Random amplified polymorphic DNA (RAPD) analysis was adapted for genomic identification of cell cultures and evaluation of DNA stability in cells of different origin at different culture passages. DNA stability was observed in cultures after no more than 5 passages. Adipose-derived stromal cells demonstrated increased DNA instability. RAPD fragments from different cell lines after different number of passages were cloned and sequenced. The chromosomal localization of these fragments was identified and single-nucleotide variations in RAPD fragments isolated from cell lines after 8–12 passages were revealed. Some of them had permanent localization, while most variations demonstrated random distribution and can be considered as de novo mutations. 相似文献

Hydrogen production by Anabaena variabilis ATCC 29413 and of its mutant PK84, grown in batch cultures, was studied in a photobioreactor. The highest volumetric H(2) production rates of native and mutant strains were found in cultures grown at gradually increased irradiation. The native strain evolved H(2) only under an argon atmosphere with the actual rate as high as the potential rate (measured in small vials under optimal conditions). In this case 61% of oxygenic photosynthesis was used for H(2) production. In contrast the mutant PK84 produced H(2) during growth under CO(2)-enriched air. Under these conditions at the maximum rate of H(2) production (10 mL h(-1) L(-1)), 13% of oxygenic photosynthesis was used for H(2) production and the actual H(2) production was only 33% of the potential. Under an atmosphere of 98% argon + 2% CO(2) actual H(2) production by mutant PK84 was 85% of the potential rate and 66% of oxygenic photosynthesis was used for H(2) production. Hydrogen production under argon + CO(2) by the mutant was strictly light-dependent with saturation at about 300 microE m(-2) s(-1). However, the rate of photosynthesis was not saturated at this irradiation. At limiting light intensities (below 250 microE m(-2) s(-1)) 33-58% of photosynthesis was used for H(2) production. Hydrogen evolution by PK84 under air + 2% CO(2) was also stimulated by light; but was not saturated at 332 microE m(-2) s(-1) and did not cease completely in darkness. The rate of oxygen photoevolution was also not saturated. A mechanism for increasing cyanobacterial hydrogen production is proposed. 相似文献

Abstact Milk and dairy products purchased at Egyptian markets and breast milk from lactating mothers in Cairo and Giza governorates
were analyzed for some mycotoxins. Three of 15 cows’ milk samples were found positive for Afl. M1 with mean value 6.3 ppb. Only one sample of dried milk was positive (5 ppb). Two of 10 hard cheese samples contained detectable
levels of Afl. M1 (3and 6 ppb), whereas one sample containing Afl. B1 and G1 (10 and 4 ppb resp.). For soft cheese one sample of 10 was positive for Afl. M1 (0.5 ppb). Blue veined cheeses were free of Afl. M1 and PR-toxins.
For breast milk two of 10 samples were positive for Afl. M1 (20%) with mean value 2.75 ppb, while 3 of 10 samples were positive for Ochratoxin A (30 %). 相似文献

Several plasmid derivatives of broad-host-range Inc P4 plasmid RSF1010 were constructed and characterized. Vector pAYC30 was constructed by insertion in vivo into the genome of RSF1010 the Hgr transposon Tn501, originating from the plasmid pVS1 of Pseudomonas aeruginosa. Plasmids with inserts of PstI or SacI fragments may be selected by inactivation of genes sul and aph, respectively. The cloning at unique site SalGI leads to the appearance of HgCl2--sensitive transformants. Versatile cloning vector pAYC1 consists of two replicons, RSF1010 and plasmid pMZ7, a derivative of R6K. The constructed plasmid is 16.9 kb in length and determines resistance to five drugs. Two promoter-probe broad-host-range vectors, pAYC36 and pAYC37, were obtained by replacing a small segment from the DNA sequence of the aph gene promoter of previously described plasmid pAYC32 with the polylinker from plasmid pUC19. Therefore, vector plasmids retained the intact gene aph (Smr); however, they have Sms phenotype because of the insertional inactivation of the promoter. The genetic structure of promoter-probe vectors allows one to select clones, containing hybrid plasmids with an active promoter for gene aph expression. 相似文献

Overall, 72 strains of lactic acid thermophilic streptococci isolated from sour milk products manufactured in various regions of Russia and European countries were analyzed using classical microbiological and molecular biological methods. Physiological and biochemical properties and genetic diversity of these Streptococcus thermophilus strains were studied, and a comparative analysis of the nucleotide sequences of the 16S rRNA gene was conducted. It has been demonstrated that the homology of proximal parts of the 16S rRNA gene of all the strains studied towards one another and towards the reference strain ATCC19258 amounts to 100%. As for the sugar fermentation, some strains display the characteristics untypical of the S. thermophilus members. The data obtained suggest that it is preferable to use gene 16S rRNA sequencing data for identification of natural isolates of closely related lactic acid bacterial species; moreover, this method is recommended for a precise species identification of industrial bacterial strains used in the food industry. 相似文献

This study reports data on the contents of organochlorine pesticides and heavy metals in the muscles and livers of eight individuals (five males and three females) of the Pacific walrus Odobenus rosmarus divergens Illiger, 1815, which were captured in the summer of 2011 in Mechigmensky Bay of the Bering Sea. Pesticides, namely α-, β-, and γ-isomers of hexachlorocyclohexane (HCH), dichlorodiphenyltrichloroethane (DDT) and its metabolites dichlorodiphenyldichloroethane (DDD) and dichlorodiphenyldichloroethylene (DDE), as well as the heavy metals Cd, Pb, and Hg were found in all samples studied. The total concentration of organochlorine pesticides in the muscles varied from 200 to 5700 ng/g lipid weight and in the liver from 4900 to 90300 ng/g lipid weight. The concentrations of cadmium, lead, and mercury were 0.04–6.7, 0.13–0.76, and 0.03–0.40 μg/g dry weight, respectively. On the whole, the contents of organochlorine pesticides and heavy metals in the organs of the Pacific walrus were lower compared to those in marine mammals from other regions of the World Ocean. 相似文献
