The goal of the study was to show that immobilized purple bacteria could photoproduce H(2) using dark fermentation effluent (FE) as substrate. Simple pretreatment of an inexpensive glass-fiber matrix accelerated the immobilization process. Photobioreactors (PhBR) containing immobilized Rhodobacter sphaeroides GL produced 0.128 L H(2) h(-1) L(-1) of PhBR volume (0.570 L h(-1) L(-1) of matrix) for up to 3 months when continuously fed artificial media with volatile fatty acids (VFAs) or FE from potato and starch fermentations. Hydrogen production was insensitive to NH(4)(+) up to 1 mM and saturated at 8 mM lactate or 1.5% potato FE (diluted in water and supplemented with critical micronutrients). The efficiency of VFA transformation to H(2) was 50-70% of theoretical. At nonlimiting substrate concentrations in artificial media or FE, acetate was utilized before butyrate. High volumetric rates of continuous H(2) photoproduction and stability of the process are advantages of using immobilized cultures. Use of H(2) photoproduction as a polishing step in the treatment of FEs from dark fermentations increased the total amount of H(2) produced from 0.9 to 4.7 mol mol(-1) glucose equivalent in the original potato homogenate.  相似文献

Neem Azal-T/S, Neudosan and Spruzit flüssig at different concentrations were evaluated against the second and fourth instar larvae of E. chrysomelina under laboratory conditions. In case of acute effects, the highest mortality was recorded when larvae were offered squash leaves treated with the highest concentration (2%) of Neem Azal-T/S, and the lowest mortality was, however recorded after treating them with the lowest concentration (0.25%). On the other hand, the second instar larvae were more susceptible to any compound at any concentration than the fourth instar one. One per cent Neudosan formulation showed the highest toxicity to both the 2nd and 4th instar larvae. As the concentration of this compound decreased the mortality percentage of treated larvae decreased to reach the minimum at the lowest concentration (0.25%). The same trend could be applied for Spruzit flüssig compound. When Neem Azal-T/S was compared with Neudosan, the later at any concentration was more toxic than the former. Generally, Neudosan and Spruzit flüssig seemed to be active against E. chrysomelina during the first three days after treatment, then, deterioration occurred among this compound. In case of latent effects, the duration of the treated fourth instar larvae of E. chrysomelina with any tested compound, was prolonged. An increase in the concentration caused prolongation of this period to reach the longest at the highest concentration. This prolongation, was more pronounced after Neem Azal-T/S treatment followed by Neudosan and Spruzit flüssig. The same trend could be applied for the duration of the pupal stage as being affected by the different concentrations of the tested compounds which were applied to the fourth instar larvae. However, adults of E. Chrysomelina treated as fourth instar larvae with high concentrations of Neem Azal-T/S showed some adult malformations. The sex ratios of adults produced from fourth instar larvae treated with different concentrations of the tested natural compounds were about 1?:?1, except for two cases; 1% Neudosan where the number of females were double the number of males and 0.5% Spruzit flüssig where the number of males were double the number of females. An increase in the concentration of any tested compound caused an obvious decrease in the percentage of emerged adults. The least emergence percentage was recorded in the case of Spruzit flüssig, followed by Neudosan and Neem Azal-T/S. Treating the fourth instar larvae with the different concentrations of the tested natural compounds significantly shortened the longevity of produced adults and decreased the fecundity.  相似文献

The influence of the degree and mode of light limitation on growth characteristics of turbidostat cultures of Rhodobacter capsulatus was investigated using mass and energy balance regularities. Light limitation was achieved by increasing the steady-state biomass concentration at constant incident light intensity ( approximately 100 W/m(2)) or by decreasing the incident light intensity at constant steady-state biomass concentration ( approximately 500 mg of dry biomass/L). It was shown that under conditions of light limitation of Rh. capsulatus, the content of P and N in the biomass as well as the biomass degree of reduction were determined by the growth rate of the cultures. The energetic yield of biomass of Rh. capsulatus and total bacteriochlorophyll a content increased when light limitation increased. These parameters were higher in the cultures, in which light limitation was achieved by lowering the incident light intensity at low biomass concentration. This seems to be due to different distribution of light within the photobioreactor when dissimilar modes of light limitation were used.  相似文献

Spiral colonies of the diatom Aulacoseira ambigua were assigned the rank of forma (Aulacoseira ambigua f. japonica). This spiral-shaped, colonial, centric diatom has limited geographical distribution and is currently reported to occur in only a few countries in the world. In Africa this species was described for the first time from the Vaal River, South Africa, but recent research on the Nile River also revealed its presence in Egypt. Physical and chemical data on water quality in these two river systems were compared to determine whether the presence of this uncommon ‘phenoecodeme’ could be ascribed to similar environmental conditions. Results indicate that the rivers are dissimilar with regard to many variables, but both rivers provide turbid, warm and eutrophic waters with medium to high mineral content and it was concluded that these factors favour and sustain the growth of this curved form.  相似文献

Role of Cbl in shear-activation of PI 3-kinase and JNK in endothelial cells   总被引:2,自引:0,他引:2  
Fluid shear stress can activate PI-3 kinase and JNK in vascular endothelial cells. This study was designed to establish the role of Cbl as an upstream molecule in the shear stress activation of PI-3 kinase and JNK. Confluent monolayers of bovine aortic endothelial cells (BAECs) were subjected to a shear stress of 12 dyn/cm(2) over intervals ranging from 0.5 to 30 min. Shear stress increased Cbl phosphorylation to 2.9-fold of control and Cbl association with the regulatory PI-3 kinase subunit p85 to 5.4-fold. The PI-3 kinase activity measured in Cbl-immunoprecipitated complexes increased to 11.7-fold in response to shear, suggesting that the shear stress activation of PI-3 kinase involves its association with Cbl. Furthermore, the shear stress induction of JNK was attenuated by a negative mutant of Cbl. Finally, shear stress caused an activation of PI 3-kinase only in BAECs seeded onto fibronectin, vitronectin, or laminin, but not poly-l-lysine. Our results suggest that Cbl plays a critical role in the shear stress induction of PI 3-kinase and JNK activities, and that this shear-induced activation requires the interaction of endothelial integrins with extracellular matrix proteins.  相似文献

The method of RAPD-PCR and comparative analysis of the PCR fingerprinting profiles similarity was used to characterize interspecific diversity of natural isolates of the lactic acid bacteria Streptococcus thermophilus. The strain genetic diversity was demonstrated using three primer variants, designed for different bacterial genome regions. The resolution of RAPD-PCR technique with different primers for identification at the species level and for certification at the strain level, was examined relative to the commercially important cultures of S. thermophilus. The results provided conclusion on preferable usage of RAPD-PCR with the primer ERIC-1 for specific identification of S. thermophilus, and with the primer M13 for certification of natural isolates of this species at the strain level.  相似文献

Twenty-five Streptococcus thermophilus isolates were analyzed using pulse-field gel electrophoresis (PFGE) and gene restriction profile analysis techniques. 16S rRNA gene sequences of the isolates were almost 100% homologous. However, genomic fingerprinting analysis has shown variability in both genome size and restriction fragments length. The genomes varied from 1417 to 2075 kb resulting in the difference between marginal genome sizes in about 600 kb. The results are indicative of Streptococcus thermophilus intraspecies genetic polymorphism, the origin of which requires further investigation.  相似文献
