增加,从而提高幼苗的抗旱性。  相似文献
