Groups of mice were given an intraperitoneal injection of one of six monoclonal antibodies to Toxoplasma gondii, a mixture of equal amounts of five monoclonal antibodies to T. gondii, or the murine myeloma protein MOPC 21, and challenged with either a highly virulent or moderately virulent parasite strain. Two monoclonal antibodies (FMC 19 and FMC 22) conferred total protection against the moderately virulent challenge, with all mice surviving, whereas 90% of control mice died. FMC 19 and FMC 22 also conferred significant protection against the highly virulent challenge as indicated by a prolonged mean time to death (MTD) of immunized compared with control groups of mice. One monoclonal antibody (FMC 23) and the mixture of five antibodies gave significant protection against the moderately virulent challenge only. Passive immunization with dilutions of FMC 22 antibody indicated that the lowest serum titer needed to confer significant protection to mice against a moderately virulent Toxoplasma challenge was 1/640. Mice challenged with highly virulent tachyzoites that had been preincubated with FMC 22 had a significantly longer MTD than mice challenged with highly virulent tachyzoites that had been preincubated with MOPC 21 or phosphate buffered saline, pH 7.2 (PBS). Immunoprecipitation and autoradiography of radiolabeled tachyzoites confirmed that FMC 19 was directed against a 35,000 molecular weight (mol. wt.) antigen and FMC 22 was directed against a 14,000 mol. wt. fraction. We have examined the possible involvement of protein kinase C (C-kinase) in the initiation of germinal vesicle breakdown (GVBD) in Chaetopterus oocytes. Two tumor-promoting phorbol esters (phorbol-12, 13-dibezoate and 12-0-tetradecanoylphorbol-13-acetate [TPA]) and a permeant diacylglycerol (1-oleoyl-2-acetylglycerol), potent activators of C-Kinase, triggered GVBD. Two other phorbol esters (phorbol-13-monoacetate and 4α-phorbol-12, 13-didecanoate), which do not activate C-kinase, were inactive. Three C-kinase antagonists (W-7, H-7 and retinol) inhibited both naturally-and TPA-induced GVBD, whereas W–5, a much less inhibitory W–7 analog, had no effect on GVBD. Triggering of GVBD by TPA was independent of extracellular Ca2+ . Although naturally-induced GVBD was blocked by micromolar concentrations of the calmodulin antagonist, calmidazolium (R24571), and by millimolar concentrations of the permeant cAMP analog, dibutryryl cAMP, TPA-induced GVBD was not affected by these agents. These results support the hypothesis that both C-kinase and calmodulin are involved in the sequence of events leading to GVBD in this species. 相似文献

SUMMARY. 1. We investigate the importance of a refuge from fish predation to the abundance, species composition and seasonal succession of zooplankton. Thirty lakes representing a range of depths were sampled twice in summer for physical/chemical parameters and zooplankton community structure.
2. We define the refuge from centrarchid predators to be that space between the thermocline and the zone of anoxia. As lakes vary in rate of oxygen depletion from the hypolimnion. the refuge size and lake depth are independent: refuge size decreases during the summer period.
3. Lake depth and refuge size independently explain variation among lakes in zooplankton species composition, but seasonal community change within lakes is best predicted by loss of refuge size.
4. Refuge size also explains the substantial variation in the relative dominance of the two major daphnid species. Lakes possessing a large refuge are dominated by D. pulicaria ; those with a small refuge are dominated by the smaller, D. galeata mendotae . We suggest that lakes of intermediate refuge size, which are characterized by high species diversity, represent a more equitable balance of predation and competition. 相似文献

The effects of temperature on the polar movement of IAA through6-mm and 12-mm segments of Zea mays roots have been investigatedover the range from 1 to 50°C. At all temperatures an acropetal polar movement of IAA predominated,although at low temperatures and at 50°C the 6-mm segmentsshowed a transient basipetal polarity, before the persistentacropetal polarity developed. At 1°C the differences betweenacropetal and basipetal movement of IAA were less distinct thanat the other temperatures. There is, however, a marked metabolically-dependentacropetal movement of IAA through the tissues at 1°C, becausewhen the segments were deprived of oxygen the acropetal movementwas severely reduced while the basipetal movement was reducedto a smaller extent. At 1°C and at 5°C there was alwaysa persistent basipetal polarity of IAA movement through 6-mmand 12-mm segments under anaerobic conditions. The velocity of acropetal movement (mm h1) was the samethrough the 6-mm and the 12-mm segments and was markedly affectedby temperature. It increased from 1°C to a maximum valueof 8 mm h1 at 31°C and then decreased again at 40and 50°C. The velocity of basipetal movement could be assessedonly at 1 and 5°C at which temperatures it was greater thanthe velocity of acropetal movement, and virtually independentof segment length. The acropetal flux of IAA (cpm h1) was much less through12-mm segments than through 6-mm segments. For both lengthsof segment, however, the flux showed a complex relationshipwith ambient temperature, increasing from 1°C to a maximumat 1015°C, declining to a minimum value at 31°Cand then rising again at 40 and 50°C. The basipetal fluxof IAA could be astimated only at 1 and 5°C at which itwas very much smaller than the acropetal flux. The amount of IAA in the receiver blocks increased linearlywith time at the lower temperatures. At temperatures withinthe range 15°C to about 31°C, however, the amount ofIAA in the receiver blocks began to decline if the transportperiods exceeded a certain length. The time at which this declinein the IAA in the receiver block began was related to the ambienttemperature. Chromatographic analysis indicated one radioactive substancein receiver blocks at the apical end of segments supplied withIAA-1-14C at the basal end after transport periods of 6 h at25°C, and 72 h at 5°C. The ant communities of two adjacent sites bearing markedly different vegetation (heath and mallee) were studied using quadrat counts and pitfall traps in semi-arid northwestern Victoria. The ants were extremely abundant and rich in species (86 species from 27 genera were recorded from two 50 m × 25 m plots within 25 m of each other) and were dominated by species ofMonomorium and Iridomyrmex. Pronounced site differences in species composition were found, with 73% of the most abundant species showing a strong site preference. The ants exhibited marked seasonality: activity was highest in summer and lowest in winter and this was accompanied by a high turnover of species in time, resulting in pronounced seasonal differences in species composition. Most foraging was nocturnal during summer but was almost exclusively diurnal during winter, and individual species displayed distinctive patterns of diet foraging activity. Although the overall temporal distribution of the communities were apparently controlled by temperature, interspecific competition may also be an important factor influencing the seasonal and diel distributions of individual species, resulting in their high degree of temporal separation. The overall consequence is that although the study area contains many abundant species with similar ecological requirements, few of these are active in the same place at the same time. 相似文献

The evaluation of a skin test to detect acquired resistance to Rhipicephalus appendiculatus Neumann ticks is described. An extract of salivary glands of partially fed female R. appendiculatus was prepared by dissection, sonication and filtration and used for intra-dermal injection. Tests were carried out on twelve calves of known resistance to R. appendiculatus and on twelve naive calves to establish threshold values for positive reactions. Four rabbits naive to ticks were skin tested repeatedly to assess immunogenicity of the test. Reactions to the test at 1 h and at 24 h after injection were significantly correlated with resistance. The correlation was higher with the reactions at 24 h. Rabbits were immunized by the test but the reactions never exceeded the positive threshold.
