10E8-like neutralizing antibodies against HIV-1 induced using a precisely designed conformational peptide as a vaccine prime

Recent studies have demonstrated that the membrane-proximal external region(MPER)of human immunodeficiency virus 1(HIV-1)glycoprotein 41 contains a series of epitopes for human monoclonal antibodies,including 2F5,Z13e1,4E10,and10E8,which were isolated from HIV-1-infected individuals and show broad neutralizing activities.This suggests that MPER is a good target for the development of effective HIV-1 vaccines.However,many studies have shown that it is difficult to induce antibodies with similar broad neutralizing activities using MPER-based peptide antigens.Here,we report that 10E8-like neutralizing antibodies with effective anti-HIV-1 activity were readily induced using a precisely designed conformational immunogenic peptide containing the 10E8-specific epitope.This immunogenic peptide(designated T10HE)contains a 15-mer MPER-derived 10E8-specific epitope fused to T-helper-cell epitopes from tetanus toxin(tt),which showed a significantly stabilized-helix structure after a series of modifications,including substitution with an(S)--(2-pentenyl)alanine containing an olefin-bearing tether and ruthenium-catalyzed olefin metathesis,compared with the unmodified T10E peptide.The stabilized-helix structure of T10HE did not affect its capacity to bind the 10E8 antibody,as evaluated with an enzyme linked immunosorbent assay(ELISA)and surface plasmon resonance binding assay(SPR assay).The efficacies of the T10HE and T10E epitope vaccines were evaluated after a standard vaccination procedure in which the experimental mice were primed with either the T10HE or T10E immunogen and boosted with HIV-1 JRFL pseudoviruses.Higher titers of 10E8-like antibodies were induced by T10HE than that by T10E.More importantly,the antibodies induced by T10HE showed enhanced antiviral potency against HIV-1 strains with both X4 and R5 tropism and a greater degree of broad neutralizing activity than the antibodies induced by T10E.These results indicate that a 10E8-epitope-based structure-specific peptide immunogen can elicit neutralizing antibodies when used as a vaccine prime.     

Biomechanism of impact resistance in the woodpecker’s head and its application

The woodpecker does not suffer head/eye impact injuries while drumming on a tree trunk with high acceleration(more than 1000×g) and high frequency.The mechanism that protects the woodpecker’s head has aroused the interest of ornithologists,biologists and scientists in the areas of mechanical engineering,material science and electronics engineering.This article reviews the literature on the biomechanisms and materials responsible for protecting the woodpecker from head impact injury and their applications in engineering and human protection.     

Physiological and genetic correlates of boldness: characterising the mechanisms of behavioural variation in rainbow trout, Oncorhynchus mykiss

Bold, risk-taking animals have previously been putatively linked with a proactive stress coping style whereas it is suggested shyer, risk-averse animals exhibit a reactive coping style. The aim of this study was to investigate whether differences in the expression of bold-type behaviour were evident within and between two lines of rainbow trout, Oncorhynchus mykiss, selectively bred for a low (LR) or high (HR) endocrine response to stress, and to link boldness and stress responsiveness with the expression of related candidate genes. Boldness was determined in individual fish over two trials by measuring the latency to approach a novel object. Differences in plasma cortisol concentrations and the expression of eight novel candidate genes previously identified as being linked with divergent behaviours or stress were determined. Bold and shy individuals, approaching the object within 180 s or not approaching within 300 s respectively, were evident within each line, and this was linked with activity levels in the HR line. Post-stress plasma cortisol concentrations were significantly greater in the HR line compared with the LR line, and six of the eight tested genes were upregulated in the brains of LR fish compared with HR fish. However, no direct relationship between boldness and either stress responsiveness or gene expression was found, although clear differences in stress physiology and, for the first time, gene expression could be identified between the lines. This lack of correlation between physiological and molecular responses and behavioural variation within both lines highlights the complexity of the behavioural-physiological complex.     

Naringenin regulates expression of genes involved in cell wall synthesis in Herbaspirillum seropedicae

Five thousand mutants of Herbaspirillum seropedicae SmR1 carrying random insertions of transposon pTnMod-OGmKmlacZ were screened for differential expression of LacZ in the presence of naringenin. Among the 16 mutants whose expression was regulated by naringenin were genes predicted to be involved in the synthesis of exopolysaccharides, lipopolysaccharides, and auxin. These loci are probably involved in establishing interactions with host plants.

Wood-inhabiting marine fungi from the coast of Shandong, China Ⅲ

Five species of higher marine fungi were observed on the incubated drift and intertidal woods collected from the coasts of Yellow Sea and Bohai Sea. Among them, Halosphaeriopsis was a genus newly recorded for China. Taxonomy and morphology of these species were discussed in this paper. The specimens were deposited in Mycology Herbarium at Qingdao Agricultural University （MHQAU）.     

The finding of the genus Clonostachys （Hyphomycetes） in China

Clonostachys phyllophila and Clonostachys rogersoniana were isolated from rhizosphere soil of Allium tuberosum in Chaoyang, Liaoning Province. It is the first report of the genus in China. The species was described morphologically and illustrated in this paper, the culture was stored in the Culture Collection of Liaoning Center of Culture Collection （LCCC）.
