Elderly female hypertensives with arterial stiffening constitute a majority of patients with heart failure with preserved ejection fraction (HFpEF), a condition characterized by inability to increase cardiac stroke volume (SV) with physical exercise. As SV is determined by the interaction between the left ventricle (LV) and its load, we wished to study the role of arterial hemodynamics for exertional SV reserve in patients at high risk of HFpEF. Twenty-one elderly (67 ± 9 yr) female hypertensive patients were studied at rest and during supine bicycle stress using echocardiography including pulsed-wave Doppler to record flow in the LV outflow tract and arterial tonometry for central arterial pressure waveforms. Arterial compliance was estimated based on an exponential relationship between pressure and volume. The ratio of aortic pressure-to-flow in early systole was used to derive characteristic impedance, which was subsequently subtracted from total resistance (mean arterial pressure/cardiac output) to yield systemic vascular resistance (SVR). It was found that patients with depressed SV reserve (NoRes; reserve <15%; n = 10) showed decreased arterial compliance during exercise, while patients with SV reserve ≥15% (Res; n = 11) showed increased compliance. Exercise produced parallel increases in LV end-diastolic volume and arterial volume in Res patients while NoRes patients exhibited a lesser decrease in SVR and a drop in effective arterial volume. Poor SV reserve in elderly female hypertensives is due to simultaneous failure of LV preload and arterial vasodilatory reserves. Abnormal arterial function contributes to a high risk of HFpEF in these patients. 相似文献

Studies of cooperation have traditionally focused on discrete games such as the well-known prisoner’s dilemma, in which players
choose between two pure strategies: cooperation and defection. Increasingly, however, cooperation is being studied in continuous
games that feature a continuum of strategies determining the level of cooperative investment. For the continuous snowdrift
game, it has been shown that a gradually evolving monomorphic population may undergo evolutionary branching, resulting in
the emergence of a defector strategy that coexists with a cooperator strategy. This phenomenon has been dubbed the ‘tragedy
of the commune’. Here we study the effects of fluctuating group size on the tragedy of the commune and derive analytical conditions
for evolutionary branching. Our results show that the effects of fluctuating group size on evolutionary dynamics critically
depend on the structure of payoff functions. For games with additively separable benefits and costs, fluctuations in group
size make evolutionary branching less likely, and sufficiently large fluctuations in group size can always turn an evolutionary
branching point into a locally evolutionarily stable strategy. For games with multiplicatively separable benefits and costs,
fluctuations in group size can either prevent or induce the tragedy of the commune. For games with general interactions between
benefits and costs, we derive a general classification scheme based on second derivatives of the payoff function, to elucidate

Chemical control of insect pests is often necessary to ensure high yields of field crops. The aim of the present study was to study whether field pesticide use influences amount of pest damage in neighbouring garden crops. Spring oilseed rape, OSR (Brassica napus L.), was established in Southern Sweden as an example of an agricultural field crop. One half of the OSR field was treated with insecticides to control flea beetles (Phyllotreta spp., Chrysomelidae) and pollen beetle (Meligethes aeneus Fab., Nitidulidae). Radish (Raphanus sativus L.) was used as an example of a common garden crop and it was sown as four replications at three different distances and on four occasions during the season. Care was taken to protect the radish plots from pesticide due to wind drift during applications. Damage to the radish by flea beetle and cabbage root fly (Delia radicum L., Anthomyiidae) along with unspecified leaf damage was recorded. Significantly lower damage by flea beetles was found on cotyledons of radish adjacent to the insecticide treated side of the OSR field compared to the untreated side in radishes from the first sowing. Unspecified damage to true leaves was also less abundant on radishes at the treated side of the OSR field as compared to the untreated side, in all three of the radish harvests analysed. However, radish plot distance from the OSR field did not influence leaf damage. Root fly damage rates in radish did not differ significantly between those adjacent to the treated and untreated sides of the OSR. Damage rates were, however, higher in the radish plots closest to the OSR field in the first sowing, which coincides with the appearance of the first ovipositing females after overwintering as pupae elsewhere. Generally, insecticide treatment in the agricultural field appeared to influence overall damage in the neighbouring garden crop, despite the fact that the garden crop was protected against wind drift of the insecticides during applications. 相似文献

Kinetic models are among the tools that can be used for optimization of biocatalytic reactions as well as for facilitating process design and upscaling in order to improve productivity and economy of these processes. Mechanism pathways for multi‐substrate multi‐product enzyme‐catalyzed reactions can become very complex and lead to kinetic models comprising several tens of terms. Hence the models comprise too many parameters, which are in general highly correlated and their estimations are often prone to huge errors. In this study, Novozym®435 catalyzed esterification reaction between oleic acid (OA) and trimethylolpropane (TMP) with continuous removal of side‐product (water) was carried out as an example for reactions that follow multi‐substrate multi‐product ping‐pong mechanisms. A kinetic model was developed based on a simplified ping‐pong mechanism proposed for the reaction. The model considered both enzymatic and spontaneous reactions involved and also the effect of product removal during the reaction. The kinetic model parameters were estimated using nonlinear curve fitting through unconstrained optimization methodology and the model was verified by using empirical data from different experiments and showed good predictability of the reaction under different conditions. 1. Some lepidopteran species have larvae that live gregariously, especially in early instars. Colony‐living species may benefit from improved protection from predators, thermoregulation, and feeding facilitation, for example. 2. While many studies have compared solitary and gregarious life styles, few data exist as to the relationship between size of the larval colony and larval performance in gregarious species. The present study was aimed at understanding the importance of colony size for growth and survival of the northern pine processionary moth (Thaumetopoea pinivora) larvae. 3. Field studies, comparing three different sizes of colonies of T. pinivora larvae, showed that individuals in larger colonies had a higher survival rate compared with those living in smaller colonies and also a faster growth rate. 4. The higher survival rate of large colonies was attributed to improved protection from predacious arthropods. 5. In early spring, the young larvae bask in the sun to increase their body temperature. In field experiments the thermal gain was higher in large colonies, and individuals in such colonies also grew faster. As growth rate was not affected by colony size when the ability to bask was experimentally removed in a laboratory experiment, the higher growth rate of the larger colonies was probably due to improved thermoregulation rather than feeding facilitation. 6. The size of larval colonies of gregarious insects depends on natural mortality events as well as on female oviposition strategy. Our results show that decreasing colony size can lead to a reduction in growth rate and survival. It is therefore important to understand whether or not small colonies will benefit equally from the gregarious behaviour. 相似文献

Objectives
ACAT2 is the exclusive cholesterol-esterifying enzyme in hepatocytes and enterocytes. Hepatic ABCA1 transfers unesterified cholesterol (UC) to apoAI, thus generating HDL. By changing the hepatic UC pool available for ABCA1, ACAT2 may affect HDL metabolism. The aim of this study was to reveal whether hepatic ACAT2 influences HDL metabolism.Design
WT and LXRα/β double knockout (DOKO) mice were fed a western-type diet for 8 weeks. Animals were i.p. injected with an antisense oligonucleotide targeted to hepatic ACAT2 (ASO6), or with an ASO control. Injections started 4 weeks after, or concomitantly with, the beginning of the diet.Results
ASO6 reduced liver cholesteryl esters, while not inducing UC accumulation. ASO6 increased hepatic ABCA1 protein independently of the diet conditions. ASO6 affected HDL lipids (increased UC) only in DOKO, while it increased apoE-containing HDL in both genotypes. In WT mice ASO6 led to the appearance of large HDL enriched in apoAI and apoE.Conclusions
The use of ASO6 revealed a new pathway by which the liver may contribute to HDL metabolism in mice. ACAT2 seems to be a hepatic player affecting the cholesterol fluxes fated to VLDL or to HDL, the latter via up-regulation of ABCA1. 相似文献999.
Tetracycline analogues (TCNAs) have been shown to inhibit matrix metalloproteinases and to induce apoptosis in several cancer cell types. In the present study, the cytotoxic effects of TCNAs doxycycline (DOXY), minocycline (MINO) and chemically modified tetracycline-3 (COL-3) were investigated in the human acute myeloid leukemia HL-60 cell line. Cells were incubated with TCNAs in final concentrations of 0.5–100 µg/ml for 24 h. Viability of the leukemic cells was inhibited in a concentration-dependent manner using resazurin assay. The estimated IC50s were 9.2 µg/ml for DOXY, 9.9 µg/ml for MINO and 1.3 µg/ml for COL-3. All three TCNAs induced potent cytotoxic effects and cell death. Apoptosis, which was assessed by morphological changes and annexin V positivity, was concentration- and time-dependent following incubation with any one of the drugs. TCNAs induced DNA double strand breaks soon after treatment commenced as detected by γH2AX and western blot. The loss of mitochondrial membrane potential (Δψm), caspase activation and cleavage of PARP and Bcl-2 were observed; however, the sequence of events differed among the drugs. Pancaspase inhibitor Z-VAD-FMK improved survival of TCNAs-treated cells and decreased TCNAs-induced apoptosis. In summary, we demonstrated that TCNAs had a cytotoxic effect on the HL-60 leukemic cell line. Apoptosis was induced via mitochondria-mediated and caspase-dependent pathways in HL-60 cells by all three TCNAs. COL-3 exerted the strongest anti-proliferative and pro-apoptotic effects in concentrations that have been achieved in human plasma in reported clinical trials.
