Three glycosilated flavonoids (diosmin, hesperidin and naringin) and respective aglycones were characterized in terms of their apparent ionisation constants and bidirectional permeability using the cellular model Caco-2 as well as the artificial membrane model PAMPA. Ionisation curves were established by capillary electrophoresis. It was confirmed that significant amounts of the aglycones are ionised at physiological pH whereas the glycosides are in the neutral form. Permeation was not detected for the glycosides in either the apical-to-basolateral or basolateral-to-apical directions confirming the need for metabolism before absorption through the intestinal membrane. The aglycones permeated in both directions with apparent permeabilities (P(app)) in the range of 1-8x10(-5) cm/s. The results from both in vitro methods correlated providing some evidence of passive transport; however, the hypothesis of active transport cannot be excluded particularly in the case of diosmetin. Metabolism of the aglycones was detected with the cell model, more extensively when loading in the apical side. Some of the metabolites were identified as glucuronide conjugates by enzymatic hydrolysis. 相似文献

The dynamics of 47 major and trace elements were examined during a long-term (up to 8 years between 1979 and 1988) litter
decomposition experiment in European boreal and nemoboreal forests (Sweden). Litterbags were incubated in 11 monocultural
stands at 10 different locations including seven with Norway spruce (Picea abies (L.) H. Karst.) and four with Scots pine (Pinus sylvestris L.). Principal component analysis and manual scatter plots revealed groups of elements behaving in a similar manner. One
group consisted mainly of nutrients, but also of the unessential or toxic elements Rb, Sc, Sr and Tl, and had a general trend
with decreasing mass-normalised (mn) concentrations during decomposition. Another group contained mostly unessential and potentially
toxic elements, whose concentrations generally increased during the decay process. An exception from this increasing trend
was found for Cd, Hf, Hg, Ta and Zr, for which the mn-concentrations increased initially followed by a net leaching from the
litter. The influence of sea spray was identified in the early stages of the experiment (in particular for Na), and the impact
of the anthropogenic component of atmospheric deposition was clearly visible (e.g. Pb, Cd, Hg). A regional northern Sb source
is indicated in the pine, but not in the spruce, needles. The fact that the samples were collected at a time of higher atmospheric

The ability of the periodontal pathogen Porphyromonas gingivalis to use different glycolipid structures as receptors has previously been demonstrated. The bacterium adhered to acid and nonacid glycolipids originating from human organs and to nonacid glycolipids of porcine origin. The aim of the present study was to analyze these binding epitopes by structural characterization. Glycolipid fractions with positive bacterial binding from e.g. human and porcine origin, were purified by the high performance liquid chromatography technique and thereafter used in bacterial overlay assays with (35)S-labeled P. gingivalis. Purified fractions with positive binding were structurally characterized by proton nuclear magnetic resonance spectroscopy. Complementing thin-layer chromatograms and bacterial overlay assays with pure reference glycolipid fractions and competition experiments with lactose were performed to define potential receptors. The P. gingivalis binding epitopes, including cerebrosides with nonhydroxy fatty acids, lactosylceramide with hydroxy fatty acids, sulfatides, lacto-, neolacto- and gangliotetraosylceramides, are in several instances similar to those found for other bacteria, e.g. H. pylori, H. influenzae and N. meningitidis. In addition P. gingivalis also bound to the Galalpha4Gal epitope of the globo series of glycolipids. In the future these results may be valuable for development of new treatment strategies, such as anti-adhesion therapies and vaccines specifically directed against P. gingivalis infection. 相似文献

The homeotic Abdominal-B (Abd-B) gene expression depends on a modular cis-regulatory region divided into discrete functional domains (iab) that control the expression of the gene in a particular segment of the fly. These domains contain regulatory elements implicated in both initiation and maintenance of homeotic gene expression and elements that separate the different domains. In this paper we have performed an extensive analysis of the iab-6 regulatory region, which regulates Abd-B expression at abdominal segment A6 (PS11), and we have characterized two new polycomb response elements (PREs) within this domain. We report that PREs at Abd-B cis-regulatory domains present a particular chromatin structure which is nuclease accessible all along Drosophila development and both in active and repressed states. We also show that one of these regions contains a dCTCF and CP190 dependent activity in transgenic enhancer-blocking assays, suggesting that it corresponds to the Fab-6 boundary element of the Drosophila bithorax complex. 相似文献

We used radiotelemetry to investigate the time of migratory flight initiation relative to available celestial orientation cues and departure direction of a nocturnal passerine migrant, the reed warbler, Acrocephalus scirpaceus, during autumn migration. The study was carried out at Falsterbo, a coastal site in southwest Sweden. The warblers initiated migration from times well after local sunset and well into the night, corresponding to sun elevations between -4 degrees and -35 degrees, coinciding with the occurrence of stars at night. They departed in the expected migratory direction towards south of southwest with a few initiating migration in reverse directions towards northeast to east. Flight directions under overcast conditions (7-8/8) were more scattered than under clear sky conditions (0-4/8). There were fewer clouds on departure nights than on nights when the birds did not initiate migration. For birds staying longer than one night at stopover the horizontal visibility was higher and precipitation was less likely on departure nights than on the previous night. The results show that the visibility of celestial cues, and stars in particular, are important for the decision to initiate migration in reed warblers. However, cloud cover, horizontal visibility and precipitation might be correlated with other weather variables (i.e. wind or air pressure) that are also likely to be important for the decision to migrate. A full-length cDNA encoding for ferredoxin-nitrite reductase (NiR, EC 1.7.7.1), has been isolated from a root cDNA library from the legume Lotus japonicus and characterised. The NiR gene ( Nii ) is present as a single copy in this plant, and encodes a protein of 582 amino acids. The Lotus NiR protein is synthesised as a precursor with an amino-terminal transit peptide consisting of 25 amino acid residues. Sequence comparisons with leaf NiRs from different plant species and with other related redox proteins identified in the root NiR the same highly conserved residues involved in the cofactor binding than previously reported for leaves. Besides, a putative binding site for ferredoxin was also found in the N-terminal region of the protein. The NiR gene is expressed in roots and leaves, although the level of expression is much higher in roots, in accordance with the fact that L. japonicus assimilates nitrate mainly in roots. NiR mRNA, protein and activity are induced by nitrate in roots and leaves, while ammonium-grown plants only showed basal levels. No oscillations of NiR mRNA, protein and activity were observed during the day/night cycle, neither in roots nor leaves, making an interesting difference with rhythms observed in other plant species. 相似文献

We investigated the orientation of juvenile pied flycatchers, Ficedula hypoleuca, during autumn migration in south Sweden using orientation cage experiments, to study the relative importance of visual and magnetic information at sunset. We performed cage tests under 12 experimental conditions that manipulated the geomagnetic and visual sunset cues available for orientation: natural clear skies in the local or a vertical magnetic field; simulated total overcast in the local or a vertical magnetic field; natural pattern of skylight polarization and directional information from stars screened off, with the sun's position as normal or shifted 120 degrees anticlockwise with mirrors; reduced polarization in the local or a vertical magnetic field; directions of polarization (e-vector) NE/SW and NW/SE, respectively, in the local or a vertical magnetic field. The pied flycatchers were significantly oriented towards slightly south of west when they could use a combination of skylight and geomagnetic cues. The mean orientation was significantly shifted along with the deflection of the sunset position by mirrors. Reduced polarization had no significant effect on orientation either in the local, or in a vertical, magnetic field. The birds tended to orient parallel with the axis of polarization, but only when the artificial e-vector was aligned NW/SE. The mean orientation under simulated total overcast in a vertical, and in the local, magnetic field was not significantly different from random. It is difficult to rank either cue as dominant over the other and we conclude that both visual and magnetic cues seem to be important for the birds' orientation when caught and tested during active migration.
