目的:存在阻塞性尿路疾患的老年男性在发生脑血管意外后,是否可通过早期症状或排尿症状类型(梗阻性还是刺激性)来预判排尿功能障碍的病因。方法:选择57例脑卒中后主诉排尿障碍的老年男性患者,所有患者均有继发于良性前列腺增生(BPH)的膀胱出口梗阻(BOO)症状。采集病史并行体检,57位患者均实行尿动力学检查,检查结果行A-G图分析并分类为:有梗阻,无梗阻及可疑梗阻。结果:患者平均年龄70岁(54-87),按排尿障碍的主诉类型分组(纯刺激症状42%,纯梗阻症状34%,两者混合24%),其中51例(89%)在脑卒中发生后即出现排尿症状,47(82%)例患者出现逼尿肌反射亢进(DH),在三组患者中无显著统计学差异。压力流率分析显示,36(63%)位患者有出口梗阻,无梗阻14(24%)例,可疑梗阻7(13%)例。在3组患者中亦无显著统计学差异。结论:所有老年男性患者呈现的症状不能预测膀胱出口梗阻或逼尿肌反射亢进的尿动力学结果。中风发生后排尿功能障碍症状的发生率明显升高,表明由脑血管意外引起的排尿功能障碍合并前期具有膀胱出口梗阻疾病时,可能会使后者的症状恶化,反之亦然。  相似文献

Beauvericin, a cyclohexadepsipeptide-possessing natural product with synergistic antifungal, insecticidal, and cytotoxic activities. We isolated and characterized the fpBeas gene cluster, devoted to beauvericin biosynthesis, from the filamentous fungus Fusarium proliferatum LF061. Targeted inactivation of the F. proliferatum genomic copy of fpBeas abolished the production of beauvericin. Comparative sequence analysis of the FpBEAS showed 74% similarity with the BbBEAS that synthesizes the cyclic trimeric ester beauvericin in Beauveria bassiana, which assembles N-methyl-dipeptidol monomer intermediates by the programmed iterative use of the nonribosomal peptide synthetase modules. Differences between the organization of the beauvericin loci in F. proliferaturm and B. bassiana revealed the mechanism for high production of beauvericin in F. proliferatum. Our work provides new insights into beauvericin biosynthesis, and may lead to beauvericin overproduction and creation of new analogs via synthetic biology approaches.  相似文献

p28, a 28kD protein from toad (Bufo bufo gargarizans) oocytes, was identified by using p13sucl-agarose affinity chromatography. Sequence homology analysis of the full-length cDNA of p28 (Gene Bank accession number: AF 314091) indicated that it encodes a protein containing 224 amino-acids with about 55% identities and more than 70% positives to human, rat or mouse UCH-L1, and contains homological functional domains of UCH family. Anti-p28 monoclonal antibody, on injecting into the oocytes, could inhibit the progesterone-induced resumption of meiotic division in a dose-dependent manner. The recombinant protein p28 showed similar SDS/PAGE behaviors to the native one, and promoted ubiquitin ethyl ester hydrolysis, a classical catalytic reaction for ubiquitin carboxyl terminal hydrolases (UCHs). Water deficit-induced abscisic acid (ABA) accumulation is one of the most important stress signaling pathways in plant cells. Redox regulation of cellular signaling has currently attracted particular attention, but much less is known about its roles and mechanisms in plant signaling. Herein, we report that water deficit-induced ABA accumulation could be regulated by ascorbic acid (AA)-controlled redox status in leave apoplast. The AA content in non-stressed leaves was approximately 3 umol/g FW, corresponding to a mean concentration of 3 mmol/L in a whole cell. Because AA is mainly localized in the cytosol and chloroplasts, the volume of which is much smaller than that of the whole cell, AA content in cytosolic and chloroplast compartments should be much higher than 3 mmol/L. Water deficit-induced ABA accumulation in both leaf and root tissues of maize seedlings was significantly inhibited by AA and reduced glutathione (GSH) at concentrations of 500 umol/L and was completely blocked by 50 mmol/L AA and GSH. These results suggest that the AA-induced inhibition of ABA accumulation should not occur at sites where AA exists in high concentrations. Although water deficit led to a small increase in the dehydroascorbic acid (DHA) content, no significant changes in AA content were observed in either leaf or root tissues. When compared with the whole leaf cell, the AA content in the apoplastic compartment was much lower (i.e. approximately 70 nmol/g FW, corresponding to 0.7 mmol/L). Water deficit induced a significant decrease (approximately 2.5-fold) in the AA content and an increase (approximately 3.4-fold) in the DHA content in the apoplastic compartment, thus leading to a considerably decreased redox status there, which may have contributed to the relief of AA-induced inhibition of ABA accumulation, alternatively, promoting water deficit-induced ABA accumulation. Reactive oxygen species (ROS) could not mimic water deficit in inducing ABA accumulation, suggesting that the inhibition of ABA accumulation by AA or GSH was not related to their ROS-scavenging ability. The results of the present study suggest that the redox status in the apoplastic compartment, as determined by AA and DHA, may play a vital role in the regulation of the signaling process for water deficit-induced ABA accumulation.  相似文献

RAPD是一种新的分子生物学技术。本实验用28个随机引物对平菇、香菇及其融合子进行PCR扩增, 在融合子谱带中,有64条与平菇相同(与平菇的相似指数SI=70.6%),14条与香菇相同(SI=33.6%),另外有26条谱带为双亲与融合子所共有。本实验结果说明融合子是双亲融合的产物,但在融合过程中,从双亲获得的遗传信息不等。本实验首次在食用真菌中获得了原生质体融合的分子生物学证据。我们认为,在食用菌研究中,RAPD将是一种有重要价值的分析方法。 Abstract:RAPD is a novel molecular biological technique.P.sapidus,L.edodes and their intergeneric fusants were tested by RAPD method using 28 randomly selected primers.In bands gained from fusants by amplification reaction,there were 64 bands with the same position as in P.sapidus (the similarity index (SI) was 70.6%),14 bands as in L.edodes.This results illustrated that fusants are true hybrid derived from protoplast fusion procedure,but the genetic information obtained from the two parents were not equal.Molecular biological evidence of protoplast fusion had obtained first time in edible mushroom by RAPD method in this experiment,we suggest that this technique is a valuable genetic marker for mushroom study.  相似文献

对159名中国辽宁汉族个体的基因组DNA进行分析,共检出42种等位基因,其中以DRB1*09012(12.8%)、*0701(10.7%)、*1501(10.4%) 最为常见,其次为DRB1*1201(7.9%)、*1202(7.5%)、*1101(6.6%)、*0301(5.0%)。并发现辽宁汉族人DRB1等位基因频率与白种人间存在明显差异,揭示不同人种有其自己的主要等位基因。同时对本技术在HLA-DRB1分型应用中的优点进行了讨论。 Abstract HLA-DRB1 alleles of 159 Chinese donors from Liaoning Han population were determined by using a set of 23 sequence specific oligonucleotide probes directed to various DRB1 alleles. The samples were first amplified and divided into 8 groups by allele/group specific primers. The SSOs enable us to identify 58 different DRB1subtypes. 42 alleles were detected in the study of this population. Among them, the DRB1*09012(12.8%), *0701(10.7%), *1501(10.4%), *1201(7.9%), *1202(7.5%), *1101(6.6%), *0301(5.0%) are the most frequent. The significant difference was found in Chinese northern Han population by comparing the gene frequencies in Caucasoid samples, suggesting that there were principal DRB1 alleles in different races.  相似文献

目的:探讨桉柠蒎对慢性阻塞性肺疾病急性加重期(AECOPD)患者肺功能及氧化应激的影响。方法:选取2010年9月-2017年6月在我院进行治疗的AECOPD患者126例,采用乱数表法将所有患者分为对照组和研究组各63例。对照组给予盐酸氨溴索口服溶液治疗,研究组给予桉柠蒎肠溶软胶囊治疗,比较两组患者的临床疗效及治疗前后的用力肺活量(FVC)、第1秒用力呼气容积(FEV1)以及第1秒用力呼气容积占预计值百分比(FEV%)、丙二醛(MDA)、8-异构前列腺素(8-isoprostane)水平,观察两组患者治疗过程中不良反应发生率。结果:研究组患者的总有效率为95.24%,高于对照组的82.54%(P0.05)。两组患者治疗前的FVC、FEV1、FEV%比较无统计学差异(P0.05),治疗28d后两组患者的FVC、FEV1、FEV%均明显上升,且研究组的FVC、FEV1、FEV%高于对照组(P0.05)。两组患者治疗前的MDA、8-isoprostane比较无统计学差异(P0.05),治疗28d后两组患者的MDA、8-isoprostane明显下降(P0.05),治疗28d后研究组的MDA、8-isoprostane低于对照组(P0.05)。对照组不良反应发生率为7.94%(5/63),与研究组的6.35%(4/63)相比,差异无统计学意义(P0.05)。结论:桉柠蒎可明显改善AECOPD患者的肺功能和氧化应激,具有较好的临床疗效,且不良反应率低,值得临床推广应用。  相似文献
