紫孢侧耳、糙皮侧耳及其融合菌株的同工酶分析   总被引:4,自引:0,他引:4  
采用聚丙烯酰胺凝胶垂直板电泳方法对紫孢侧耳、糙皮侧耳及其融合菌株的酯酶和过氧化物酶同工酶进行了分析。经多次试验,得到了两亲株和融合株恒定的同工酶谱, 在酶谱中融合菌株与亲株有明显差异,并表现出典型的双亲株互补酶带特征,从而证实融合菌株为两亲株基因重组之产物。 Abstract: After many repetitions,the patterns of esterase isozyme and peroxidase isozyme of Pleurotus sapidus,Pleurotus ostreatus and their fusant strains were obtained.There were differences among the parent strains and theirfusant strains in theirisozyme bands.Parental isozyme bands appeared in patterns of fusant strains.Itdemonstrated that fusant were recombinants of P.sapidus and P.ostreatus.  相似文献

Many ecological studies and conservation management plans employ noninvasive scat sampling based on the assumption that species’ scats can be correctly identified in the field. However, in habitats with sympatric similarly sized carnivores, misidentification of scats is frequent and can lead to bias in research results. To address the scat identification dilemma, molecular scatology techniques have been developed to extract DNA from the donor cells present on the outer lining of the scat samples. A total of 100 samples were collected in the winter of 2009 and 2011 in Taxkorgan region of Xinjiang, China. DNA was extracted successfully from 88% of samples and genetic species identification showed that more than half the scats identified in the field as snow leopard(Panthera uncia) actually belonged to fox(Vulpes vulpes). Correlation between scat characteristics and species were investigated, showing that diameter and dry weight of the scat were significantly different between the species. However it was not possible to define a precise range of values for each species because of extensive overlap between the morphological values. This preliminary study confirms that identification of snow leopard feces in the field is misleading. Research that relies upon scat samples to assess distribution or diet of the snow leopard should therefore employ molecular scatology techniques. These methods are financially accessible and employ relatively simple laboratory procedures that can give an indisputable response to species identification from scats.  相似文献

The atypical PKC isoforms(ζandι)play essential roles in regulating various cellular processes.Both the hetero-interaction between PKCζand p62 through their N-terminal PB1 domains and the homo-oligomerization of p62 via its PB1 domain are critical for the activation of NF-B signaling;however,the molecular mechanisms concerning the formation and regulation of these homotypic complexes remain unclear.Here we determined the crystal structure of PKCζ-PB1 in complex with a monomeric p62-PB1 mutant,where the massive electrostatic interactions between the acidic OPCA motif of PKCζ-PB1 and the basic surface of p62-PB1,as well as additional hydrogen bonds,ensure the formation of a stable and specific complex.The PKCζ-p62 interaction is interfered with the modification of a specific Cys of PKCζby the antiarthritis drug aurothiomalate,though all four cysteine residues in the PKCζ-PB1 domain can be modified in in vitro assay.In addition,detailed structural and biochemical analyses demonstrate that the PB1 domains of aPKCs belong to the type I group,which can depolymerize the high-molecular-weight p62 aggregates into homo-oligomers of lower order.These data together unravel the molecular mechanisms of the homo-or hetero-interactions between p62 and PKCζand provide the basis for designing inhibitors of NF-B signaling.  相似文献
