偃松次生木质部的结构及其对环境的适应

用光学显微镜,偏振光显微镜和扫描电镜野生与栽培偃松的次生木质部结构,研究表明,茎与要支条生长轮均呈现偏心生长、心;边材区明显,管胞具螺纹裂隙及径裂隙及径列条,胞间具明显的胞间隙。射线管胞分布于射线边缘或中间,或只由射线管胞组成低射线。横生树脂道位于纺锤形射线中,纵生树脂道的数目与生长轮宽度有负的相关性,交叉场纹孔式窗格状。野生的矮化偃松,表现出一定的应压木特征。     

甜菊糖苦涩味质的改善

甜叶菊(Stevia rebaudiana)原产南美洲,系菊科甜菊属多年生草本植物。甜菊属中,仅甜叶菊因其干叶中含有6—12％的甜菊糖,而具有甜味。甜菊糖属天然甜味剂,无毒,无副作用,甜度为蔗糖的250—300倍,含热量低,对糖尿病、高血压症有一定保健效果,在预防龋齿及肥胖症方面也有积极的作用,因此是一个很有发展前景的甜味剂。然而由于甜菊糖固有的苦涩味,异杂味,在使用时必需添加蔗糖来改善口味,因而用量及应用范围均受到一定限制。这给甜叶菊种植的进一步扩大及甜菊糖的发展带来极大的不利因素。     

外源DNA直接导入小麦及其在育种上的应用

本研究选用两个白粒小麦品种作供体,提取其总ＤＮＡ,采用花粉管直接携带法导入７５（１９８）红粒品种受体植株。ＤＮＡ导入的第一代（Ｄ１）,目的性状的转化频率为１．７５％和２．９４％。Ｄ２代变异率显著低于Ｄ１代。对Ｄ１,Ｄ２代所得目的性状变异后代,按照常规育种程序进行Ｄ３代观察与鉴定,得到已稳定遗传的后代,从中选取保持原品种其它优良性状而籽粒为的白色的变异类型混合脱粒,获得７５（１９８）改良新品系。

The inhibitory effect of transthyretin gene on growth of human hepatoma cells

Transthyretin(TTR) gene was highly expressed in normal liver and it has been found to be deleted in part of DNA samples from human hepatic cancer.Its mRNA expression was suppressed in most hepatoma samples.In order to study the biological effect of TTR gene on the growth of hepatoma cells,a recombinant vector containing TTR cDNA was constructed by pCMV,then it was transfected into hepatoma cell lines SMMC-7721 and Q3.It has been demonstrated that the inhibition of growth rate of TTR cDNA transfected hepatoma cells was about 50% in strength compared with that of the control.This inhibition was further enhanced when the transfected hepatoma cells were treated with all-trans retinoic acid.Hepatoma cells of cell lines PLC/PRF/5,SMMC-7721 and Q3 as well as hepatoma cells SMMC-7721 transfected with pCMV or pCMV-TTR were analyzed for TTR expression by Northern hybridization.The low level of TTR expression was found in both hepatoma cell lines and in SMMC-7721 cells transfected with pCMV alone.However,a remarkable TTR mRNA expression was observed in hepatoma SMMV-7721 cells transfected with pCMV-TTR.It seems possible that TTR gene might be a candidate of cancer suppressor gene for human hepatic cancer.     

A stage—specific protein factor binding to a CACCC motif in both human β—globin gene promoter and 5‘—HS2 region

The DNaseI hypersensitive site 2 (HS2) of human β-globin locus control region(LCR) is required for the high level expression of human β-globin genes.In the present study,a stage-specific protein factor (LPF-β) was identified in the nuclear extract prepared from mouse fetal liver at d 18 of gestation,which could bind to the HS2 region of human β-globin LCR.We also found that the shift band of LPF-β factor could be competed by human β-globin promoter.However,it couldn‘t be competed by human ε-globin promoter or by human ^Aγ-globin promoter.Furthermore,our data demonstrated that the binding-sequence of LPF-β factor is 5‘CACACCCTA 3‘,which is located at the HS2 region of β-LCR(from-10845 to-10853 bp)and human β-globin promoter(from-92 to -84 bp).We speculated that these regions containing the CACCC box in both the human β-globin promoter and HS2 might function as stage selector elements in the regulation of human β-globin switching and the LPF-β factor might be a stage-specific protein factor involved in the regulation of human β-globin gene expression.
