Seventy-eight cattle samples from three Creole Caribbean islands and one Brazilian breed were analyzed for sequence variation in the hypervariable segment of the mitochondrial DNA control region. Seventy-three samples displayed Bos taurus haplotypes, and five samples exhibited haplotypes that were of Bos indicus ancestry. Phylogenetic analysis revealed that all sampled B. taurus sequences fell into two distinct clusters with separate African and European origins. European sequences were encountered in each population; however, the distribution of African haplotypes was uneven, with the highest proportion of African influence found in the Guadeloupe Creole. The reduced levels of African haplotypic variation within the Caribbean and Brazilian are consistent with prior founder effects. Additionally, genetic variation at three microsatellite loci illustrated African influence uniquely in the Guadeloupe Creole. Collectively, the data suggest that this African influence is, at least in part, attributable to the historical importation of African cattle to the Americas. Furthermore, alleles of B. indicus ancestry were detected at appreciable frequencies in all Caribbean Creole populations and may reflect zebu introgressions from either West Africa or the Indian subcontinent.  相似文献

The genome of Dehalococcoides ethenogenes strain 195, an anaerobic dehalorespiring bacterium, contains 18 copies of putative reductive dehalogenase genes, including the well-characterized tceA gene, whose gene product functions as the key enzyme in the environmentally important dehalorespiration process. The genome of D. ethenogenes was analyzed using a bioinformatic tool based on the frequency of oligonucleotides. The results in the form of a genomic signature revealed several local disruptions of the host signature along the genome sequence. These fractures represent DNA segments of potentially foreign origin, so-called atypical regions, which may have been acquired by an ancestor through horizontal gene transfer. Most interestingly, 15 of the 18 reductive dehalogenase genes, including the tceA gene, were found to be located in these regions, strongly indicating the foreign nature of the dehalorespiration activity. The GC content and the presence of recombinase genes within some of these regions corroborate this hypothesis. A hierarchical classification of the atypical regions containing the reductive dehalogenase genes indicated that these regions were probably acquired by several gene transfer events.  相似文献

A left maxilla fragment from the newly discovered Miocene vertebrate locality of Polastron (Gers, southwestern France) is described and referred to the longsnouted eusuchian crocodilian Gavialosuchus, which had never been reported from the Miocene of this area, in which crocodilian remains are scarce. On the basis of its mammal fauna, the locality is slightly older than Sansan. Although Gavialosuchus is usually found in marine beds, its occurrence in the fluviatile deposits of Polastron shows that it also inhabited rivers.  相似文献

Paranoplocephala maseri n. sp. is described from Lemmiscus curtatus (Cope) in the USA. The new species is related to Paranoplocephala omphalodes (Hermann, 1783), P. caucasica (Kirschenblat, 1938), P. kirbyi Voge 1948, P. microti (Hansen, 1947) and P. macrocephala (Douthitt, 1915) sensu Genov et al. (1996). P. maseri n. sp. differs from P. omphalodes in the position of the genital pores, testes and cirrus-sac; from P. caucasica, in which there is an unarmed cirrus, in both the distribution and larger number of testes; from P. kirbyi in the distribution of the testes, the position of the genital pores and egg dimensions; from P. microti in the distribution and smaller number of testes, the smaller egg dimensions and the position of the genital pores; and from P. macrocephala in the position of genital pores and cirrus-sac.  相似文献

This work was undertaken to investigate the functional consequences of mutations of the essential alpha His45 residue of Vibrio harveyi luciferase, especially with respect to the yield and reactivity of the flavin 4a-hydroperoxide intermediate II. A total of 14 luciferase variants, each with a different single-residue replacement for the alpha His45, were examined. These variants showed changes, mostly slight, in their light decay rates of the nonturnover luminescence reaction and in their Km values for decanal and reduced riboflavin 5'-phosphate (FMNH2). All alpha His45 mutants, however, showed markedly reduced bioluminescence activities, the magnitude of the reduction ranging from about 300-fold to 6 orders of magnitude. Remarkably, a good correlation was obtained for the wild-type luciferase, 12 alpha His45-mutated luciferases, and six additional variants with mutations of other alpha-subunit histidine residues between the degrees of luminescence activity reduction and the dark decay rates of intermediate II. Such a correlation further indicates that the activation of the O-O bond fission is an important function of the flavin 4a-hydroperoxide intermediate II. Both alpha H45G and alpha H45W were found to bind near-stoichiometric amounts of FMNH2. Moreover, each variant catalyzed the oxidation of bound FMNH2 by two mechanisms, with a minor pathway leading to the formation of a luminescence-active intermediate II and a major dark pathway not involving any detectable flavin 4a-hydroperoxide species. This latter pathway mimics that in the normal catalysis by flavooxidases, and its elicitation in luciferase was demonstrated for the first time by single-residue mutations.  相似文献

Population genetics and phenotypic structures are often predicted to vary along the geographic range of a species. This phenomenon would be accentuated for species with large range areas, with discontinuities and marginal populations. We herein compare the genetic patterns of central populations of Coccinella septempunctata L. with those of two phenotypically differentiated populations considered as rear‐edge populations and subspecies based on phenotype (Algeria and Japan). According to the central‐marginal model and expected characteristics of rear‐edge populations, we hypothesize that these rear‐edge populations have (1) a reduced genetic diversity, resulting from their relative isolation over long periods of time, (2) a higher population genetic differentiation, explained by low contemporary gene flow levels, and (3) a relationship between genetic diversity characteristics and phenotypes, due to historical isolation and/or local adaptation. Based on genotyping of 28 populations for 18 microsatellite markers, several levels of regional genetic diversity and differentiation are observed between and within populations, according to their localization: low within‐population genetic diversity and higher genetic differentiation of rear‐edge populations. The genetic structuring clearly dissociates the Algerian and Eastern Asia populations from the others. Geographical patterns of genetic diversity and differentiation support the hypothesis of the central‐marginal model. The pattern observed is in agreement with the phenotypic structure across species range. A clear genetic break between populations of Algeria, the Eastern Asia, and the remaining populations is a dominant feature of the data. Differential local adaptations, absence of gene flow between marginal and central populations, and/or incapacity to mate after colonization, have contributed to their distinct genotypic and phenotypic characteristics.  相似文献

Summary The distribution of serotonin-, tyrosine hydroxylase-, and FMRFamide-immunoreactive neuronal elements, as well as the concentrations of serotonin and dopamine in the different parts of the gastrointestinal tract, were studied in the snail Helix pomatia. The sensitivity of the spontaneous contractions of the alimentary tract to serotonin, dopamine, and FMRFamide was also tested. Serotonin-, tyrosine hydroxylase-, and FMRFamide-immunoreactive elements could be demonstrated in each part of the gastrointestinal tract, but they showed different innervation patterns. Serotonin- and tyrosine hydroxylase-immunoreactive elements were dominant in the submucosal layer, whereas FMRFamide-immunoreactive elements were dominant in both the mucosal and submucosal layers. Tyrosine hydroxylase-immunoreactive elements were confined to the longitudinal muscle trabeculae of submucosa, whereas serotonin-immunoreactive elements were distributed throughout the submucosal layer. No serotonin-immunoreactive cell bodies, but only fibers, could be detected in the gastrointestinal tract, and therefore they represent extrinsic elements. Tyrosine hydroxylase- and FMRFamide-immunoreactive cell bodies represent intrinsic elements of the tract. The occurrence and density of the serotonin- and tyrosine hydroxylase-immunoreactive elements showed significant differences in the different parts of the alimentary tract, in accordance with HPLC assays, which revealed a significant frontocaudal decrease in both the serotonin (from 2.11 to 1.21 pM/mg) and dopamine (from 3.28 to 0.52 pM/mg) contents of the different parts of the alimentary tract. Dopamine at 10-5 M concentration proved to be effective only on the longitudinal muscles by increasing the tone and frequency of contractions, but was ineffective on the circular muscles. Serotonin affected both the longitudinal and circular muscles. Serotonin at 10-5 M concentration decreased the tone and increased the frequency of low-amplitude contractions of the longitudinal muscles of the esophagus and the gizzard but increased both the tone and frequency of the crop. Serotonin at 10-9 M concentration slightly decreased the tone and blocked the contractions of the circular muscles in the crop but at 10-5 M concentration induced contractions of the circular muscles in the gizzard. FMRFamide at 10-6 M concentration decreased the tone and was shown to block the contractions of both the longitudinal and circular muscles.  相似文献

Intact rich fen soil cores with controlled water levels near thesurface were installed in an open greenhouse. To simulate short termsummer drought, water levels were lowered (20 cm) after two weeks inhalf of the cores (experimental cores) and remained near the surface inthe other half (blanks). After two more weeks, the water levels werebrought back to the surface in the experimental cores and remainedthere for another two weeks. In the blanks, reduction and alkalinizationof the top peat layer occurred. In the experimental soil cores oxidationand acidification started within one week after drawdown. An indicationfor a drought induced rise in soluble reactive phosphorus has beenfound. The velocity of the acidification process illustrates the dynamicnature of the hydrochemical conditions in fen soils during drought. Theprocesses controlling the acid/base status of rich fen, the effect ofdrought induced acidification on P availability and the significance forthe vegetation are discussed.  相似文献

As reported from marine systems, we found that also in15 prealpine lakes N2O concentrations werestrongly correlated with O2 concentrations. Inoxic waters below the mixed surface layer, N2Oconcentrations usually increased with decreasingO2 concentrations. N2O is produced in oxicepilimnia, in oxic hypolimnia and at oxic-anoxicboundaries, either in the water or at the sediment-waterinterface. It is consumed, however, incompletely anoxic layers. Anoxic water layers weretherefore N2O undersaturated. All studied lakeswere sources for atmospheric N2O, including thosewith anoxic, N2O undersaturated hypolimnia.However, compared to agriculture, lakes seem not tocontribute significantly to atmospheric N2Oemissions.  相似文献
