目的分析金黄色葡萄球菌所致肺部感染的耐药性特点及其Panton—Valentine杀白细胞素基因的携带状况。方法回顾性调查了温州医学院第一附属医院2005年1月至2006年1月医院感染的金黄色葡萄球菌所致肺部感染患者132例,对其体外药敏试验进行分析;并利用多重PCR检测其PVL基因,应用多位点基因序列分型(multilocus sequence typing,MLST)技术对PVL基因阳性的菌株进行序列分型。耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcus aureus,MRSA)的SCCmec基因分型采用多重聚合酶链反应。结果致肺部感染的132株金黄色葡萄球菌的耐药现象较为严重,仅对万古霉素、呋喃妥因及复方新诺明等药物的敏感率较高;其中经多重PCR筛选出10株携带PVL基因的金葡菌,全部为MRSA菌株,3株为ST239-SCCⅢ,2株为ST398-SCCmecⅢ,2株为ST398-SCCmecⅣ,ST25-SCCmecⅢ、ST59-SCCmecⅠ和ST88-SCCmecⅢ各1株。结论肺部感染的金黄色葡萄球菌对多种抗生素耐药,呈多重耐药性;其携带PVL基因占一定比例。  相似文献

A new species and two new Chinese records in the genus Ulocladium from soil are reported. New species is Ulocladium leve, and the new records are U. alternariae and U. consortiale. Latin diagnosis of the new species is presented. A total of 50 isolates of soil dematiaceous hyphomycetes belonging to 39 species in 18 genera were found from the area of Dahingganling Mountain Range. Among them, Monodictys arxanensis is a new species. Chrysosporium fastidium, Exserohilum pedicellatum, Monodictys asperospora, Papulaspora brachiata, Scolecobasidium anelli, Scolecobasidium verruculosum and Ulocladium tuberculatum are new to China. The other 31 taxa which have been previously reported from China are also listed. Specimens (dried cultures) and living cultures of all fungi studied have been deposited in the Herbarium of Shandong Agricultural University: Plant Pathology (HSAUP).  相似文献

从除虫菊叶中筛选到1株内生真菌Fusarium sp.Y2,对其进行了基础培养基的筛选及培养条件优化研究。结果表明,Y2菌株的较佳发酵培养基为:每L培养基中含有麦芽糖10g、蛋白胨6g、KH2PO42g、KCl 1g、FeSO40.02g、MgSO4·7H2O1g;较佳发酵条件为:初始pH8.5,250mL三角瓶装液100mL,摇床转速180r/min,培养温度28.0℃,接种量21%。在此条件下与原始发酵条件相比,Y2菌株发酵液对番茄灰霉病菌菌丝生长抑制率达98.6%,提高了6.8%;对其孢子萌发抑制率达95.4%,提高了16.1%;其茵丝生长量(干质量)达8.975mg/mL,增加了7.8%。  相似文献

Genetic mapping provides a powerful tool for quantitative trait loci (QTL) analysis at the molecular level. A simple sequence repeat (SSR) genetic map containing 590 markers and a BCI population from two cultivated tetraploid cotton (Gossypium hirsutum L.) cultivars, namely TM-1 and Hai 7124 (G. barbadense L.), were used to map and analyze QTL using the composite interval mapping (CIM) method. Thirty one QTLs, 10 for lobe length, 13 for lobe width, six for lobe angle, and two for leaf chlorophyll content, were detected on 15 chromosomes or linkage groups at logarithm of odds (LOD)≥2.0, of which 15 were found for leaf morphology at LOD≥3.0. The genetic effects of the QTL were estimated. These results are fundamental for marker-assisted selection (MAS) of these traits in tetraploid cotton breeding.  相似文献

Leaves from annual young grape plants (Vitis vinifera L. cv. Jingxiu) were used as experimental materials. The ultrastructural characteristics of mesophyll cells in chilling-treated plants after heat acclimation (HA) and in heat-treated plants after cold acclimation (CA) were observed and compared using transmission electron microscopy. The results showed that slight injury appeared in the ultrastructure of mesophyll cells after either HA (38℃ for 10 h) or CA (8℃ for 2.5 d), but the tolerance to subsequent extreme temperature stress was remarkably improved by HA or CA pretreatment. The increases in membrane permeability and malondialdehyde concentration under chilling (0℃) or heat (45℃) stress were markedly inhibited by HA or CA pretreatment. The mesophyll cells of plants not pretreated with HA were markedly damaged following chilling stress. The chloroplasts appeared irregular in shape, the arrangement of the stroma lamellae was disordered, and no starch granules were present. The cristae of the mitochondria were disrupted and became empty. The nucleus became irregular in shape and the nuclear membrane was digested. In contrast, the mesophyll cells of HA-pretreated plants maintained an intact ultrastructure under chilling stress. The mesophyll cells of control plants were also severely damaged under heat stress. The chloroplast became round in shape, the stroma lamellae became swollen, and the contents of vacuoles formed clumps. In the case of mitochondria of control plants subjected to heat stress, the outer envelope was digested and the cristae were disrupted and became many small vesicles. Compared with cellular organelles in control plants, those in CA plant cells always maintained an integrated state during whole heat stress, except for the chloroplasts, which became round in shape after 10 h heat stress. From these data, we suggest that the stability of mesophyll cells under chilling stress can be increased by HA pretreatment. Similarly, CA pretreatment can protect chloroplasts, mitochondria, and the nucleus against subsequent heat stress; thus, the thermoresistance of grape seedlings was improved. The results obtained in the present study are the first, to our knowledge, to offered cytological evidence of cross-adaptation to temperature stresses in grape plants.  相似文献

为了应用更多的Y染色体特异性STR基因座以用于法医学和人类遗传学研究,本文用PCR结合PAGE技术检测128例山西汉族无关男性DYS605等位基因分布状况。结果显示:山西地区汉族男性DYS605基因座观察到22,21,20,19,18共5个等位基因,基因频率分别为0.0156;0.1797;0.4531;0.2891;0.0625。等位基因20和19之间的电泳距离在非变性胶上非常接近,要有足够的电泳距离才能区分。测序表明该基因座包括3个串联重复区,其中一个为可变重复区。20例女性DNA未发现扩增产物。Abstract: We study the polymorphism at DYS605 ,a new tetranucleotide Y-STR locus,in a Chinese Han population of Shanxi to meet the need of more genetic markers in forensic practice and genetic analysis. DNA were extracted from 128 unrelated male venous blood, and amplified using GDB primers. PCR products were detected using non-denaturing polyacrylamide gel electrophoresis and silver staining. Five alleles named 22,21,20,19,18 were observed with frequency of 0.0156;0.1797;0.4531;0.2891;0.0625. PCR products were not found in female DNA. Using a long enough gel for a long electrophoresis time is strongly encouraged because the rung between allele 20 and allele19 is smaller than expected. Allele sequences show that the repetitive units of DYS605 were composed with the variant units and non-variant units.  相似文献

A non-radioactive labeling system was used to label the probe. During the process, the quality of probe was normalized three times to rule out the possibility of false results.
