We have investigated methionine adenosyltransferase activity (MAT) in extracts of a variety of normal and malignant human tissues and cultured cell lines. MAT activity assayed from 17 different cultured cell lines varied to a great extent. Ramos (human, Burkitt's lymphoma) and EL4 (mouse, T cell lymphoma) cell showed MAT activity near 300 pmol/mg per min. Daudi (human, Burkitt's lymphoma) and almost all monolayer cells had MAT activity below 100 pmol/mg per min. Human peripheral blood lymphocytes had MAT activity of 36 pmol/mg permin. The MAT activity of the cell lines can be related to doubling time: cell lines with short doubling times have much higher MAT activity than other cell lines. A large variation in MAT activity in different human tissues was observed. In autopsy samples MAT activity was highest in the brain and in the colon. Malignant tissue samples gave much higher MAT activity than normal tissues. Lung cancer (carcinoma squamocellulare pulmonis) had MAT activity of 30.7 pmol/mg per min, while in normal lung it was 2.4 pmol/mg per min. 相似文献

Exposure to vibration is suggested as a risk factor for developing neck and shoulder disorders in working life. Mechanical vibration applied to a muscle belly or a tendon can elicit a reflex muscle contraction, also called tonic vibration reflex, but the mechanisms behind how vibration could cause musculoskeletal disorders has not yet been described. One suggestion has been that the vibration causes muscular fatigue. This study investigates whether vibration exposure changes the development of muscular fatigue in the trapezius muscle. Thirty-seven volunteers (men and women) performed a sub-maximal isometric shoulder elevation for 3 min. This was repeated four times, two times with induced vibration and two times without. Muscle activity was measured before and after each 3-min period to look at changes in the electromyography parameters. The result showed a significantly smaller mean frequency decrease when performing the shoulder elevation with vibration (?2.51 Hz) compared to without vibration (?4.04 Hz). There was also a slightly higher increase in the root mean square when exposed to vibration (5.7% of maximal voluntary contraction) compared to without (3.8% of maximal voluntary contraction); however, this was not statistically significant. The results of the present study indicate that short-time exposure to vibration has no negative acute effects on the fatiguing of upper trapezius muscle. 相似文献

Flexible goniometers are useful for direct movement measurements. Crosstalk due to rotation between the endblocks is well known. However, even without any rotation, some crosstalk can occur. The objective of this study was to elucidate the effect of, and compensate for, the inherent crosstalk in biaxial goniometers, with specific relevance for applications with one dominating movement direction. Six biaxial goniometers (M110, Biometrics Ltd., Gwent, UK) were evaluated. A precision jig, for simulating pure flexion/extension angles, was constructed. Each sensor produced a consistent and specific crosstalk pattern, when tested over a ±100° range of motion. A procedure for correction for the inherent crosstalk of individual goniometer, based on polynomial adjust, is presented. The method for compensation, which reduced the root mean square error from, on average for the six goniometers, 3.7° (range 1.8–10.1°) to 0.35° (0.12–0.55°), might be required for obtaining valid goniometer measurements, e.g. of valgus/varus of the knee during gait flexion/extension movements. 相似文献

Chloroplast membranes contain a substantial excess of the nonbilayer-prone monogalactosyldiacylglycerol (GalDAG) over the biosynthetically consecutive, bilayer-forming digalactosyldiacylglycerol (GalGalDAG), yielding a high membrane curvature stress. During phosphate shortage, plants replace phospholipids with GalGalDAG to rescue phosphate while maintaining membrane homeostasis. Here we investigate how the activity of the corresponding glycosyltransferase (GT) in Arabidopsis thaliana (atDGD2) depends on local bilayer properties by analyzing structural and activity features of recombinant protein. Fold recognition and sequence analyses revealed a two-domain GT-B monotopic structure, present in other plant and bacterial glycolipid GTs, such as the major chloroplast GalGalDAG GT atDGD1. Modeling led to the identification of catalytically important residues in the active site of atDGD2 by site-directed mutagenesis. The DGD synthases share unique bilayer interface segments containing conserved tryptophan residues that are crucial for activity and for membrane association. More detailed localization studies and liposome binding analyses indicate differentiated anchor and substrate-binding functions for these separated enzyme interface regions. Anionic phospholipids, but not curvature-increasing nonbilayer lipids, strongly stimulate enzyme activity. From our studies, we propose a model for bilayer "control" of enzyme activity, where two tryptophan segments act as interface anchor points to keep the substrate region close to the membrane surface. Binding of the acceptor substrate is achieved by interaction of positive charges in a surface cluster of lysines, arginines, and histidines with the surrounding anionic phospholipids. The diminishing phospholipid fraction during phosphate shortage stress will then set the new GalGalDAG/phospholipid balance by decreasing stimulation of atDGD2. 相似文献

We have earlier shown that d-lysergic acid diethylamide, LSD and its 2-bromo derivative, BOL like the dopamine (DA) antagonists haloperidol increased the rate of the tyrosine hydroxylation in the striatum measured as the accumulation of DOPA after decarboxylase inhibition.Now we have found that several agents structurally similar to LSD increase the tyrosine hydroxylation in the striatum. Psilocybin (50 mg/kg i.p.) and N,N-dimethyltryptamine (50 mg/kg i.p.) caused a short-lasting increase of DOPA accumulation, while mescaline (10 – 100 mg/kg i.p.) did not increase the DOPA accumulation. A marked increase of DOPA accumulation was observed after the 5-hydroxytryptamine (5-HT) antagonist cyproheptadine. The effects of LSD and structurally related drugs on the DOPA accumulation in the striatum appear to be mediated via DA antagonism at receptor level. However, these agents may control the DOPA accumulation via other receptors than DA receptors e.g. 5-HT receptors. A control of DOPA accumulation via receptors other than DA receptors appears to be predominant after treatment with N,N-dimethyltryptamine or psilocybin. 相似文献

Sexual behaviour was studied in castrated rabbits injected every third day with 1, 7·5, 15 or 30 mg TP, during a 120-day period. Castration led to a very pronounced reduction of sexual activity. This was restored to precastration levels by the three larger doses used about 60 days after the beginning of hormone treatment. The lowest dose did not cause any significant increase in sexual activity. About 70 days after the hormone treatment ceased, almost all sexual activity had disappeared. The correlation coefficients obtained between the various parameters of sexual activity ranged between 0·21 and 0·54. Ragweed pollen, which is a major cause of allergic rhinitis in North America and during the last decades, also in parts of the European continent, has not been regarded as a risk in Sweden, since occurrences of Ambrosia have been rare and with two exceptions, ephemeral. During the last four years, however, long-distance transported pollen has been registered in South Sweden. Moreover, seeds are dispersed by man with birdseed and reports on ragweed plants growing at birdtables are becoming more and more common. In warm years, the fruits of these plants mature and are germinative. We argue that there is a risk that ragweed pollen may be a problem in the future in Sweden. Long-distance transport from the continent is likely to be increasingly more common, and in larger amounts, as ragweed is rapidly spreading in Europe. Among the plants now appearing within Sweden, selection is likely to favour phenotypes which are able to fulfil their life-cycle during the indigenous vegetation period, especially in a scenario of global warming. 相似文献

Today's intensive farming practices are known to have affected farmland biodiversity negatively in many different ways. As far as birds are concerned, they are known to have suffered during both summer and winter. Relatively little is known about the effects on birds during migration. We studied the stopover ecology of the Eurasian golden plover Pluvialis apricaria, a species listed in EU Birds Directive, in intensively farmed arable land in southernmost Sweden in the autumns of 2003–2007. We used key ecological variables (length of stay, fat deposition and moult) as fitness proxies to evaluate how the birds manage in this habitat. Eurasian golden plovers were present in large numbers mainly on arable fields from early August to November and radio‐tagged birds were found to stay in the area for up to three months. Adult birds carried out a substantial part of their flight feather moult during their stay. Body mass increased only somewhat during moult, but from the last stages of moult and onwards fuel loads corresponding to 24% above lean body mass (LBM) were accumulated at a rate of 0.5% of LBM per day, before the birds departed. Juveniles arrived later, from mid Sep., and had a similar pattern of fuel deposition. The fact that the birds choose to stay for long periods, moult in the area, and manage to store substantial fuel loads strongly suggests that Eurasian golden plovers do well in this intensively farmed arable land. 相似文献

The european lynx (Lynx lynx) might be expected to have a high intake of radiocaesium in the parts of Sweden where the main prey of the lynx, namely reindeer and roe deer have high activity concentrations of radiocaesium because of high ground deposition. We have measured 137Cs in muscle samples from 733 lynx during 1996–2003. The aim was to quantify the extent to which radiocaesium is transferred from fallout deposition to lynx, to test whether the transfer was higher in areas where there are reindeer present, to see if there was any decline in radiocaesium over time, and to calculate the radiation dose to lynx. Most samples were collected in central and northern Sweden during January–April. Activity concentrations in lynx varied from 13 Bq kg–1 to about 15 kBq kg–1 fresh weight, with the highest value corresponding to a radiation dose at 18 mGy/year. Aggregated transfer coefficients (Tag), calculated by dividing the 137Cs activity concentration in lynx muscle by the average ground deposition (total from Chernobyl and nuclear weapon tests) within a 50 km radius around the location of the lynx, varied from 0.004 to 1.3 m2 kg–1 and were significantly higher within the reindeer herding area than outside. The concentration ratio (CR) for lynx/reindeer was 2.6 on average, whilst the average for lynx/roe deer outside the reindeer herding area was lower at 1.3. Based on these results, a CR of around 2 could be considered representative for the general ratio between predator and prey. A long-term decline of radiocaesium in prey species was reflected in lynx, with an effective half-life of 7 years from 1996 to 2003. The study shows that the accumulation of radiocaesium in predators, especially predators of reindeer, makes them more vulnerable to high radiocaesium deposition than most other wild species. 相似文献
