Activation of tumor-reactive T lymphocytes is a promising approach for the prevention and treatment of patients with metastatic
cancers. Strategies that activate CD8+ T cells are particularly promising because of the cytotoxicity and specificity of CD8+ T cells for tumor cells. Optimal CD8+ T cell activity requires the co-activation of CD4+ T cells, which are critical for immune memory and protection against latent metastatic disease. Therefore, we are developing
“MHC II” vaccines that activate tumor-reactive CD4+ T cells. MHC II vaccines are MHC class I+ tumor cells that are transduced with costimulatory molecules and MHC II alleles syngeneic to the prospective recipient. Because
the vaccine cells do not express the MHC II-associated invariant chain (Ii), we hypothesized that they will present endogenously synthesized tumor peptides that are not presented by professional Ii+ antigen presenting cells (APC) and will therefore overcome tolerance to activate CD4+ T cells. We now report that MHC II vaccines prepared from human MCF10 mammary carcinoma cells are more efficient than Ii+ APC for priming and boosting Type 1 CD4+ T cells. MHC II vaccines consistently induce greater expansion of CD4+ T cells which secrete more IFNγ and they activate an overlapping, but distinct repertoire of CD4+ T cells as measured by T cell receptor Vβ usage, compared to Ii+ APC. Therefore, the absence of Ii facilitates a robust CD4+ T cell response that includes the presentation of peptides that are presented by traditional APC, as well as peptides that

The phylogeography of species associated with European steppes and extrazonal xeric grasslands is poorly understood. This paper summarizes the results of recent studies on the phylogeography and conservation genetics of animals (20 taxa of beetles, butterflies, reptiles and rodents) and flowering plants (18 taxa) of such, "steppic" habitats in Eastern Central Europe. Most species show a similar phylogeographic pattern: relatively high genetic similarity within regional groups of populations and moderate-to-high genetic distinctiveness of populations from currently isolated regions located in the studied area. This distinctiveness of populations suggests a survival here during glacial maxima, including areas north of the Bohemian Massif-Carpathians arc. Steppic species generally do not follow the paradigmatic patterns known for temperate biota (south-north “contraction–expansion”), but to some extent are similar to those of arctic-alpine taxa. There are three main groups of taxa within Eastern Central Europe that differ in their contemporary distribution pattern, which may reflect historical origin and expansion routes. Present diversity patterns of the studied steppic species suggest that they share a unique genetic signature and distinct assemblages exist in each of the now isolated areas rich in steppic habitats. At least some of these areas probably act as present “interglacial refugia” for steppic species. This study strongly supports the need to protect steppic species throughout their entire ranges in the region, as the continuous destruction of steppic habitats in some areas may lead not only to the disappearance of local populations, but also to the extinction of unique evolutionary units. 相似文献

Extracellular protein profiles were compared for broth-grown cultures of Burkholderia pseudomallei and its avirulent close relative Burkholderia thailandensis. A number of protein bands were present in the B. pseudomallei profile but absent or less abundant in the B. thailandensis profile. Four such prominent secreted proteins were identified by using N-terminal sequencing coupled to searches of the B. pseudomallei genome sequence database. The genes for two proteins with similarity to carbohydrate-binding proteins, and a further protein homologous to known bacterial collagenases, were present in both B. pseudomallei and B. thailandensis. The putative collagenase gene was cloned and expressed as a fusion protein in Escherichia coli. Waste from cattle production contains protozoa, such as Cryptosporidium spp. and Giardia, which can be transmitted to humans. People residing in areas of high cattle density may be at increased risk for protozoan infections. The objective of this study was to assess spatial and temporal associations between cattle density and hospitalizations for protozoan infections in the U.S. elderly. Data on protozoan infections were abstracted from Centers for Medicare and Medicaid Services datasets for a 14-year period (1991–2004). Cattle inventory data were abstracted from the 2002 U.S. Census of Agriculture. Counties were classified into one of five exposure categories based on both cattle density and human density. Our analyses considered differences in rates, trends, and variations in seasonal patterns based on exposure categories. Cryptosporidiosis demonstrated a trend of increasing annual rates related to increased potential exposure to cattle. Both cryptosporidiosis and giardiasis demonstrated significant seasonal patterns peaking during the fourth week of October in areas of high cattle/low population density and the second week of September in counties with low cattle/low human density, respectively. Counties with low human population density (regardless of cattle density) had the highest rate of all protozoan infections, peaking in the summer. These results demonstrate the elderly population is at increased risk of protozoan infections in areas of high cattle density, particularly cryptosporidiosis. The seasonal patterns and higher annual rates seen in rural areas suggest time-variant environmental exposures, which may be affected with geographical and temporal targeting of agricultural policies and interventions to improve public health. 相似文献

A new design method of a broadband wide-angle metal-dielectric-metal plasmonic absorber is presented based on the cavity mode theory. The broadband absorption is implemented by filling a unit cell with multi-size square metal patches resonant at adjacent wavelengths, with the widths of the patches and thickness of the dielectric layer optimized with the presented method. A broadband plasmonic absorber working in the visible range is designed, the absorption of which is insensitive to the azimuth angle of incident field and keeps over 0.7 at incident angle up to 60° for p polarization and above 0.6 at up to 40° for s polarization. 相似文献

An investigation was undertaken to evaluate a set of cyanobacterial strains in terms of production of biocidal compounds exhibiting
allelochemical and fungicidal properties. Two cyanobacterial strains — Anabaena sp. and Calothrix sp. were selected for further investigation, on the basis of their larger inhibition zones on the lawn of Synechocystis and Synechococcus sp. and two phytopathogenic fungi — Rhizoctonia bataticola and Pythium debaryanum. The diameter of the inhibition zone was largest when extracellular filtrates of the two cultures incubated at high light
intensity (90–100 μmol photons m−2 s−1) and temperature (40 ± 2 °C) or grown in medium containing two-folds higher P (1.4 mg/L, as compared to 0.7 mg/L in BG 11
medium) were taken. A pH of 8 was the most optimal for both strains, in terms of growth and biocidal activity. Partial purification
of ethyl acetate extract using TLC, followed by GLC revealed a single peak. This study highlights the importance of environmental
factors in aggravating or reducing the toxic effects of these harmful cyanobacteria and their potential as a biocontrol agent. 相似文献

In this special issue of the Glycoconjugate Journal focusing on glycosciences and development, we summarize recent advances in our understanding of the role of mucin-type O-glycans
in development and disease. The presence of this widespread protein modification has been known for decades, yet identification
of its biological functions has been hampered by the redundancy and complexity of the enzyme family controlling the initiation
of O-glycosylation, as well as the diversity of extensions of the core sugar. Recent studies in organisms as diverse as mammals
and Drosophila have yielded insights into the function of this highly abundant and evolutionarily-conserved protein modification. Gaining
an understanding of mucin-type O-glycans in these diverse systems will elucidate crucial conserved processes underlying many
