Background aimsAdoptive transfer of tumor-specific lymphocytes is a promising strategy in the treatment of cancer. We conducted intratumoral administration of an allogeneic irradiated continuous T-cell line (C-Cure 709) expressing an HLA-A2-restricted MART-1-specific T-cell receptor (TCR) into HLA-A2+ melanoma patients. The C-Cure 709 cell line is cytotoxic against MART-1+ HLA-A2+ melanoma cell lines and secretes several immune stimulatory cytokines upon stimulation.MethodsAnti-tumor immune responses against the commonly expressed tumor antigen (Ag) MART-1 were longitudinally analyzed in peripheral blood by fluorescence-activated cell sorting (FACS) before and after intratumoral injection of C-Cure 709.ResultsNo treatment-induced increase in Ag-specific T-cell frequencies was observed in peripheral blood, and the phenotype of MART-1-specific T cells was very stable during the treatment. Interestingly, despite a very stable frequency of MART-1-specific T cells over the course of treatment, clonotype mapping revealed that the response was in fact highly diverse and dynamic, with new clonotypes emerging during treatment. Only a few clonotypes were recurrently detected in consecutive samples. One MART-1-specific T-cell clone disappearing from peripheral blood was later detected in a metastatic lesion.ConclusionsSequence analyzes of the CDR3 region revealed conserved structural characteristics in the MART-1-specific TCR used by T-cell clones.  相似文献

1. Lake eutrophication has increased phytoplankton blooms and sediment organic matter. Among higher plants, small, oligotrophic rosette species (isoetids) have disappeared, while a few tall, eutrophic species (elodeids) may have persisted. Despite recent reduction of nutrient loading in restored lakes, the vegetation has rarely regained its former composition and coverage. Patterns of recovery may depend on local alkalinity because HCO3? stimulates photosynthesis of elodeids and not of isoetids. In laboratory growth experiments with two isoetids (Lobelia dortmanna and Littorella uniflora) and two elodeids (Potamogeton crispus and P. perfoliatus), we test whether organic enrichment of lake sediments has a long‐lasting influence by: (i) reducing plant growth because of oxygen stress on plant roots and (ii) inhibiting growth more for isoetids than elodeids. We also test whether (iii) increasing alkalinity (from 0.17 to 3.20 meq. L?1) enhances growth and reduces inhibition of organic sediment enrichment for elodeids but not for isoetids. 2. In low organic sediments, higher oxygen release from roots of isoetids than elodeids generated oxic conditions to greater sediment depth for Lobelia (4.3 cm) and Littorella (3.0 cm) than for Potamogeton species (1.6–2.2 cm). Sediment oxygen penetration depth fell rapidly to 0.4–1.0 cm for all four species at even modest organic enrichment and oxygen consumption in the sediments. Roots became shorter and isoetid roots became thicker to better supply oxygen to apical meristems. 3. Growth of elodeids was strongly inhibited across all levels of organic enrichment of sediments being eight‐fold lower at the highest enrichment compared to the unenriched control. Leaf biomass of isoetids increased three‐fold by moderate organic enrichment presumably because of greater CO2 supply from sediments being their main CO2 source. At higher organic enrichment, isoetid biomass was reduced, leaf chlorophyll declined up to 10‐fold, root length declined from 7 to <2 cm and mortality rose (up to 50%) signalling high plant stress. 4. Lobelia was not affected by HCO3? addition in accordance with its use of sediment CO2. Biomass of elodeids increased severalfold by rising alkalinity from 0.17 to 3.20 meq. L?1 in accordance with their use of HCO3? for photosynthesis, while the negative impact of organically enriched sediments remained. 5. Overall, root development of all four species was so strongly restricted in sediments enriched with labile organic matter that plants if growing in situ may lose root anchorage. Other experiments demonstrate that this risk is enhanced by greater water content and reduced consolidation in organically rich sediments. Therefore, formation of more muddy and oxygen‐demanding sediments during eutrophication will impede plant recovery in restored lakes while high local alkalinity will help elodeid recovery.  相似文献

1. Fish community structure and habitat distribution of the abundant species roach, perch and ruffe were studied in Lake Nordborg (Denmark) before (August 2006) and after (August 2007) aluminium treatment to reduce internal phosphorus loading. 2. Rapid changes in fish community structure, abundance and habitat distribution occurred following a decline in in‐lake phosphorus concentrations from 280 to 37 μg P L?1 and an increase in Secchi depth transparency from 1.1 to 1.9 m (August). The proportion of perch in overnight gill net catches increased, whilst roach decreased, and the average weight of all key species increased. 3. The habitat distribution of perch and roach changed from a high proportion in the upper pelagic and littoral zones in 2006, towards enhanced proportions in the deeper pelagic and profundal zone in 2007. The abundance of large‐bodied zooplankton increased and the abundance of benthic invertebrates decreased in the same period, suggesting that the habitat shift was not induced by food limitation. 4. Ruffe shifted from the littoral and upper profundal zones towards the deep profundal zone, likely reflecting an increased predation risk in the littoral zone and better oxygen conditions in the deep profundal. 5. Our results indicate that enhanced risk of predation in the upper pelagic and the littoral zones and perhaps improved oxygen concentrations in the deeper profundal zone at decreasing turbidity are responsible for the observed habitat shift. The results indicate that fish respond rapidly to changes in nutrient state, both in terms of community structure and habitat use.  相似文献

The Eurasian lynx (Lynx lynx) is a common predator of both roe deer (Capreolus capreolus) and reindeer (Rangifer tarandus) in Sweden. We investigated the influence of prey availability, latitude, sex, and age on body size and body mass variation of the Eurasian lynx in Sweden, using data from 243 specimens whose locality of capture, year of capture, sex, and age were known. We found that both body size and body mass of the lynx in Sweden are mainly affected by the lynx sex and age but also by the availability of prey during the first year of life. Body size and body mass of lynx as well as the density of roe deer increased from Central Sweden to South. Furthermore, body size and body mass of lynx increased from Central Sweden to North (i.e. within the reindeer husbandry area). Lynx body size was slightly smaller within the reindeer husbandry area (approximately north of latitudes 62°–63°N) compared to outside, probably because reindeer are more difficult prey to hunt, as well as being migratory and thus an unpredictable prey for the Eurasian lynx compared to the non-migratory roe deer. Our results support a growing body of evidence showing that food availability at growth has a major effect on body size of animals.  相似文献

Seasonal dynamics of suspended minerals, organic matter, particulate, and dissolved organic carbon (DOC), chlorophyll, and their vertical fluxes were studied in a small Arctic fjord (Adventfjorden, Spitsbergen) from November 2006 to October 2007. The fjord was usually covered with fast ice in winter, but remained open throughout the year since 2005. The open-water winter period caused increased wave action and resuspension of organic and mineral particles. The lack of sea-ice in spring accelerated the onset of the productive season. The earlier light signal also caused an earlier appearance of mesozooplanktonic organisms, accompanied by a significant increase of the DOC pool in the water. In the cold period (winter and spring) 47% of the annual organic matter settled on the sampling site, a lot coming from the spring diatom bloom in April. Summer melt (July, August) resulted in turbid and brackish fjord surface water with stratification and increase of both suspended particles and sedimentation, causing 60% of the annual minerals and 53% of the annual organic matter to settle at the sampling site. Increased dissolved organic carbon (DOC) through sloppy feeding on the mixo- and heterotrophic protista by the abundant mesozooplankton indicated intensive secondary production, resulting in a maximum fecal pellets flux of >5 mg C m−2 day−1. A warmer climate with reduced sea-ice cover in fjords will advance the onset of the spring bloom and will also result in a larger input of turbid melt water in summer, restricting the light availability and enhancing flocculation and thereby sedimentation.  相似文献

Most of the Earth's biodiversity resides in the tropics. However, a comprehensive understanding of which factors control range limits of tropical species is still lacking. Climate is often thought to be the predominant range‐determining mechanism at large spatial scales. Alternatively, species’ ranges may be controlled by soil or other environmental factors, or by non‐environmental factors such as biotic interactions, dispersal barriers, intrinsic population dynamics, or time‐limited expansion from place of origin or past refugia. How species ranges are controlled is of key importance for predicting their responses to future global change. Here, we use a novel implementation of species distribution modelling (SDM) to assess the degree to which African continental‐scale species distributions in a keystone tropical group, the palms (Arecaceae), are controlled by climate, non‐climatic environmental factors, or non‐environmental spatial constraints. A comprehensive data set on African palm species occurrences was assembled and analysed using the SDM algorithm Maxent in combination with climatic and non‐climatic environmental predictors (habitat, human impact), as well as spatial eigenvector mapping (spatial filters). The best performing models always included spatial filters, suggesting that palm species distributions are always to some extent limited by non‐environmental constraints. Models which included climate provided significantly better predictions than models that included only non‐climatic environmental predictors, the latter having no discernible effect beyond the climatic control. Hence, at the continental scale, climate constitutes the only strong environmental control of palm species distributions in Africa. With regard to the most important climatic predictors of African palm distributions, water‐related factors were most important for 25 of the 29 species analysed. The strong response of palm distributions to climate in combination with the importance of non‐environmental spatial constraints suggests that African palms will be sensitive to future climate changes, but that their ability to track suitable climatic conditions will be spatially constrained.  相似文献

Surface electromyograms(EMG) during test contractions (TCs) were studied to assess the muscle strain in simulated mushroom picking. Additionally, the duration of the TC for the effective assessment was investigated. Nine female subjects performed standardized shoulder abduction and a stooped posture for one minute as TCs. Each experiment consisted of a 60-min rest, three work periods (W1-W3), a 30-min rest, and two work periods (W4 and W5) separated by a 30-min rest period. The duration of each work period was about 20 min. A total of 18 TCs was performed between the work periods and every 10 minutes in the rest periods. EMGs were recorded from the trapezius, infraspinatus, deltoid, and erector spinae muscles. The amplitude of EMG (AEMG) and mean power frequency (MPF) of EMG were calculated. Each TC was divided equally into three parts. Ratings of perceived exertion (RPE) in the neck, shoulder and low-back were reported during TCs. The work increased RPE of all the parts. AEMG and RPE were increased and MPF was decreased by W1, W2 and W3 in the neck and shoulder muscles. MPF of the erector spinae was increased by the work. The results were not affected by the duration of TCs and the parts during the TCs. AEMG and MPF fluctuated before W1 although the changes of RPE were small. Averaging several TCs was recommended to get stable results from TCs. EMG changes and appropriate TC conditions were discussed in relation to the adaptation in fatiguing contractions.  相似文献

Returns from Harvesting a Commercial Non-timber Forest Product andParticular Characteristics of Harvesters and Their Strategies: Aquilaria crassna and Agarwood in Lao PDR. This paper explores associations between the returns from harvesting a commercial non-timber forest product and particular characteristics of harvesters and their strategies. Data originate from a survey in Laos of the harvest and trade of agarwood, a highly valuable commodity harvested mainly from tree species in the genus Aquilaria. Five groups of harvesters were identified, with considerable differences in returns between them. Foreign harvesters, i.e., ethnic Vietnamese, earned approximately USD 13 per day on average, while nonlocal full-time harvesters earned approximately USD 9, and nonlocal part-time harvesters USD 7 per day. Local people earn approximately USD 5 per day and only USD 4 if harvesting only took place in village forests. Factors observed to determine returns are market access, especially access to national and international markets, and knowledge and experience in trading, harvesting, and the organization of harvesting trips. It is suggested that interventions should be made to address these factors, to level out returns from harvesting, and to increase the share of returns captured by local people.   相似文献

Lignocellulosic plant material is potentially a sustainable source of fermentable sugars for bioethanol production. However, a barrier to this is the high resistance or recalcitrance of plant cell walls to be hydrolyzed. Therefore, a detailed knowledge of the structural features of plant cell walls that contribute to recalcitrance is important for improving the efficiency of bioethanol production. In this work we have used a technique known as Comprehensive Microarray Polymer Profiling (CoMPP) to analyze wheat straw before and after being subjected to hydrothermal pre‐treatments at four different temperatures. The CoMPP technique combines the specificity of monoclonal antibodies with the high‐throughput capacity of microarrays. Changes in the relative abundance of cell wall polysaccharides could be tracked during processing, and a reduction in xylan, arabinoxylans, xyloglucan, and mixed‐linked glucan epitopes was detected at the two highest temperatures of pre‐treatment used. This work demonstrates the potential of CoMPP as a complementally technique to conventional methods for analyzing biomass composition. Biotechnol.
