Olfaction is of major importance for survival and reproduction in moths. Males possess highly specific and sensitive olfactory receptor neurones to detect female sex pheromones. However, the capacity of male moths to respond to host‐plant volatiles is relatively neglected and the role that such responses could play in the sensory ecology of moths is still not fully understood. The present study aims to identify host‐plant stimuli for the European grape berry moth Eupoecilia ambiguella Hb. (Tortricidae, Lepidoptera), a major pest of vine in Europe. Headspace volatiles from Vitis vinifera L. cv. Pinot Noir, Vitis vinifera subsp. sylvestris and five other host‐plant species comprising five different families are analyzed by gas chromatography linked to electroantennogram (EAG) recording from male E. ambiguella antennae and by gas chromatography‐mass spectrometry. This procedure identifies 32 EAG‐active compounds, among them the aliphatic compounds 1‐hexanol, (Z)‐3‐hexenol, (Z)‐3‐hexenyl acetate and 1‐octen‐3‐ol; the terpenes limonene, β‐caryophyllene and (E)‐4,8‐dimethyl‐1,3,7‐nonatriene; and the aromatic compounds benzaldehyde and methyl salicylate. Male and female E. ambiguella show similar EAG response amplitudes to individual chemical stimuli and also to mixtures of plant volatiles, as represented by essential oils from ten other plant species. Cytosolic NADPH may act as one of the signals that couple glucose metabolism to insulin secretion in the pancreatic ß-cell. NADPH levels in the cytoplasm are largely controlled by the cytosolic isoforms of malic enzyme and isocitrate dehydrogenase (IDHc). Some studies have provided evidence for a role of malic enzyme in glucose-induced insulin secretion (GIIS) via pyruvate cycling, but the role of IDHc in ß-cell signaling is unsettled. IDHc is an established component of the isocitrate/α–ketoglutarate shuttle that transfers reducing equivalents (NADPH) from the mitochondrion to the cytosol. This shuttle is energy consuming since it is coupled to nicotinamide nucleotide transhydrogenase that uses the mitochondrial proton gradient to produce mitochondrial NADPH and NAD+ from NADP+ and NADH. To determine whether flux through IDHc is positively or negatively linked to GIIS, we performed RNAi knockdown experiments in ß-cells. Reduced IDHc expression in INS 832/13 cells and isolated rat islet ß-cells resulted in enhanced GIIS. This effect was mediated at least in part via the KATP-independent amplification arm of GIIS. IDHc knockdown in INS 832/13 cells did not alter glucose oxidation but it reduced fatty acid oxidation and increased lipogenesis from glucose. Metabolome profiling in INS 832/13 cells showed that IDHc knockdown increased isocitrate and NADP+ levels. It also increased the cellular contents of several metabolites linked to GIIS, in particular some Krebs cycle intermediates, acetyl-CoA, glutamate, cAMP and ATP. 1. Conifer‐feeding budworms (Choristoneura) hibernate in sheltered locations on their host trees from late summer of 1 year to spring of the next. During this period, they do not feed but rely on sustenance provided in the egg. Overwinter survival is dependent on the rate of consumption of these limited reserves. 2. A process model was developed that quantifies the relationship between the rate of consumption and survival at variable temperatures and exposure times for western spruce budworm. The model supported physiological evidence that warm weather conditions early in the diapause period have a dominant influence on overwinter survival. Output compared favourably with field observations of poorer budworm survival at lower elevations where late‐summer and autumn temperatures were warmer compared to those overwintering at cooler, higher elevations. 3. Field experiments demonstrated these weather‐dependent rates of survival were modulated significantly by the degree of shelter experienced by hibernating budworms. 4. Dissection of whole trees harbouring overwintering western spruce budworms showed a significant portion of the population had travelled a considerable distance from the periphery of the tree canopy where eggs were laid to overwinter successfully on the tree bole where sheltered niches are common. 5. In this paper the organic and inorganic geochemistry of fossil and extant Persististrombus latus are compared, together with other strombid species (Lentigo lentiginosus, Lobatus gigas, Strombus alatus, Lobatus raninus, Laevistrombus canarium and Tricornis latissimus). Using a large sample of well‐preserved fossil P. latus shells from the Mediterranean realm, we examined the warming period of sea water in the Middle Pleistocene. A mineralogical study of the shells demonstrates the continuous presence of calcite and a complex organic matter distribution, which was well preserved in many cases, thereby making the U/Th dating of strombid shells unreliable. U/Th analysis of coral samples and amino acid racemization dating of pelecypod shells confirmed that P. latus entered the Mediterranean realm in MIS 7 and 5. The genus Nais is a group of oligochaetous clitellates, common in eutrophic freshwater habitats. About 30 species are described. Species identification is based primarily on chaetal characters, which are often subtle, inconsistent, and even overlapping between nominal species. We investigated the correlation between genetic variation and chaetal morphology in this genus. Eighty‐one individuals from Europe, North America, and China were included in the study. Seventy‐five of these were preserved as vouchers. They were scrutinized with regard to chaetal morphology, and ten different morphotypes were identified. Three molecular markers, two mitochondrial (the COI gene and 16S rDNA) and one nuclear (the ITS region), were used to establish the genetic lineages in the material. Genetic variation was found to be largely congruent with chaetal character patterns. However, at least nine separately evolving lineages (all supported by mitochondrial as well as nuclear data) correspond to at most six nominal species. Four morphotypes/lineages are recognized as Nais barbata, Nais christinae, Nais elinguis, and Nais stolci, respectively, whereas five, or possibly more, lineages represent a morphological continuum covering the variation of the Nais communis/variabilis complex. Thus, cryptic speciation is revealed. Our results indicate that a taxonomic revision of the genus will be needed in the future. 相似文献

The copper(I) chloride, pyridine system has shown marked tryptophan 2,3-dioxygenase activity with tryptophan and indole derivatives. However, only in the cases of tryptophan and 3-methylindole was it possible to isolate the primary products N-formylkynurenine (~1%) and 2-formamidoacetophenone (70%), respectively. In other cases, such as 2-methylindole, methyl indole-3-acetate, tryptamine, indole-3-propionic acid, and acetyltryptophan methyl ester, only secundary products could be isolated or determined by GC-MS methods. 相似文献
