Growth and characterization of human skin epithelial cell cultures   总被引:6,自引:0,他引:6  
Summary In 129 of 140 attempts, human skin cells were successfully cultured on the dermal collagen bed of sterile, dead pigskin. Diploid epithelial cells grew selectively on the collagen bed; fibroblasts grew on the glass surfaces of the culture dishes. The cultures could be subdivided physically up to six times at a 1:2 split ratio, but at least 24 to 48 cell generations were produced over the months the cells could be carried. Much of the cell multiplication resulted in maturation into distinct basal, squamous, granular, and keratinized cell layers. The cultured cells were considered epithelial because of their shape, possession of intercellular bridges, desmosomes and tonofibrils, and because they formed maturating epithelium in vitro and upon transplantation back to the original human donor. As the cells grew they digested the pigskin collagen, thus producing clear zones that could be used to monitor and quantitate cell growth. Multiplication of epithelial cells, rather than migration, was indicated by mitotic figures in colchicine-treated cultures and by DNA synthesis. L cells incubated at 19 °C synthesize mitochondrial rRNA but not cytosol rRNA. In addition to 16 and 13S mitochondrial rRNA, mitochondrial RNA sedimenting at higher S values was also synthesized. The processing of mitochondrial rRNA is slower at 19 °C than at 37 °C.  相似文献

At least two types of Robertsonian exchange are now known in the acrocentric chromosomes of man. Both types involve breakage in the arms adjacent to the centromere. Evidence is presented for a third type of exchange, one involving breakage within the centromere itself, in the grasshopper Percassa rugifrons. In this species, which is regularly homozygous for a single fusion metacentric, the eighteen rod autosomes have small but pronounced granules at the centric end of the chromosome. When C-banded these granules show differential Giemsa staining and appear to represent centromeric chromomeres; these chromomeres are lacking in the metacentric fusion product. Equivalent fusions may have occurred in some mammal species too and possible examples of this are discussed in sheep and mice. The Percassa fusion has led to a modification in both the frequency and the distribution of chiasmata as judged by a comparison of these properties in the metacentric relative to the two next smallest rod equivalents. Comparable modifications are known to occur in other naturally occurring fusions but these changes are certainly not automatic consequences of fusion since they are not shown in at least some newly produced fusion mutants.  相似文献

1. The proton NMR spectra of oxidised and reduced French bean plastocyanin have been recorded on a 270 MHz pulsed spctrometer. 2. The spectrum of a mixture containing the protein in the paramagnetic Cu(II) and diamagnetic Cu(I) states is a superposition of the separate spectra. When ferrirate spectra. 3. The results show that self-exchange between Cu(II)- and Cu(I)-plastocyanin is slow on the NMR time scale (kex less than 2-10(4) M-1-s-1 at 50 degrees C), and that electron transfer in the presence of ferricyanide is rapid (k greater than 1-10(5) M-1-s-1).  相似文献

Four behavioral experiments involving a total of 19 adult male and 27 adult spayed female rhesus monkeys failed to reveal significant sexual stimulatory properties of vaginal lavages obtained from estrogen-treated donor females when the material was applied to spayed nonestrogenized recipient females. However, all but one of the males copulated to ejaculation when paired with estrogenized females. Two of three males showed moderate increases in sexual behavior with recipients when the vaginal lavage tested was contaminated with 24-hr-old ejaculate. When purified aliphatic acids were applied to spayed nonestrogenized recepients, one of two males showed increased frequencies of mounting behavior, but intromissions and ejaculations were not displayed.Quantitative analyses of short chain aliphatic acids in vaginal lavages, the hypothesized active component, revealed that (a) spayed females had nondetectable levels of aliphatic acids; (b) daily estradiol benzoate treatment for 6–10 days (25 μg/day i.m.) induced detectable levels of acetic, propionic, and butyric acids; (c) exposure to estradiol for six months resulted in a fairly constant plateau of aliphatic acid concentrations; and (d) ejaculate from the male caused up to fivefold elevations in the aliphatic acid concentrations.Three intact females were studied throughout a menstrual cycle, and the peak values of aliphatic acids occurred in the luteal phase, several days after presumed ovulation. Three spayed females treated chronically with estradiol were given four daily injections of 5 mg progesterone, and the mean concentrations of aliphatic acids increased from 199.7 to 801.0 μg/ml. However, the endocrine conditions associated with maximum concentrations of aliphatic acids in either intact or spayed females are known from other studies to be associated with decreased likelihood of copulation.We conclude that for the majority of males studied, the application of vaginal lavages obtained from estrogenized donors did not significantly increase copulatory behavior with spayed, nonestrogenized recipient females. Moreover, the data from aliphatic acid determinations suggest that increases in concentrations of these substances are not always associated with facilitation of copulation, since the largest increases were found either (1) following copulation to ejaculation, (2) during the luteal phase in cycles free of copulation, or (3) following progesterone treatment of spayed, estrogenized females. Finally, comparison of our results with those from other laboratories suggests that the mechanism involved in positive effects may depend upon associative learning or upon extinction and disinhibition of sexual interest.  相似文献

—The kinetics of plasma choline (Ch) and the uptake of plasma Ch into the brain were studied by means of intravenous infusion of [2H4]Ch at various rates into anaesthetized and conscious rats. [2H4]Ch levels in both arterial and venous plasma at steady state were linearly related to the infusion rate; however, unlabelled Ch levels were independent of infusion rate. [2H4]Ch levels were higher in the arterial plasma than in the venous plasma, while unlabelled Ch levels were higher in the venous plasma than in the arterial plasma. It was concluded that Ch is being generated in the brain and is released into the venous effluent. The supply of Ch to the plasma is not decreased if the plasma Ch level is increased. The clearance and turnover of Ch in the compartment of its initial distribution are 75 ml kg-1 min-1 and 716 nmol kg-1 min-1, respectively. The uptake of plasma Ch into the brain is not saturated even at very high levels of plasma Ch.  相似文献

A simple method of isolating and characterizing RNA from L-cell mitochondria is described. The mitochondrial fraction is lysed by sodium dodecyl sulphate, and the RNA fractionated by sucrose-density-gradient centrifugation. Pinocytosis as measured by the uptake of 125I labelled PVP by the isolated cultured day 12 rat yolk sac was observed to be linear over a 4 h incubation period and to proceed at a rate of approximately 2.5 microliters/mg protein/h. Cadmium, anti-visceral yolk sac antibody (AVYS) and trypan blue all inhibited pinocytosis in a concentration-dependent fashion when added to the culture medium, although at low concentrations trypan blue was slightly stimulatory. The effect of zinc on the inhibition of pinocytosis by these three teratogens was studied. It was observed that zinc ameliorated the inhibitory effects of cadmium and AVYS, but had no effect on inhibition by trypan blue. These results indicate that the previously demonstrated protective action of zinc against cadmium-induced yolk sac dysfunction is not specific to that agent but extends to inhibition of pinocytosis by AVYS, and further suggest that, because of its refractoriness to zinc, trypan blue-induced inhibition of pinocytosis by yolk sac occurs by a mechanism different from that effected by cadmium and AVYS.  相似文献

Animal populations have undergone substantial declines in recent decades. These declines have occurred alongside rapid, human‐driven environmental change, including climate warming. An association between population declines and environmental change is well established, yet there has been relatively little analysis of the importance of the rates of climate warming and its interaction with conversion to anthropogenic land use in causing population declines. Here we present a global assessment of the impact of rapid climate warming and anthropogenic land use conversion on 987 populations of 481 species of terrestrial birds and mammals since 1950. We collated spatially referenced population trends of at least 5 years’ duration from the Living Planet database and used mixed effects models to assess the association of these trends with observed rates of climate warming, rates of conversion to anthropogenic land use, body mass, and protected area coverage. We found that declines in population abundance for both birds and mammals are greater in areas where mean temperature has increased more rapidly, and that this effect is more pronounced for birds. However, we do not find a strong effect of conversion to anthropogenic land use, body mass, or protected area coverage. Our results identify a link between rapid warming and population declines, thus supporting the notion that rapid climate warming is a global threat to biodiversity.  相似文献
