Blood samples were collected from a local strain of chickens associated with serious tumor cases in Shandong Province.The samples were inoculated into chicken embryo fibroblast and DF-1 cells for virus isolation and identification,respectively.The inoculated cells were screened for three common chicken tumor viruses.Nine strains of avian leukosis virus subgroup J(ALV-J) were identified,and were designated LY1201‐LY1209.The env gene from the LY1201 strain was amplified and cloned.All nine resultant env clones(clones 01-09) were sequenced,and the gp85 and gp37 amino acid regions were subjected to homology analysis.Clones 01 and 03 had 10 amino acid deletions in the gp85 region compared to the other seven clones,suggesting that at least two quasispecies with obvious mutations coexist in the same field strain.Among these nine clones,three had identical gp85 and gp37 sequences,and were recognized as the dominant LY1201 quasispecies.The amino acid sequence homology of gp37 and gp85 among the nine clones was 98.5%-100.0% and 96.6%-100.0% respectively,suggesting that the gp85 region of the env gene can better display the quasispecies diversity of ALV-J than gp37.  相似文献

This study aimed to discuss the energy budget of Elliot’s pheasant Syrmaticus ellioti in different seasons, with life and health, good growth and normal digestion of Elliot’s pheasant as the tested objects, The energy budget of Elliot’s pheasant was measured by daily collection of the trial pheasants’ excrement in the biological garden of Guangxi Normal University from March 2011 to February 2012. The results showed that the gross energy consumption, metabolic energy and excrement energy varied by season, increasing as temperature decreased. There was significant difference in gross energy consumption, metabolic energy, excrement energy between adults and nonages. There was also a trend that food digestibility of pheasants increases as temperature increases. In the same season, the food digestibility of adults was better than that of nonages. Throughout spring, summer, autumn and winter, the metabolic energy of 4-year adults were 305.77±13.40 kJ/d, 263.67±11.89 kJ/d, 357.23±25.49 kJ/d and 403.12±24.91 kJ/d, respectively, and the nonages were 284.86±17.22 kJ/d, 284. 66±15.16 kJ/d, 402. 26±31.46 kJ/d and 420. 30±31.98 kJ/d, respectively. The minimum metabolic energies were 247.65±21.81 g, 265.86±26.53 g, respectively for each group, detected between 4-year adults and 1-year nonages. Further study is needed to determine whether 29.6 C is the optimal temperature for the Elliot’s pheasant.  相似文献

Elucidation of the mechanisms of liver fibrogenesis is important to treat liver fibrosis.In this study,we established rat models of liver fibrosis with stages from 0–1,2,and 3–4 to 4 at 2,4,6,and 8 weeks,respectively,by injection of pig serum.Liver fibrogenesis was detected by Masson’s trichrome staining.Rat non-parenchymal cells(NPCs)were enriched 4-fold by Percoll density gradient centrifugation.Protein extracts from NPCs were prepared at 4 and 8 weeks,separated by two-dimensional electrophoresis,and then stained with Coomassie Blue G-250.At 4 weeks,we identified 18 non-redundant differentially expressed proteins of which protein disulfide-isomerase associated protein 3(PDIA3)and NDUV showed consistent expression at protein and mRNA levels from 4 to 8 weeks.PDIA3 was found to be down-regulated by Western blotting in the rat model and immunohistochemically in human liver.Our results revealed important aspects of the pathogenesis/progression of liver fibrosis and demonstrated important changes in protein expression levels of NPCs at various stages of liver fibrosis.  相似文献

本文对大规模人类cDNA 测序过程中获得的一条高保守基因进行了初步功能研究,生物信息学研究发现该基因在人类、小鼠、果蝇、拟南芥和裂殖酵母中都有很高的保守性,其他分析预测该基因可能具有肿瘤相关性。RT-PCR分析表明,该基因在成人和胎儿组织中广谱表达。利用基因芯片分析该基因在7例肝癌、5例胰腺癌、2例喉癌和2例肺癌中表达情况,结果证实了该基因的肿瘤相关性,并且提示该基因在不同肿瘤类型中可能处于不同的地位。 Abstract:Preliminary function research of a highly conserved human gene,which was cloned from human fetal cDNA library during large-scale cDNA sequencing,is illustrated in this article.Bioinformatics analysis indicates that this gene is highly conserved in human、mouse、fruit fly、thaliana and fission yeast.Other bioinformatics analysis implies its relevance with tumors.RT-PCR analysis shows its wide-ranging expression patterns.Its expression in 16 cancer cases(including 7 liver cancer cases、5 pancreas cancer cases、2 larynx cancer cases and 2 lung cancer cases) is studied by using gene microarray analysis.The result shows its relevance with tumors and implies it may have different status in different classification of tumors.  相似文献

应用模糊数学中的模糊综合评价方法对广东省1999—2001 年春植玉米区域试验新组合进行定量化评价。结果表明,粤单2号、南玉 18号与南玉35号分别居于1999—2001年区试组合评价第一位;参试两年的组合粤单2号、南玉18号、登海3号与华引115号综合评价较高,表现稳定,可以在广东省内推广应用。 Abstract:The fuzzy comprehensive evaluation method was applied to the quantitative evaluation of the maize new cross combinations from 1999 to 2001 in Guangdong province.The results showed that Yuedan 2,Nanyu 18 and Nanyu 35 ranked the first among the combination evaluations of regional trials in 1999-2000.The comprehensive evaluation values of the combination of Yuedan 2,Nanyu 18 and Denghai 3 were high and stable in the successive two-year experiment.So they were recommended to popularize in Guangdong province.  相似文献

The phylogeny of Ptychostomum was first spacer (ITS) region of the nuclear ribosomal (nr) DNA DNA rps4 sequences. Maximum parsimony, maximum undertaken based on analysis of the internal transcribed and by combining data from nrDNA ITS and chloroplast likelihood, and Bayesian analyses all support the conclusion that the reinstated genus Ptychostomum is not monophyletic. Ptychostomum funkii (Schwagr.) J. R. Spence (≡ Bryum funkii Schwaigr.) is placed within a clade containing the type species of Bryum, B. argenteum Hedw. The remaining members of Ptychostomum investigated in the present study constitute another well-supported clade. The results are congruent with previous molecular analyses. On the basis of phylogenetic evidence, we agree with transferring B. amblyodon Mull. Hal. (≡ B. inclinatum (Brid.) Turton≡ Bryum archangelicum Bruch & Schimp.), Bryum lonchocaulon Mull. Hal., Bryum pallescens Schleich. ex Schwaigr., and Bryum pallens Sw. to Ptychostomum.  相似文献
