荞麦属植物淀粉酶和甲酸脱氢酶同功酶研究(英文)

以聚丙烯酰胺凝胶电泳方法研究了荞麦属植物8个种42个收集系干种子和发芽种子的淀粉酶和甲酸脱氢酶同功酶。结果表明,荞麦淀粉酶在于种子中缺乏活性,但是在发芽种子中活性很强。在供试材料的发芽种子中共发现23个淀粉酶谱带,其中甜荞和苦荞分别有10条和8条。不同荞麦种间淀粉酶谱带差异很大,但是同种内不同收集系间差异较小。谱带聚类分析表明大野荞和毛野荞分别与甜荞和苦荞较近缘,支持它们分别为甜荞和苦荞祖先种的假说。在干种子和发芽种子中,发现所有荞麦种类均只有1条位置一致的甲酸脱氢酶谱带,暗示该酶在进化中具有高度稳定性。     

Genetic analysis of fluvastatin response and dyslipidemia in renal transplant recipients

The Assessment of Lescol in Renal Transplantation clinical trial demonstrated the efficacy of fluvastatin in reducing cardiovascular (CV) disease in renal transplant recipients. The study included a voluntary pharmacogenetic component, enrolling 1,404 patients, which allowed association testing of baseline measures and longitudinal analysis of the 707 fluvastatin-treated and 697 placebo-treated individuals. A candidate gene approach, examining 42 polymorphisms in 18 genes, was used to test for association between selected polymorphisms and major adverse cardiac events, graft failure, change in LDL and HDL cholesterol, and baseline LDL and HDL cholesterol. Reported associations between cholesteryl ester transfer protein (CETP) and baseline HDL cholesterol were replicated, with four previously implicated single nucleotide polymorphisms significantly associated in males and one in females; tests of reported associations between CETP and CV disease yielded varying results. We found no evidence for genetic factors affecting fluvastatin response. Polymorphisms in 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGCR) previously reported to affect the efficacy of pravastatin did not show a similar effect on the reduction of LDL cholesterol by fluvastatin.     

Admixture mapping of an allele affecting interleukin 6 soluble receptor and interleukin 6 levels

Circulating levels of inflammatory markers can predict cardiovascular disease risk. To identify genes influencing the levels of these markers, we genotyped 1,343 single-nucleotide polymorphisms (SNPs) in 1,184 African Americans from the Health, Aging and Body Composition (Health ABC) Study. Using admixture mapping, we found a significant association of interleukin 6 soluble receptor (IL-6 SR) with European ancestry on chromosome 1 (LOD 4.59), in a region that includes the gene for this receptor (IL-6R). Genotyping 19 SNPs showed that the effect is largely explained by an allele at 4% frequency in West Africans and at 35% frequency in European Americans, first described as associated with IL-6 SR in a Japanese cohort. We replicate this association (P<1.0x10-12) and also demonstrate a new association with circulating levels of a different molecule, IL-6 (P<3.4x10-5). After replication in 1,674 European Americans from Health ABC, the combined result is even more significant: P<1.0x10-12 for IL-6 SR, and P<2.0x10-9 for IL-6. These results also serve as an important proof of principle, showing that admixture mapping can not only coarsely localize but can also fine map a phenotypically important variant.

屏障环境的门禁控制系统的设计与应用

实验动物屏障环境设施在动态运行时是一个封闭的系统,进入的人、动物、饲料、空气、垫料及用品均须进行严格的微生物控制。传递仓、渡槽、风淋室和缓冲间设备上的控制环节缺少必要的门禁自动化控制设备,在日常工作上极容易造成违章操作和失误,该门禁系统可控制设备的开关门程序和时间,并可启动消毒设备,实现门禁控制和消毒灭菌的目的。     

Complete Genomic Sequence Analysis of a New SV40 Isolate

The genome of a new SV40 strain (SV-IMB) isolated from a rhesus monkey was completely sequenced and compared with other isolates. The results showed that the whole genome contains 5246bp, and the average identity of SV-IMB was 98.1% as compared to other SV40 isolates. Its regulatory region is composed of a complete enhancer and a defective enhancer. Amino acid changes occurred to some extent in both the large T antigen (T-Ag) and VP1 region. The findings demonstrate that the SV-IMB is a new SV40 isolate.     

Recent Advances in Biological Control of Pest Insects by Using Viruses in China

Insect viruses are attractive as biological control agents and could be a feasible alternative to chemical insecticides in the management of insect infestations. This review describes recent advances in the development of wild-type and genetically modified viruses as insecticides. A new strategy of application of insect viruses in China is reviewed. Also, the assessment of biosafety of genetically modified Helicoverpa armigera Nucleopolyhedovirus (HearNPV) is emphasized as a case-study.     

用超大孔树脂吸附和低乙醇浓度洗脱提纯异黄酮的研究

传统大孔吸附树脂吸附分离中草药有效成分通常需要高浓度的有机溶剂作为洗脱剂,在大规模生产中具有安全隐患和对环境的污染。本文以异黄酮为目标产物,考察了8种不同骨架、不同孔径的树脂对异黄酮的吸附分离性能,其中包括本实验室自制的孔径为1000nm的超大孔树脂。实验结果表明,与传统大孔树脂吸附层析法相比,超大孔树脂吸附层析不仅工艺简单、快速而且大幅度降低了有机溶剂的使用量。洗脱时,乙醇浓度由70％降低到20％。     

冻融作用对陆地生态系统氮循环关键过程的影响效应及其机制

冻融作用是中、高纬度及高海拔地区土壤普遍存在的一种自然现象,是非生长季陆地生态系统氮循环的重要影响因素.冻融作用主要通过改变土壤的理化性质及生物学性状来影响氮素在土壤中的迁移与转化.目前,冻融作用对陆地生态系统氮循环各个过程影响的研究结果不尽一致,理论机制尚不明晰,研究方法也需进一步地探索与创新,因此有必要对现有成果进行梳理和分析,以更好地把握冻融作用下的氮循环过程.本文结合国内外已有研究成果,论述了冻融作用对陆地生态系统氮循环关键过程（氮矿化、固持、硝化与反硝化过程、氮淋溶及气态损失）的影响效应及其主要机制,对目前研究中存在的不足进行了剖析,并对未来研究中迫切需要关注的重点研究方向进行了探讨与展望.     

上海市近地面CO2浓度及其与下垫面特征的定量关系

城市化所带来的土地利用变化和化石燃料燃烧对全球碳循环和气候变化产生了深远影响.明确城市区域CO₂浓度的空间变化特征,对于认识和控制温室气体排放、减少人类活动对全球气候变化的影响具有重要意义.本研究以高强度人类扰动和快速城市化背景下的上海市为研究对象,于2014年春季利用近红外气体分析仪Li 840A开展近地面CO2浓度样带监测,结合遥感数据获取的城市下垫面特征信息,在明确上海市近地面CO₂浓度空间分布格局的基础上,进一步定量分析其对城市下垫面特征的响应机制.结果表明： 上海市近地面CO₂浓度为(443.4±22.0) μmol·mol^-1,城市中心CO₂浓度比郊区平均高12.5%(52.5 μmol·mol^-1).近地面CO₂浓度空间异质性显著,呈现西北高、西南次之、东南低的趋势,总体表现为随着下垫面城市化水平的降低而降低. 城市下垫面植被覆盖率(C_Veg)是城市近地面CO₂浓度的重要指示因子,两者呈现负相关;不透水层覆盖率(C_ISA)次之,两者呈正相关.CO₂浓度（C_CO2）与C_ISA及C_Veg的相关性(R²)在缓冲距离为5 km时同时达到峰值,三者之间的定量关系可通过建立逐步回归方程表征：C_CO2=0.32C_ISA-0.89C_Veg+445.13 (R²=0.66, P<0.01).
