Summary Our field surveys conducted in 10 Hungarian grape producing areas have revealed that the nutrient contents of vine leaves of different grape varieties were closely correlated with the EUF-nutrient contents of different soil types with different contents of clay. Resulting from these relationships the following EUF-nutrient values are considered as required for optimal nutrition of the vine-stock to attain grape yields of 10–12 t/ha in the areas under investigation:  相似文献

In this review, we have mainly included studies in which whole-body autoradiography was used. In lipid research, most studies have been done with fatty acids. These studies showed some common characteristics in the pattern of tissue distribution. A major uptake was seen in the brown fat, liver and adrenal cortex but also to some extent in other tissues with a high metabolic activity or high cell turn-over, e.g. the gastric and intestinal mucosa, diaphragm, kidney cortex and bone marrow. Low levels of radioactivity were generally found in the brain, testes, thymus, white fat, skeletal muscles, lungs and spleen. Most fatty acids showed some specific features, e.g the strong uptake of erucic, arachidonic and docosahexaenoic acid in myocardium and of eicosapentaenoic acid in the adrenal cortex. Studies with PGE1 and LTC3 showed that the liver and kidney and to a lesser degree the lungs were the major sites of metabolism. The distribution of free cholesterol and triolein emulsion labelled in the fatty acid moieties did show some similarities with respect to the general pattern found with most fatty acids. Specific for cholesterol was a very strong uptake in the adrenal cortex. There was also a significant uptake in the spleen whereas the uptake in the brown fat was not as marked as for most fatty acids. Specific for triolein was a marked uptake in the spleen and myocardium, in fed animals also in the white adipose tissue. These studies show that whole-body autoradiography can give much valuable information of the uptake and distribution of lipids that would be rather difficult to obtain with conventional methods. Combined with electron-microscopy, autoradiography can be used to study cellular and even subcellular distribution, and thus given further data on the metabolism of lipids in the body.  相似文献

A proteoglycan was isolated from fetal membranes which had been separated from human postpartum placenta. The glycosaminoglycan side chains (Mr = 55,000) were found to be composed of 75% chondroitin sulfate and 23% dermatan sulfate as determined by chondroitinase ABC or AC II digestion. NH2-terminal microsequencing of the intact proteoglycan revealed a single amino acid sequence of (sequence; see text) A rabbit antiserum raised against the intact proteoglycan reacted in sodium dodecyl sulfate-polyacrylamide gel electrophoresis immunoblotting with Mr = 45,000 and 43,000 core polypeptides from chondroitinase-treated proteoglycan. Affinity-purified antibodies from this antiserum precipitated from human embryonic fibroblast culture fluid a proteoglycan which has an approximate Mr = 120,000 in sodium dodecyl sulfate-polyacrylamide gel electrophoresis. This proteoglycan has on the average two polysaccharide side chains. As defined by chondroitinase digestion, these chains consist of 66% dermatan sulfate and 20% chondroitin sulfate. Digestion of the glycosaminoglycan with chondroitinase ABC converted the proteoglycan to a Mr = 45,000 major and a Mr = 43,000 minor core polypeptide. Tissue immunofluorescence localized the proteoglycan to interstitial matrices, suggesting that it is a product of mesenchymal cells. The methods we have devised for the purification of the fetal membrane proteoglycan in chemical amounts and the antibodies we have prepared against it will allow studies on the structural and functional properties of the proteoglycan and on the expression of immunologically cross-reactive proteoglycans by various cells and tissues.  相似文献

Summary Clostridium acetobutylicum ATCC 824 was submitted to repeated subculturing at 24-hour intervals for 218 days. The organism retained its ability to form solvents, although the fermentation slowly became increasingly acidogenic during the first 200 days. Except for the initial spore inoculum, the cultures were not subjected to heat shocking between the serial transfers. When the inoculum volume was doubled from 3.3% to 6.7% after 200 days of subculturing, the product formation pattern quickly shifted back from acids to primarily butanol. Acetone production also resumed after being undetectable for more than 50 days. The relative formation of acetate and ethanol remained nearly constant throughout the experiments, while the formation of butyrate mirrored that of butanol.  相似文献

Brain capillaries (microvessels) were isolated from the rabbit and bovine brain. Extensive morphological examinations were performed at the light and electron microscopical levels. The relative contribution of endothelium (52%), basal membrane (32%) and pericytes (16%) to the composition of the microvessel was assessed. The ability of the endothelium from bovine brains to maintain a membrane potential, i.e. to accumulate the lipophilic cation [3H]TPMP, was shown. The transmitter catabolizing enzymes MAO and AchE were shown to be, and COMT and GABA-T not to be associated with the microvessel fraction isolated from rabbits.  相似文献

A 23 kDa protein has recently been demonstrated to participate in photosynthetic oxygen evolution by reconstitution experiments on inside-out thylakoid vesicles (Åkerlund H-E, Jansson C and Andersson B (1982) Biochim Biophys Acta 681:1–10). Here we describe the isolation of the 23 kDa protein from a spinach chloroplast extract using ion-exchange chromatography. The protein was obtained in a yield of 25% and with less than 1% of contaminating proteins. The ability of the protein to stimulate oxygen evolution in inside-out thylakoids was preserved throughout the various fractionation steps. The isolated protein was highly water soluble and appeared as a monomer. Its isoelectric point was at pH=7.3. The amino acid composition showed a high content of polar amino acids, resulting in a polarity index of 49%. The isolated protein lacked metals and other prosthetic groups. Its function as a catalytic or regulating subunit in the oxygen evolving complex is discussed.  相似文献

Acetone extracts of filter-collected urban airborne particulate matter contain compounds which can competitively inhibit 2,3,7,8-[1,6-3H]tetrachlorodibenzo-p-dioxin (TCDD) binding to the rat liver TCDD-receptor protein. The concentration of conventional polycyclic aromatic hydrocarbons (PAHs) or chlorinated dioxins and dibenzofurans cannot account for more than 1-30% of the observed competition for [3H]TCDD binding to the receptor protein. The difference in potency between samples collected in urban areas during different periods of the year and a background sample is 25-400-fold. Collecting samples in the presence of increased concentrations of nitrogen dioxide, nitrous acid, nitric acid or ozone did not increase the amount of compounds with receptor affinity. However, with nitrogen dioxide and especially with nitric acid, a substantial increase of the mutagenic effects in the Ames Salmonella assay in the absence of mammalian activation as well as a degradation of several PAHs were noted. Affinity for the TCDD-receptor protein, mutagenicity in the absence of mammalian metabolic activation in the Ames Salmonella assay and PAH-content are characteristics of urban particulate matter showing the presence of compounds, that represent potential health risks. The compounds with affinity for the receptor may constitute a group of substances different from both conventional PAHs and direct-acting mutagens.  相似文献

The expression and core protein structure of two proteoglycans, the major cartilage proteoglycan isolated from a rat chondrosarcoma and a small molecular weight chondroitin sulfate proteoglycan isolated from a rat yolk sac tumor, have been compared. The cartilage proteoglycan was not detectable in the cartilage tissue of cartilage matrix deficient (cmdcmd) neonatal mice by immunofluorescence, but the cmd cartilage did react with antibodies against the core protein of the yolk sac tumor proteoglycan. Radioimmunoassays showed that the core proteins of these proteoglycans are not cross-reactive with each other. Analysis of the core proteins by sodium dodecyl sulfate/polyacrylamide gel electrophoresis after chondroitinase ABC treatment of the proteoglycan revealed a large difference in their sizes. The cartilage proteoglycan core protein had a molecular weight of about 200,000 while the yolk sac tumor proteoglycan core protein migrated with an apparent molecular weight of about 20,000. In addition, the cultured yolk sac tumor cells that make the small proteoglycan did not react with antiserum against the cartilage proteoglycan. These results indicate that the proteoglycan isolated from the yolk sac tumor is similar to the small chondroitin sulfate proteoglycan species found in cartilage and support the existence of at least two dissimilar and genetically independent chondroitin sulfate proteoglycan core proteins.  相似文献
